The past two decades have been monumental in the advancement of microchips designed for a diverse range of medical applications and bio-analysis. Owing to the remarkable progress in micro-fabrication technology, complex chemical and electro-mechanical features can now be integrated into chip-scale devices for use in biosensing and physiological measurements. Some of these devices have made enormous contributions in the study of complex biochemical processes occurring at the molecular and cellular levels while others overcame the challenges of replicating various functions of human organs as implant systems. This thesis presents test data and analysis of two such systems. First, an ISFET based pH sensor is characterized for its performance in a continuous pH monitoring application. Many of the basic properties of ISFETs including I-V characteristics, pH sensitivity and more importantly, its long term drift behavior have been investigated. A new theory based on frequent switching of electric field across the gate oxide to decrease the rate of current drift has been successfully implemented with the help of an automated data acquisition and switching system. The system was further tested for a range of duty cycles in order to accurately determine the minimum length of time required to fully reset the drift. Second, a microfluidic based vestibular implant system was tested for its underlying characteristics as a light sensor. A computer controlled tilt platform was then implemented to further test its sensitivity to inclinations and thus it‟s more important role as a tilt sensor. The sensor operates through means of optoelectronics and relies on the signals generated from photodiode arrays as a result of light being incident on them. ISFET results show a significant drop in the overall drift and good linear characteristics. The drift was seen to reset at less than an hour. The photodiodes show ideal I-V comparison between photoconductive and photovoltaic modes of operation with maximum responsivity at 400nm and a shunt resistance of 394 MΩ. Additionally, post-processing of the tilt sensor to incorporate the sensing fluids is outlined. Based on several test and fabrication results, a possible method of sealing the open cavity of the chip using a UV curable epoxy has been discussed.

Over the past fifty years, the development of sensors for biological applications has increased dramatically. This rapid growth can be attributed in part to the reduction in feature size, which the electronics industry has pioneered over the same period. The decrease in feature size has led to the production of microscale sensors that are used for sensing applications, ranging from whole-body monitoring down to molecular sensing. Unfortunately, sensors are often developed without regard to how they will be integrated into biological systems. The complexities of integration are underappreciated. Integration involves more than simply making electrical connections. Interfacing microscale sensors with biological environments requires numerous considerations with respect to the creation of compatible packaging, the management of biological reagents, and the act of combining technologies with different dimensions and material properties. Recent advances in microfluidics, especially the proliferation of soft lithography manufacturing methods, have established the groundwork for creating systems that may solve many of the problems inherent to sensor-fluidic interaction. The adaptation of microelectronics manufacturing methods, such as Complementary Metal-Oxide-Semiconductor (CMOS) and Microelectromechanical Systems (MEMS) processes, allows the creation of a complete biological sensing system with integrated sensors and readout circuits. Combining these technologies is an obstacle to forming complete sensor systems. This dissertation presents new approaches for the design, fabrication, and integration of microscale sensors and microelectronics with microfluidics. The work addresses specific challenges, such as combining commercial manufacturing processes into biological systems and developing microscale sensors in these processes. This work is exemplified through a feedback-controlled microfluidic pH system to demonstrate the integration capabilities of microscale sensors for autonomous microenvironment control.

Analysing and measuring of biological or biochemical processes are of utmost importance for medical, biological and biotechnological applications. Point of care diagnostic system, composing of biosensors, have promising applications for providing cheap, accurate and portable diagnosis. Owing to these expanding medical applications and advances made by semiconductor industry biosensors have seen a tremendous growth in the past few decades. Also emergence of microfluidics and non-invasive biosensing applications are other marker propellers. Analyzing biological signals using transducers is difficult due to the challenges in interfacing an electronic system to the biological environment. Detection limit, detection time, dynamic range, specificity to the analyte, sensitivity and reliability of these devices are some of the challenges in developing and integrating these devices. Significant amount of research in the field of biosensors has been focused on improving the design, fabrication process and their integration with microfluidics to address these challenges. This work presents new techniques, design and systems to improve the interface between the electronic system and the biological environment. This dissertation uses CMOS circuit design to improve the reliability of these devices. Also this work addresses the challenges in designing the electronic system used for processing the output of the transducer, which converts biological signal into electronic signal.

Three-dimensional (3D) inductors with square, hexagonal and octagonal geometries have been designed and simulated in ANSYS HFSS. The inductors have been designed on Silicon substrate with through-hole via with different width, spacing and thickness. Spice modeling has been done in Agilent ADS and comparison has been made with results of custom excel based calculator and HFSS simulation results. Single ended quality factor was measured as 12.97 and differential ended quality factor was measured as 15.96 at a maximum operational frequency of 3.65GHz. The single ended and differential inductance was measured as 2.98nH and 2.88nH respectively at this frequency. Based on results a symmetric octagonal inductor design has been recommended to be used for application in RF biosensing. A system design has been proposed based on use of this inductor and principle of inductive sensing using magnetic labeling.

Despite the various driver assistance systems and electronics, the threat to life of driver, passengers and other people on the road still persists. With the growth in technology, the use of in-vehicle devices with a plethora of buttons and features is increasing resulting in increased distraction. Recently, speech recognition has emerged as an alternative to distraction and has the potential to be beneficial. However, considering the fact that automotive environment is dynamic and noisy in nature, distraction may not arise from the manual interaction, but due to the cognitive load. Hence, speech recognition certainly cannot be a reliable mode of communication.

The thesis is focused on proposing a simultaneous multimodal approach for designing interface between driver and vehicle with a goal to enable the driver to be more attentive to the driving tasks and spend less time fiddling with distractive tasks. By analyzing the human-human multimodal interaction techniques, new modes have been identified and experimented, especially suitable for the automotive context. The identified modes are touch, speech, graphics, voice-tip and text-tip. The multiple modes are intended to work collectively to make the interaction more intuitive and natural. In order to obtain a minimalist user-centered design for the center stack, various design principles such as 80/20 rule, contour bias, affordance, flexibility-usability trade-off etc. have been implemented on the prototypes. The prototype was developed using the Dragon software development kit on android platform for speech recognition.

In the present study, the driver behavior was investigated in an experiment conducted on the DriveSafety driving simulator DS-600s. Twelve volunteers drove the simulator under two conditions: (1) accessing the center stack applications using touch only and (2) accessing the applications using speech with offered text-tip. The duration for which user looked away from the road (eyes-off-road) was measured manually for each scenario. Comparison of results proved that eyes-off-road time is less for the second scenario. The minimalist design with 8-10 icons per screen proved to be effective as all the readings were within the driver distraction recommendations (eyes-off-road time < 2sec per screen) defined by NHTSA.

Surface Plasmon Resonance (SPR), as a surface-sensitive analytical tool, can very sensitively respond to minute

Biosensors aiming at detection of target analytes, such as proteins, microbes, virus, and toxins, are widely needed for various applications including detection of chemical and biological warfare (CBW) agents, biomedicine, environmental monitoring, and drug screening. Surface Plasmon Resonance (SPR), as a surface-sensitive analytical tool, can very sensitively respond to minute changes of refractive index occurring adjacent to a metal film, offering detection limits up to a few ppt (pg/mL). Through SPR, the process of protein adsorption may be monitored in real-time, and transduced into an SPR angle shift. This unique technique bypasses the time-consuming, labor-intensive labeling processes, such as radioisotope and fluorescence labeling. More importantly, the method avoids the modification of the biomarker’s characteristics and behaviors by labeling that often occurs in traditional biosensors. While many transducers, including SPR, offer high sensitivity, selectivity is determined by the bio-receptors. In traditional biosensors, the selectivity is provided by bio-receptors possessing highly specific binding affinity to capture target analytes, yet their use in biosensors are often limited by their relatively-weak binding affinity with analyte, non-specific adsorption, need for optimization conditions, low reproducibility, and difficulties integrating onto the surface of transducers. In order to circumvent the use of bio-receptors, the competitive adsorption of proteins, termed the Vroman effect, is utilized in this work. The Vroman effect was first reported by Vroman and Adams in 1969. The competitive adsorption targeted here occurs among different proteins competing to adsorb to a surface, when more than one type of protein is present. When lower-affinity proteins are adsorbed on the surface first, they can be displaced by higher-affinity proteins arriving at the surface at a later point in time. Moreover, only low-affinity proteins can be displaced by high-affinity proteins, typically possessing higher molecular weight, yet the reverse sequence does not occur. The SPR biosensor based on competitive adsorption is successfully demonstrated to detect fibrinogen and thyroglobulin (Tg) in undiluted human serum and copper ions in drinking water through the denatured albumin.

This work explores how flexible electronics and display technology can be applied to develop new biomedical devices for medical, biological, and life science applications. It demonstrates how new biomedical devices can be manufactured by only modifying or personalizing the upper layers of a conventional thin film transistor (TFT) display process.

This work explores how flexible electronics and display technology can be applied to develop new biomedical devices for medical, biological, and life science applications. It demonstrates how new biomedical devices can be manufactured by only modifying or personalizing the upper layers of a conventional thin film transistor (TFT) display process. This personalization was applied first to develop and demonstrate the world's largest flexible digital x-ray detector for medical and industrial imaging, and the world's first flexible ISFET pH biosensor using TFT technology. These new, flexible, digital x-ray detectors are more durable than conventional glass substrate x-ray detectors, and also can conform to the surface of the object being imaged. The new flexible ISFET pH biosensors are >10X less expensive to manufacture than comparable CMOS-based ISFETs and provide a sensing area that is orders of magnitude larger than CMOS-based ISFETs. This allows for easier integration with area intensive chemical and biological recognition material as well as allow for a larger number of unique recognition sites for low cost multiple disease and pathogen detection.

The flexible x-ray detector technology was then extended to demonstrate the viability of a new technique to seamlessly combine multiple smaller flexible x-ray detectors into a single very large, ultimately human sized, composite x-ray detector for new medical imaging applications such as single-exposure, low-dose, full-body digital radiography. Also explored, is a new approach to increase the sensitivity of digital x-ray detectors by selectively disabling rows in the active matrix array that are not part of the imaged region. It was then shown how high-resolution, flexible, organic light-emitting diode display (OLED) technology can be used to selectively stimulate and/or silence small groups of neurons on the cortical surface or within the deep brain as a potential new tool to diagnose and treat, as well as understand, neurological diseases and conditions. This work also explored the viability of a new miniaturized high sensitivity fluorescence measurement-based lab-on-a-chip optical biosensor using OLED display and a-Si:H PiN photodiode active matrix array technology for point-of-care diagnosis of multiple disease or pathogen biomarkers in a low cost disposable configuration.

Driver distraction research has a long history spanning nearly 50 years, intensifying in the last decade. The focus has always been on identifying the distractive tasks and measuring the respective harm level. As in-vehicle technology advances, the list of distractive activities grows along with crash risk. Additionally, the distractive activities become more common and complicated, especially with regard to In-Car Interactive System. This work's main focus is on driver distraction caused by the in-car interactive System. There have been many User Interaction Designs (Buttons, Speech, Visual) for Human-Car communication, in the past and currently present. And, all related studies suggest that driver distraction level is still high and there is a need for a better design. Multimodal Interaction is a design approach, which relies on using multiple modes for humans to interact with the car & hence reducing driver distraction by allowing the driver to choose the most suitable mode with minimum distraction. Additionally, combining multiple modes simultaneously provides more natural interaction, which could lead to less distraction. The main goal of MMI is to enable the driver to be more attentive to driving tasks and spend less time fiddling with distractive tasks. Engineering based method is used to measure driver distraction. This method uses metrics like Reaction time, Acceleration, Lane Departure obtained from test cases.

Driving is already a complex task that demands a varying level of cognitive and physical load. With the advancement in technology, the car has become a place for media consumption, a communications center and an interconnected workplace. The number of features in a car has also increased. As a result, the user interaction inside the car has become overcrowded and more complex. This has increased the amount of distraction while driving and has also increased the number of accidents due to distracted driving. This thesis focuses on the critical analysis of today’s in-car environment covering two main aspects, Multi Modal Interaction (MMI), and Advanced Driver Assistance Systems (ADAS), to minimize the distraction. It also provides deep market research on future trends in the smart car technology. After careful analysis, it was observed that an infotainment screen cluttered with lots of small icons, a center stack with a plethora of small buttons and a poor Voice Recognition (VR) results in high cognitive load, and these are the reasons for the increased driver distraction. Though the VR has become a standard technology, the current state of technology is focused on features oriented design and a sales driven approach. Most of the automotive manufacturers are focusing on making the VR better but attaining perfection in VR is not the answer as there are inherent challenges and limitations in respect to the in-car environment and cognitive load. Accordingly, the research proposed a novel in-car interaction design solution: Multi-Modal Interaction (MMI). The MMI is a new term when used in the context of vehicles, but it is widely used in human-human interaction. The approach offers a non-intrusive alternative to the driver to interact with the features in the car. With the focus on user-centered design, the MMI and ADAS can potentially help to reduce the distraction. To support the discussion, an experiment was conducted to benchmark a minimalist UI design. An engineering based method was used to test and measure distraction of four different UIs with varying numbers of icons and screen sizes. Lastly, in order to compete with the market, the basic features that are provided by all the other competitors cannot be eliminated, but the hard work can be done to improve the HCaI and to make driving safer.

Driving is the coordinated operation of mind and body for movement of a vehicle, such as a car, or a bus. Driving, being considered an everyday activity for many people, still has an issue of safety. Driver distraction is becoming a critical safety problem. Speed, drunk driving as well as distracted driving are the three leading factors in the fatal car crashes. Distraction, which is defined as an excessive workload and limited attention, is the main paradigm that guides this research area. Driver behavior analysis can be used to address the distraction problem and provide an intelligent adaptive agent to work closely with the driver, fay beyond traditional algorithmic computational models. A variety of machine learning approaches has been proposed to estimate or predict drivers’ fatigue level using car data, driver status or a combination of them.

Three important features of intelligence and cognition are perception, attention and sensory memory. In this thesis, I focused on memory and attention as essential parts of highly intelligent systems. Without memory, systems will only show limited intelligence since their response would be exclusively based on spontaneous decision without considering the effect of previous events. I proposed a memory-based sequence to predict the driver behavior and distraction level using neural network. The work started with a large-scale experiment to collect data and make an artificial intelligence-friendly dataset. After that, the data was used to train a deep neural network to estimate the driver behavior. With a focus on memory by using Long Short Term Memory (LSTM) network to increase the level of intelligence in two dimensions: Forgiveness of minor glitches, and accumulation of anomalous behavior., I reduced the model error and computational expense by adding attention mechanism on the top of LSTM models. This system can be generalized to build and train highly intelligent agents in other domains.
