Autonomous Vehicles (AV) are inevitable entities in future mobility systems thatdemand safety and adaptability as two critical factors in replacing/assisting human drivers. Safety arises in defining, standardizing, quantifying, and monitoring requirements for all autonomous components. Adaptability, on the other hand, involves efficient handling of uncertainty and inconsistencies in models and data. First, I address safety by presenting a search-based test-case generation framework that can be used in training and testing deep-learning components of AV. Next, to address adaptability, I propose a framework based on multi-valued linear temporal logic syntax and semantics that allows autonomous agents to perform model-checking on systems with uncertainties. The search-based test-case generation framework provides safety assurance guarantees through formalizing and monitoring Responsibility Sensitive Safety (RSS) rules. I use the RSS rules in signal temporal logic as qualification specifications for monitoring and screening the quality of generated test-drive scenarios. Furthermore, to extend the existing temporal-based formal languages’ expressivity, I propose a new spatio-temporal perception logic that enables formalizing qualification specifications for perception systems. All-in-one, my test-generation framework can be used for reasoning about the quality of perception, prediction, and decision-making components in AV. Finally, my efforts resulted in publicly available software. One is an offline monitoring algorithm based on the proposed logic to reason about the quality of perception systems. The other is an optimal planner (model checker) that accepts mission specifications and model descriptions in the form of multi-valued logic and multi-valued sets, respectively. My monitoring framework is distributed with the publicly available S-TaLiRo and Sim-ATAV tools.

Many real-world engineering problems require simulations to evaluate the design objectives and constraints. Often, due to the complexity of the system model, simulations can be prohibitive in terms of computation time. One approach to overcome this issue is to construct a surrogate model, which approximates the original model. The focus of this work is on the data-driven surrogate models, in which empirical approximations of the output are performed given the input parameters. Recently neural networks (NN) have re-emerged as a popular method for constructing data-driven surrogate models. Although, NNs have achieved excellent accuracy and are widely used, they pose their own challenges. This work addresses two common challenges, the need for: (1) hardware acceleration and (2) uncertainty quantification (UQ) in the presence of input variability. The high demand in the inference phase of deep NNs in cloud servers/edge devices calls for the design of low power custom hardware accelerators. The first part of this work describes the design of an energy-efficient long short-term memory (LSTM) accelerator. The overarching goal is to aggressively reduce the power consumption and area of the LSTM components using approximate computing, and then use architectural level techniques to boost the performance. The proposed design is synthesized and placed and routed as an application-specific integrated circuit (ASIC). The results demonstrate that this accelerator is 1.2X and 3.6X more energy-efficient and area-efficient than the baseline LSTM. In the second part of this work, a robust framework is developed based on an alternate data-driven surrogate model referred to as polynomial chaos expansion (PCE) for addressing UQ. In contrast to many existing approaches, no assumptions are made on the elements of the function space and UQ is a function of the expansion coefficients. Moreover, the sensitivity of the output with respect to any subset of the input variables can be computed analytically by post-processing the PCE coefficients. This provides a systematic and incremental method to pruning or changing the order of the model. This framework is evaluated on several real-world applications from different domains and is extended for classification tasks as well.

Floating trash objects are very commonly seen on water bodies such as lakes, canals and rivers. With the increase of plastic goods and human activities near the water bodies, these trash objects can pile up and cause great harm to the surrounding environment. Using human workers to clear out these trash is a hazardous and time-consuming task. Employing autonomous robots for these tasks is a better approach since it is more efficient and faster than humans. However, for a robot to clean the trash objects, a good detection algorithm is required. Real-time object detection on water surfaces is a challenging issue due to nature of the environment and the volatility of the water surface. In addition to this, running an object detection algorithm on an on-board processor of a robot limits the amount of CPU consumption that the algorithm can utilize. In this thesis, a computationally low cost object detection approach for robust detection of trash objects that was run on an on-board processor of a multirotor is presented. To account for specular reflections on the water surface, we use a polarization filter and integrate a specularity removal algorithm on our approach as well. The challenges faced during testing and the means taken to eliminate those challenges are also discussed. The algorithm was compared with two other object detectors using 4 different metrics. The testing was carried out using videos of 5 different objects collected at different illumination conditions over a lake using a multirotor. The results indicate that our algorithm is much suitable to be employed in real-time since it had the highest processing speed of 21 FPS, the lowest CPU consumption of 37.5\% and considerably high precision and recall values in detecting the object.

How to teach a machine to understand natural language? This question is a long-standing challenge in Artificial Intelligence. Several tasks are designed to measure the progress of this challenge. Question Answering is one such task that evaluates a machine's ability to understand natural language, where it reads a passage of text or an image and answers comprehension questions. In recent years, the development of transformer-based language models and large-scale human-annotated datasets has led to remarkable progress in the field of question answering. However, several disadvantages of fully supervised question answering systems have been observed. Such as generalizing to unseen out-of-distribution domains, linguistic style differences in questions, and adversarial samples. This thesis proposes implicitly supervised question answering systems trained using knowledge acquisition from external knowledge sources and new learning methods that provide inductive biases to learn question answering. In particular, the following research projects are discussed: (1) Knowledge Acquisition methods: these include semantic and abductive information retrieval for seeking missing knowledge, a method to represent unstructured text corpora as a knowledge graph, and constructing a knowledge base for implicit commonsense reasoning. (2) Learning methods: these include Knowledge Triplet Learning, a method over knowledge graphs; Test-Time Learning, a method to generalize to an unseen out-of-distribution context; WeaQA, a method to learn visual question answering using image captions without strong supervision; WeaSel, weakly supervised method for relative spatial reasoning; and a new paradigm for unsupervised natural language inference. These methods potentially provide a new research direction to overcome the pitfalls of direct supervision.

It is not merely an aggregation of static entities that a video clip carries, but alsoa variety of interactions and relations among these entities. Challenges still remain for a video captioning system to generate natural language descriptions focusing on the prominent interest and aligning with the latent aspects beyond observations. This work presents a Commonsense knowledge Anchored Video cAptioNing (dubbed as CAVAN) approach. CAVAN exploits inferential commonsense knowledge to assist the training of video captioning model with a novel paradigm for sentence-level semantic alignment. Specifically, commonsense knowledge is queried to complement per training caption by querying a generic knowledge atlas ATOMIC, and form the commonsense- caption entailment corpus. A BERT based language entailment model trained from this corpus then serves as a commonsense discriminator for the training of video captioning model, and penalizes the model from generating semantically misaligned captions. With extensive empirical evaluations on MSR-VTT, V2C and VATEX datasets, CAVAN consistently improves the quality of generations and shows higher keyword hit rate. Experimental results with ablations validate the effectiveness of CAVAN and reveals that the use of commonsense knowledge contributes to the video caption generation.

There has been an explosion in the amount of data on the internet because of modern technology – especially image data – as a consequence of an exponential growth in the number of cameras existing in the world right now; from more extensive surveillance camera systems to billions of people walking around with smartphones in their pockets that come with built-in cameras. With this sudden increase in the accessibility of cameras, most of the data that is getting captured through these devices is ending up on the internet. Researchers soon took leverage of this data by creating large-scale datasets. However, generating a dataset – let alone a large-scale one – requires a lot of man-hours. This work presents an algorithm that makes use of optical flow and feature matching, along with utilizing localization outputs from a Mask R-CNN, to generate large-scale vehicle datasets without much human supervision. Additionally, this work proposes a novel multi-view vehicle dataset (MVVdb) of 500 vehicles which is also generated using the aforementioned algorithm.There are various research problems in computer vision that can leverage a multi-view dataset, e.g., 3D pose estimation, and 3D object detection. On the other hand, a multi-view vehicle dataset can be used for a 2D image to 3D shape prediction, generation of 3D vehicle models, and even a more robust vehicle make and model recognition. In this work, a ResNet is trained on the multi-view vehicle dataset to perform vehicle re-identification, which is fundamentally similar to a vehicle make and recognition problem – also showcasing the usability of the MVVdb dataset.

Alpha herpesviruses are a family of neuroinvasive viruses that infect multiplevertebrate species. Alpha herpesviruses are responsible for human and livestock infections, most notably Herpes Simplex Virus (HSV), Varicella Zoster virus (VZV), and Pseudorabies Virus (PRV). PRV is a potent swine virus that can infect other mammals, and results in lethal encephalitis that can be devastating to livestock and of great financial expense to farmers. HSV, types 1 and 2, and VZV are widespread throughout the global human population, with estimates of the HSV-1 burden at about 60% of people worldwide. The hallmark of alpha herpesvirus infection is a persistent, lifelong infection that can reactivate throughout the lifespan of the host. Currently, the precise mechanisms of how these viruses undergo intracellular trafficking to emerge from the infected cell in epithelial tissues is not well understood. Many insights have been made with PRV in animal neurons, both in culture systems and animal models, about the viral genes and host factors involved in these processes. However, understanding of these mechanisms, and the interplay between viral and host proteins, in the human pathogen HSV-1 is even more lacking. Using recombinant fluorescent virus strains of HSV-1 and Total Internal Reflection Microscopy to image the transport of mature viral progeny in epithelial cells, it was determined that the egress of HSV-1 uses constitutive cellular secretory pathways. Specifically, the viral progeny traffic from the trans-Golgi network to the site of exocytosis at the plasma membrane via Rab6a secretory vesicles. This work will contribute to the understanding of how alpha herpesviruses complete their lifecycles in host cells, particularly at the sites where infection initially occurs and can spread to a new organism. Knowledge of these processes may lead to the development of therapeutics or prophylactics to reduce the burden of these viruses.
