Background: Continuous monitoring technologies such as accelerometers and pedometers are the gold standard for physical activity (PA) measurement. However, inconsistencies in use, analysis, and reporting limit the understanding of dose–response relationships involving PA and the ability to make comparisons across studies and population subgroups. These issues are particularly detrimental to the study of PA across different ethnicities with different PA habits. This systematic review examined the inclusion of published guidelines involving data collection, processing, and reporting among articles using accelerometers or pedometers in Hispanic or Latino populations.

Methods: English (PubMed; EbscoHost) and Spanish (SCIELO; Biblioteca Virtual en Salud) articles published between 2000 and 2013 using accelerometers or pedometers to measure PA among Hispanics or Latinos were identified through systematic literature searches. Of the 253 abstracts which were initially reviewed, 57 met eligibility criteria (44 accelerometer, 13 pedometer). Articles were coded and reviewed to evaluate compliance with recommended guidelines (N = 20), and the percentage of accelerometer and pedometer articles following each guideline were computed and reported.

Results: On average, 57.1 % of accelerometer and 62.2 % of pedometer articles reported each recommended guideline for data collection. Device manufacturer and model were reported most frequently, and provision of instructions for device wear in Spanish was reported least frequently. On average, 29.6 % of accelerometer articles reported each guideline for data processing. Definitions of an acceptable day for inclusion in analyses were reported most frequently, and definitions of an acceptable hour for inclusion in analyses were reported least frequently. On average, 18.8 % of accelerometer and 85.7 % of pedometer articles included each guideline for data reporting. Accelerometer articles most frequently included average number of valid days and least frequently included percentage of wear time.

Discussion: Inclusion of standard collection and reporting procedures in studies using continuous monitoring devices in Hispanic or Latino population is generally low.

Background: The shift from solitary to social behavior is one of the major evolutionary transitions. Primitively eusocial bumblebees are uniquely placed to illuminate the evolution of highly eusocial insect societies. Bumblebees are also invaluable natural and agricultural pollinators, and there is widespread concern over recent population declines in some species. High-quality genomic data will inform key aspects of bumblebee biology, including susceptibility to implicated population viability threats.

Results: We report the high quality draft genome sequences of Bombus terrestris and Bombus impatiens, two ecologically dominant bumblebees and widely utilized study species. Comparing these new genomes to those of the highly eusocial honeybee Apis mellifera and other Hymenoptera, we identify deeply conserved similarities, as well as novelties key to the biology of these organisms. Some honeybee genome features thought to underpin advanced eusociality are also present in bumblebees, indicating an earlier evolution in the bee lineage. Xenobiotic detoxification and immune genes are similarly depauperate in bumblebees and honeybees, and multiple categories of genes linked to social organization, including development and behavior, show high conservation. Key differences identified include a bias in bumblebee chemoreception towards gustation from olfaction, and striking differences in microRNAs, potentially responsible for gene regulation underlying social and other traits.

Conclusions: These two bumblebee genomes provide a foundation for post-genomic research on these key pollinators and insect societies. Overall, gene repertoires suggest that the route to advanced eusociality in bees was mediated by many small changes in many genes and processes, and not by notable expansion or depauperation.

One of the fundamental questions in molecular biology is how genes and the control of their expression give rise to so many diverse phenotypes in nature. The mRNA molecule plays a key role in this process as it directs the spatial and temporal expression of genetic information contained in the DNA molecule to precisely instruct biological processes in living organisms. The region located between the STOP codon and the poly(A)-tail of the mature mRNA, known as the 3′Untranslated Region (3′UTR), is a key modulator of these activities. It contains numerous sequence elements that are targeted by trans-acting factors that dose gene expression, including the repressive small non-coding RNAs, called microRNAs.

Recent transcriptome data from yeast, worm, plants, and humans has shown that alternative polyadenylation (APA), a mechanism that enables expression of multiple 3′UTR isoforms for the same gene, is widespread in eukaryotic organisms. It is still poorly understood why metazoans require multiple 3′UTRs for the same gene, but accumulating evidence suggests that APA is largely regulated at a tissue-specific level. APA may direct combinatorial variation between cis-elements and microRNAs, perhaps to regulate gene expression in a tissue-specific manner. Apart from a few single gene anecdotes, this idea has not been systematically explored.

This dissertation research employs a systems biology approach to study the somatic tissue dynamics of APA and its impact on microRNA targeting networks in the small nematode C. elegans. In the first aim, tools were developed and applied to isolate and sequence mRNA from worm intestine and muscle tissues, which revealed pervasive tissue-specific APA correlated with microRNA regulation. The second aim provides genetic evidence that two worm genes use APA to escape repression by microRNAs in the body muscle. Finally, in aim three, mRNA from five additional somatic worm tissues was sequenced and their 3′ends mapped, allowing for an integrative study of APA and microRNA targeting dynamics in worms. Together, this work provides evidence that APA is a pervasive mechanism operating in somatic tissues of C. elegans with the potential to significantly rearrange their microRNA regulatory networks and precisely dose their gene expression.

Sustainable purchasing has become and increasingly salient way by which local governments can decrease their resource consumptions, while also addressing broader climate action goals. Successfully implemented sustainable purchasing policies have the potential to reduce consumption and waste, expand green purchasing markets, and catalyze spillover benefits such as financial savings. Furthermore, city-level actions have become increasingly significant as the federal government ceases critical climate research and pulls out of collaborative climate deals (i.e. The Paris Climate Accord). Using data from the Sustainable Purchasing Researching Initiative at Arizona State University’s Center for Organization Research and Design, as well as qualitative policy analyses, the author investigates the elements of a city’s sustainable purchasing policy (SPP) that are related to its implementation success. Furthermore, the author compares these initial findings to the case study of Phoenix, AZ where she explores whether these elements are also present in the City of Phoenix’s sustainable purchasing policy. The author finds that six key policy elements are generally associated with higher SPP implementation success rates — mandatory requirements, accountability, multi-level governance, vendors requirements, advocacy, and continual improvement. While additional policy elements undoubtedly play a role in the successful implementation of a SPP, the author concludes cities that incorporate these specific elements are better positioned for successful and sustainable implementation. Conclusions further show that the City of Phoenix’s 2007 EPP contained only two of these policy elements. As a result of this project and the author’s work with the City, the 2017 revised SPP incorporates all six policy elements.

Adaptive comanagement endeavors to increase knowledge and responsiveness in the face of uncertainty and complexity. However, when collaboration between agency and nonagency stakeholders is mandated, rigid institutions may hinder participation and ecological outcomes. In this case study we analyzed qualitative data to understand how participants perceive strengths and challenges within an emerging adaptive comanagement in the Agua Fria Watershed in Arizona, USA that utilizes insight and personnel from a long-enduring comanagement project, Las Cienegas. Our work demonstrates that general lessons and approaches from one project may be transferable, but particular institutions, management structures, or projects must be place-specific. As public agencies establish and expand governance networks throughout the western United States, our case study has shed light on how to maintain a shared vision and momentum within an inherently murky and shared decision-making environment.

Resource-poor social environments predict poor health, but the mechanisms and processes linking the social environment to psychological health and well-being remain unclear. This study explored psychosocial mediators of the association between the social environment and mental health in African American adults. African American men and women (n = 1467) completed questionnaires on the social environment, psychosocial factors (stress, depressive symptoms, and racial discrimination), and mental health. Multiple-mediator models were used to assess direct and indirect effects of the social environment on mental health. Low social status in the community (p < .001) and U.S. (p < .001) and low social support (p < .001) were associated with poor mental health. Psychosocial factors significantly jointly mediated the relationship between the social environment and mental health in multiple-mediator models. Low social status and social support were associated with greater perceived stress, depressive symptoms, and perceived racial discrimination, which were associated with poor mental health. Results suggest the relationship between the social environment and mental health is mediated by psychosocial factors and revealed potential mechanisms through which social status and social support influence the mental health of African American men and women. Findings from this study provide insight into the differential effects of stress, depression and discrimination on mental health. Ecological approaches that aim to improve the social environment and psychosocial mediators may enhance health-related quality of life and reduce health disparities in African Americans.

Collaborative Research Networks can serve as models of proactive and purposive Collective Action and catalysts of societal change, if they function as more than hubs of research and knowledge. It is the goal of this Honors Thesis to examine the current nature under which collaborative research networks, focused on matters of Global Health or Sustainability, operate., how they are organized, what type of collaboration they engage in, and who collaborates with whom. A better understanding of these types of networks can lead to the formation of more effective networks that can develop innovative solutions to our collective Global Health and Sustainability problems.

As obesity rates continue to rise in adolescents and young children, the concern for poor future health of the younger population grows. Physical activity and improving nutrition are two ways to combat obesity rates, and the Sustainability via Active Gardening Education (SAGE) project addresses this in underserved and low-income communities

As obesity rates continue to rise in adolescents and young children, the concern for poor future health of the younger population grows. Physical activity and improving nutrition are two ways to combat obesity rates, and the Sustainability via Active Gardening Education (SAGE) project addresses this in underserved and low-income communities in Maricopa County. This project employs a curriculum designed to promote physical activity and healthy eating for Early Care and Education (ECE) sites, most of which are daycares. Further, utilizing indicators of future health can also allow for us to understand and lower obesity rates. One indicator of future health is grip strength: greater grip strength is associated with healthier outcomes such as lower triglycerides, blood pressure, and body mass index. Grip strength has been observed in the older population; however, there are few studies looking at grip strength in younger children, namely preschoolers. As grip strength is a predictor of health, it follows that it should be observed in preschoolers, and improved, if possible, by factors such as physical activity, which would ultimately improve obesity rates. This study aimed to see if there was any relationship between physical activity and grip strength in preschoolers aged 3-5 years old. To do so, grip strength, hand length, height, weight, and information regarding physical activity of preschoolers enrolled in the SAGE project were collected. Physical activity and grip strength were not found to be significantly associated in this study; however, hand length and hand strength were associated. Among secondary outcomes, it was observed that males of ages 3 to 5-years-old may have greater hand grip strength than females of the same age group. Although this was not statistically significant, there was a trend toward statistical significance. Small sample size hampered observation of expected relationships between hand grip strength and dominant hand of the participants, and hand grip strength was not significantly related with BMI. Future directions would consist of collecting longitudinal data, as well as calling back previous years’ participants for additional data, so that there is a larger sample size for data analysis.

This project aims to tackle two perspectives: to design and express an enzyme that can perform single-molecule modifications for identification, and to determine the inclusion of the last adenosine in mature mRNAs within the metazoan, Caenorhabditis elegans. Starting with the first perspective, the enzymatic group that was utilized was methyltransferases.

This project aims to tackle two perspectives: to design and express an enzyme that can perform single-molecule modifications for identification, and to determine the inclusion of the last adenosine in mature mRNAs within the metazoan, Caenorhabditis elegans. Starting with the first perspective, the enzymatic group that was utilized was methyltransferases. Methyltransferases have gained great interest in biotechnology and academia due to their ability to make single-molecule modifications to a wide variety of biomolecules, ranging from proteins to RNA. Of these methyltransferases, the subset that has the greatest interest for this study are RNA methyltransferases. Of the known RNA methyltransferases, human METTL16 was chosen for this project, due to its ability to modify adenosines at the N6 position (m6A), specificity for its consensus motif, and its promise in chimeric enzymatic complexes. As a result of these properties, this study looks to design METTl16-based complexes for the purpose of identifying single nucleotides in RNA. The second perspective involves pre-mRNA cleavage and polyadenylation of the 3’ untranslated region (3’UTR). Cleavage of pre-mRNAs within C.elegans appears to prefer an adenosine, leading to the term “terminal adenosine” (terminal-A). Since RNA cleavage and polyadenylation is highly conserved across metazoans, we can utilize the model system, C. elegans, to apply our findings to humans. Utilizing METTL16’s ability to modify adenosines, it is theorized that it may be possible to modify the terminal-A in vivo within C. elegans. To confirm the functionality and utilization of METTL16, a novel methodology is currently being developed called the terminal adenosine methylation (TAM) assay. The TAM assay takes advantage of METTL16’s N-terminal RNA binding domain (RBD) and methyltransferase domain – called the “core” – to methylate the terminal adenosine of probe mRNA transcripts prior to cleavage in vivo. To determine if the adenosine is present within mature mRNAs, sequencing will determine if there is a m6A present, confirming that CPSF-3 cleaves either upstream or downstream of the terminal-A. Ultimately, this project focuses on designing METTL16 complexes for mRNA modification, testing the functionality of these constructs in vitro, and developing transgenic C. elegans strains to express the METTL16 complexes. The bioconjugation capabilities of RNA methyltransferases allow for concepts such as the TAM assay to be viable, as well as make way for future prospects of methyltransferases as a biotechnical tool.
