Cuticular hydrocarbons (CHCs) play a crucial role in social insect recognition systems. In this study we investigated mate choice in the red harvester ant, Pogonomyrmex barbatus. In Phoenix, this species has two lineages, J1 and J2, which look identical, but are genetically isolated. In the genetic caste determination (GCD) system workers and queens are determined by their genotype (i.e., workers develop from interlineage crosses, queens from intralineage crosses). As such, J1 and J2 lineages are dependent on each other in order for colonies to produce both workers and reproductive queens. Given their genetic isolation and interdependence, we hypothesized that the CHCs of alate males and queens are affected by lineage, and that differences in the CHC profile are used for mate recognition. We tested these hypotheses by analyzing the lineage distributions of actively mating pairs (n=65), and compared them with the overall distribution of male and female sexuals (n=180). We additionally analyzed the five most abundant CHC compounds for 20 of the actively mating P. barbatus alate male and queen pairs to determine how variable the two lineages are between each sex. We found that mating pair distributions did not significantly differ from those expected under a random mating system (�2= 1.4349, P= 0.6973), however, CHC profiles did differ between J1 and J2 lineages and sexes for the five most abundant CHC compounds. Our results show that random mating is taking place in this population, however given the differences observed in CHC profiles, mate recognition could be taking place.

I tested the hypothesis that in mature colonies of the seed harvester Pogonomyrmex californicus ant species, paired pleometrotic queens would produce workers more efficiently after a massive removal of their work force than haplometrotic queens, paired pleometrotic with haplometrotic queens, and single pleometrotic queens. I suggested that the paired pleometrotic queens would have an advantage of cooperating together in reproducing more workers quicker than the other conditions to make up for the lost workers. This would demonstrate a benefit that pleometrosis has over haplometrosis for mature colonies, which would explain why pleometrosis continues for P.californicus after colony foundation. After removing all but twenty workers for every colony, I took pictures and counted the emerging brood for 52 days. Analyses showed that the paired pleometrotic queens and the haplometrotic queens both grew at an equally efficient rate and the paired pleometrotic and haplometrotic queens growing the least efficiently. However, the results were not significant and did not support the hypothesis that paired pleometrotic queens recover from worker loss more proficiently than other social systems.

Wolbachia is a genus of obligately intracellular bacterial endosymbionts of arthropods and nematodes, infecting up to 66% of all such species. In order to ensure its transmission, it may modify host reproduction by inducing one of four phenotypes: cytoplasmic incompatibility, feminization of genetic males, killing of male embryos, and induction of thelytokous parthenogenesis. This investigation was a characterization of the so-far unexamined Wolbachia infection of Pogonomyrmex ants. Five main questions were addressed: whether Wolbachia infection rates vary between North and South America, whether infection rates are dependent on host range, whether Wolbachia affects the caste determination of P. barbatus, whether infection rates in Pogonomyrmex are similar to those of other ants, and whether Wolbachia phylogeny parallels the phylogeny of its Pogonomyrmex hosts. Using PCR amplification of the wsp, ftsZ, and gatB loci, Wolbachia infections were detected in four of fifteen Pogonomyrmex species (26.7%), providing the first known evidence of Wolbachia infection in this genus. All infected species were from South America, specifically Argentina. Therefore, Wolbachia has no role in the caste determination of the North American species P. barbatus. Additionally, while it appears that the incidence of Wolbachia in Pogonomyrmex may be limited to South America, host range did not correlate with infection status. The incidence of Wolbachia in Pogonomyrmex as a whole was similar to that of invasive Solenopsis and Linepithema species, but not to Wasmannia auropunctata or Anoplolepis gracilipes, which retain Wolbachia infection in non-native locations. This suggests that there may be a parallel in Wolbachia infection spread in certain short-term models of species colonization and long-term models of genus radiation. Finally, there was no congruity between host and parasite phylogeny according to maximum likelihood analyses, necessarily due to horizontal transfer of Wolbachia between hosts and lateral gene transfer between Wolbachia strains within hosts.

All organisms perform best at a balanced point of intake where nutrients are ingested in specific amounts to confer optimal performance. However, when faced with limited nutrient availability, organisms are forced to make decisions which prioritize intake of certain macronutrients. While intake regulation has been more thoroughly studied in omnivores and carnivores, no research exists regarding lipid regulation in generalist herbivores. Traditionally, proteins and carbohydrates were thought to be the most important macronutrient for herbivore intake; however the large differences in lipid nutritional content between different plant species offers lots of potential for regulation of an important macronutrient. We studied whether generalist herbivores can regulate lipid intake, using the migratory locust (Locusta migratoria). Though herbivore protein and carbohydrate intake is well studied, less research studies regulation of lipid intake. We tested this by offering choice diets of varying carbohydrate and lipid content makeup and measuring consumption of each diet choice to determine overall carbohydrate and lipid intake. Four different lipid sources were used in order to control for taste or texture related confounds; canola oil, sunflower oil, grapeseed oil, and a lab designed synthetic oil based on the four most abundant fatty acids in common plant oils. On three out of four diet sources, groups evidences strong regulation of narrow intake target, with little disparity in overall intake of carbohydrate and lipid content between various choice diet treatments. Groups feeding on canola oil and sunflower oil based diets displayed the best regulation based on their having small disparities in intake between treatments, while those feeding on grapeseed oil based diets displayed wide variation in feeding behavior between treatments. Groups feeding on the synthetic oil based diet choice unexpectedly consumed much more carbohydrates than lipids when compared to all other groups. In conclusion, generalist herbivores are capable of regulating lipid intake.

Mathematical epidemiology, one of the oldest and richest areas in mathematical biology, has significantly enhanced our understanding of how pathogens emerge, evolve, and spread. Classical epidemiological models, the standard for predicting and managing the spread of infectious disease, assume that contacts between susceptible and infectious individuals depend on their relative frequency in the population. The behavioral factors that underpin contact rates are not generally addressed. There is, however, an emerging a class of models that addresses the feedbacks between infectious disease dynamics and the behavioral decisions driving host contact. Referred to as “economic epidemiology” or “epidemiological economics,” the approach explores the determinants of decisions about the number and type of contacts made by individuals, using insights and methods from economics. We show how the approach has the potential both to improve predictions of the course of infectious disease, and to support development of novel approaches to infectious disease management.

Insects are small creatures highly susceptible to water loss. A major factor in the prevention of water loss through an insect’s cuticle are their cuticular hydrocarbons (CHC), a lipid layer consisting mostly of long-chain hydrocarbons. CHCs consist of different molecules called alkanes, alkenes, and methyl branched hydrocarbons which all have varying levels of hydrophobicity. Ants are a massively abundant family of insects with important roles in the ecosystem that also utilize CHCs. Camponotus floridanus isare athe native ant species of the Florida Keys which areis known to have variable environmental temperature. Being exposed to temperatures as high as 35 °C, these ants are expected to have mechanisms that allow them to adapt to their environment. It was hypothesized that CHCs may change in concentration or composition as a means to combat the changes in cuticular permeability due to the variable temperatures that the ants experience. We therefore used C. floridanus worker ants to learn more about CHC plasticity in insects when exposed to elevated temperatures. We found four CHC componentspeaks that showed a statistically significant increase in concentration when comparing the control to treatment colonies: 3,7 dimethyl C31, an underdetermined methyl branched C31, 3,7,11 trimethyl C31, and an undetermined tetramethylbranched C31. These significant changes in concentration occurred on longer chain hydrocarbons. Under further examination, it was found that there was a strong positive correlation between elution time and the differences in medians of peak area between control and treatment colonies. This shows that there was a shift in the CHC profile resulting in an increased concentration of longer chained methyl-branched hydrocarbons. It also suggests that branched hydrocarbons also play some role in the water proofing mechanism of C. floridanus.

Hypoxia-responses help coordinate the growth of oxygen-transporting tissues with the growth of other tissues during development. In Drosophila, hypoxia strongly affects development with flies being reared in a low oxygen environment showing smaller body sizes and diminished tracheal growth. The primary regulator of cellular hypoxic-responses is the hypoxia-inducible factor (HIF), and under normoxic conditions, HIF-alpha is hydroxylated by prolyl hydroxylase domain (PHD) on a proline residue inside the alpha leading to the proteins proteasome degradation downstream. However, in response to reduced oxygen, cells accumulate HIF- alpha, which then joins with the constituent HIF-beta in the cytosol, forming a HIF- alpha/beta heterodimer. Which, in turn, enters the nucleus and binds to hypoxic response elements, activating the hypoxic response genes. Hyperoxia has recently been shown to stimulates metabolic rates only at the last stage Drosophila's larval development (L3), indicating oxygen limitation occurs towards the end of development. Green fluorescent protein (GFP) was added to the oxygen-dependent domain of Drosophila HIF- Alpha (Sima) and a monomeric red fluorescent protein with a nuclear localization signal (mRFP-nls) was added to a protein under the same ubiquitin-69E promoter but is not affected by changing O2 levels. Using a Leica SP5 AOBS Spectral Confocal, third instar larvae were analyzed at the cellular level with attention focused on HIF- signaling in the central nervous system (CNS). L3 Drosophila were divided into groups of 0-12h, 12-24h, 24-48h, and 48-60h corresponding to their development. In each group, flies were either treated for 10-12 hours in 5% O2 or were left normoxic before fixation. What was overwhelmingly found is that HIF-signaling was most prominent during their early development (0-12h), with a significant decline as age increased (P=<0.001). There was also an observed hypoxic effect as animals treated in lower oxygen concentrations had significantly higher HIF signaling (P=<0.001). However, this effect still declines as larvae continued developing. This data supports the idea that internal hypoxia does not become severe during late third instar growth but may occur during the actual molt of the flies.

While non-invasive breast cancer treatments may be considered less costly in the short-term, over the course of a lifetime, a more aggressive treatment can be overall less costly, especially with recurrence cases; however, these more aggressive treatments are not necessarily covered by insurance and are difficult to discuss in the short amount of time in physician consultations. This analysis studied data from 982 women diagnosed with breast cancer over a five-year period to evaluate monetary costs associated with treatment options and incorporated five in-depth interviews to understand experiences and non-monetary costs. Data showed the most expensive option was a unilateral mastectomy with radiation therapy and the least costly option was breast conserving surgery. Interviews determined each woman evaluated the monetary costs with each treatment but most heavily focused on personal values, biases and recommended opinions when deciding on a treatment. The use of prompt sheets before physician appointments and consultations, along with the addition of financial counselor meeting with each patient can improve patient satisfaction and alleviate stress by simplifying a woman's choice in deciding a treatment. In addition, increased insurance coverage to include every treatment chosen by women (rather than on a case-by-case basis), specifically contralateral prophylactic mastectomy and additional screening options, could decrease long term costs \u2014 both monetarily and in quality of life for patients.

In 2016, in the United States alone, the cosmetics industry made an estimated 62.46 billion dollars in revenue (Revenue of the Cosmetic Industry in the U.S. 2002-2016 | Forecast). With a consistent increase in sales in the last several years, the industry has reached continued success even during times of hardship, such as the Great Recession of 2008. The use of Corporate Social Responsibility (CSR), external campaigns, and thoughtful packaging and ingredients resonates with targeted consumers. This has served as an effective strategy to maintain growth in the industry. Cosmetic companies promote their brand image using these sustainability tactics, but there seems to be a lack of transparency in this unregulated industry. The purpose of this thesis is to determine if the cosmetics industry is a good steward of the sustainability movement. Important terms and concepts relating to the industry will be discussed, then an analysis of sustainability focused cosmetic brands will be provided, which highlights the extent to which these brands engage in activities that promote sustainability. This is followed by an application of findings to a company that could benefit from using such practices. Overall, the analysis of the different brands proved to be shocking and disappointing. This is due to the sheer amount that scored very poorly based on the sustainability criteria developed. The cosmetics industry is too inconsistent and too unregulated to truly act as a good steward for sustainability. Though some companies in the industry succeed, these accomplishments are not consistent across all cosmetic companies. Hence, the cosmetics industry as a good steward for sustainability can only be as strong as its weakest link.

This report was commissioned to provide an analysis and evaluation of consumer perceptions and branding as it relates to the political and social climate in America. To be able to do this, the paper analyzes shifts in the external environment as well as researching case studies and online consumer perception surveys. Overall, this paper aims to examine the distributed survey and attempt to correlate and identify how branding, consumer perceptions, and social and political issues all can work and affect one another. Through the administration of this survey, we were able to formulate a conclusion that points towards the importance of brands actively adhering to changing consumer preferences, ideals, and expectations.
