The bulk of this thesis is the translation itself—of Episodes 8-10—with timed subtitles. However, I also wanted to provide commentary on a few issues of interest that I came across during the translation. Conveying meaning from one language to the other does not simply involve translating the denotations of each word, but also the social context, humor, tone, and other aspects surrounding the situation. Whether words are used individually to interject, or come together to form unique idioms or phrases, the challenge presented to the translator is the choices: what words do I select to best convey not only the dictionary sense, but also the contextual meaning? This is especially difficult in wordplay-type situations. If there’s no perfect option, can I find a cultural analog that differs from the original denotation, but preserves the intention of the words? Otherwise, do I find the original important enough that it is worth educating the reader in order to preserve it—or, do I see no other translation option but to educate as a last resort? These are all choices that I had to make, and in this thesis I will be discussing the most interesting situations that came up, and my choices in dealing with them.

Influencer marketing has exploded into a multi-billion dollar industry, with millions of influencers spanning thousands of different industries. The beauty industry is one of the most popular industries for influencer marketing, with influencers having the ability to collaborate with popular beauty brands like L’Oréal as well as being able to create their own successful beauty companies. With consumers exposed to thousands of brand deals every day, brands are now searching for the keys to creating an authentic brand partnership with influencers. This thesis looks at the factors that go into authentic influencer marketing to help brands understand what they should be looking for when creating a partnership that consumers would deem as authentic, with a specific emphasis on the beauty industry. This thesis includes a multi-method study using a focus group and a survey. Specifically, the focus group focused on finding common themes related to authenticity and the survey uncovered whether follower count influences consumers’ perception of authenticity. The studies revealed that follower count doesn’t impact how consumers’ perceived authenticity, but authenticity and trust do play a role in whether consumers will interact with an influencer’s paid advertisement. To better understand the research presented in this thesis, an analysis of influencers and the psychology of influence are presented, along with a case study of a successful beauty brand partnership and an in depth look at FTC guidelines.

This Barrett thesis is a praxis-oriented research project designed to address issues that women face in hip hop and to give voice to female artists in the Arizona hip hop community specifically. In creating this project, I sought to encourage women in my community to create a conversation about their roles in the hip hop scene, to listen to and unite with other female artists in the valley, and to help create the networks and spaces that center the diverse narratives female hip hop artists express which the industry at large currently fails to represent. This paper examines similar research done in the field of hip hop sexism and hip hop feminism across the world, with an emphasis on US hip hop culture, to identify what many hip hop scholars and feminists point to as the sources of sexism in hip hop and the systems that maintain it, focusing on (1) the rap industry’s favoring of sexist lyrics and disfavoring of womxn’s points of view throughout the commercialization of hip hop culture and music, (2) the media’s discrimination against female hip hop artists in their coverage, and (3) the unequal distribution of space and resources allocated to women of color in hip hop resulting from black men’s need for those spaces and resources in order to reassert their masculinity in the face of their exclusion from the widley accepted white hegemonic masculinity they have been barred from. Combining the methods of successful activists in the US with findings from a series of interviews with women from the Arizona hip hop scene, I make recommendations on actions to take in the Arizona scene in order to combat the sexism found in hip hop while maintaining a pro-hip-hop stance throughout.

This paper explores a cluster analysis and data-driven marketing recommendations for Cartel Coffee Lab, a local coffee shop. Building a loyal customer base is the biggest asset to a company’s success. This is crucial when Cartel’s main location has a fast-changing environment, such as a university, located nearby. The reason for this is mainly due to the influx and customer churn that a hub produces. Our team’s focus was on analyzing customer buying patterns to determine the different customer segments within Cartel Coffee Lab. Our methods included creating temporary databases in Microsoft SQL Server and performing a cluster analysis in SAS Enterprise Miner. The cluster analysis results were then used to propose data-driven marketing recommendations for each customer segment in order to provide a well-designed value proposition. Additionally, a loyalty program was recommended in order to increase customer profitability and loyalty, depending on the cluster segment. Through identifying the current situation, business questions, research methodology, analysis results, and plan of action, this creative project will cover multiple key areas of a business analytics project.

In “The Trouble of Wilderness,” William Cronon (1995) states the concept of wilderness, historically, is based on the romanticized ideal of the “untamed” frontier that influenced the American expansion ideals in the late 1800s and early 1900s, including the initial conservation movement. This idea of wilderness is defined by “empty” lands that needed to be utilized by the civilized Anglo-Americans, or lands that needed to be preserved from human alterations. Wilderness was separate from humans and, therefore, was also thought to be land that had been unaltered by human touch. The disappearing frontier was being turned into farmlands and civilization, so the Anglo-Americans, the ones who culturally viewed undeveloped land as a place for recreation, wished to save the ‘wilderness’ that was not yet being used. But as will be discussed it was in fact being used just not by the Anglo-Americans. This wilderness that they were trying to preserve became the national parks, such as Yellowstone and Yosemite. Under this rationale, Indigenous peoples were forced off the land to create the illusion of these places fitting this romanticized idea of wilderness. This essay examines the national parks in context of this concept of wilderness being free from humans and how national parks rationalized the removal of Indigenous people from these “wild” lands by using this concept of wilderness. Specifically, it uses the history of Yellowstone and Yosemite parks, which are some of the first parks to enter the National Park System, as sites of understanding how the idea wilderness was conceptualized by the American government during the late 1800s as places that are separate from humans. This essay argues that these ideals are based on racist and xenophobic approaches that the early United States government used in regards to relationships with Indigenous people. To discuss these ideas, this paper will examine the language used in early government documents regarding the policies of the national parks along with art and writings from this time period to show how the public and government viewed these national parks and the Indigenous people in the surrounding areas. Particularly, this paper will consider the original documents that established the national parks and the language that was used in these documents. It will then compare these policies from the origins of the national parks to the policies in place now regarding Indigenous people, such as the reparations that are trying to be made in these areas.

Though high doses of ketorolac can be toxic,

NIPAAm co-DEAEMA hydrogels are a potential solution for sustained, local delivery of ketorolac tromethamine. Current methods of postoperative pain management, such as local anesthetics, NSAIDs, and opioids, can be improved by minimizing side effects while still effectively treating severe and extreme pain. Though high doses of ketorolac can be toxic, sustained, local delivery via hydrogels offers a promising solution. Four ketorolac release studies were conducted using PNDJ hydrogels formulated by Sonoran Biosciences. The first two studies tested a range of JAAm concentration between 1.4 and 2.2 mole percent. Both had high initial release rates lasting less than 7 days and appeared to be unaffected by JAAm content. Tobramycin slowed down the release of ketorolac but was unable to sustain release for more than 6 days. Incorporating DEAEMA prolonged the release of ketorolac for up to 14 days with significant reductions in initial burst release rate. Low LCST of NIPAAM co-DEAEMA polymer is problematic for even drug distribution and future in vivo applications.

Chemoreception is an important method for an octopus to sense and react to its surroundings. However, the density of chemoreceptors within different areas of the skin of the octopus arm is poorly documented. In order to assess the relative sensitivity of various regions and the degree to which chemoreception is locally controlled, octopus arms were amputated and exposed to acetic acid, a noxious chemical stimulus that has previously been shown to elicit movement responses in amputated arms (Hague et al., 2013). To test this, 11 wild-caught Octopus bimaculoides (6 females, 5 males) were obtained. Acetic acid vapor was introduced in the distal oral, distal aboral, proximal oral, and proximal aboral regions of amputated arms. The frequency of the occurrence of movement was first analyzed. For those trials in which movement occurred, the latency (delay between the stimulus and the onset of movement) and the duration of movement were analyzed. The distal aboral and distal oral regions were both more likely to move than either the proximal oral or proximal aboral regions (p < 0.0001), and when they did move, were more likely to move for longer periods of time (p < 0.05). In addition, the proximal oral region was more likely to exhibit a delay in the onset of movement compared to the distal oral or distal aboral regions (p < 0.0001). These findings provide evidence that the distal arm is most sensitive to noxious chemical stimuli. However, there were no significant differences between the distal oral and distal aboral regions, or between the proximal oral and proximal aboral regions. This suggests that there may not be a significant difference in the density of chemoreceptors in the aboral versus oral regions of the arm, contrary to claims in the literature. The other independent variables analyzed, including sex, body mass, arm length, anterior versus posterior arm identity, and left versus right arm identity, did not have a significant effect on any of the three dependent variables analyzed. Further analysis of the relative density of chemoreceptors in different regions of the octopus arm is merited.

This study aims to examine how the use of consensus-based transactions, smart contracts,and interoperability, provided by blockchain, may benefit the blood plasma industry. Plasmafractionation is the process of separating blood into multiple components to garner benefitsof increased lifespan, specialized allocation, and decreased waste, thereby creating a morecomplex and flexible supply chain. Traditional applications of blockchain are developed onthe basis of decentralization—an infeasible policy for this sector due to stringent governmentregulations, such as HIPAA. However, the trusted nature of the relations in the plasmaindustry’s taxonomy proves private and centralized blockchains as the viable alternative.Implementations of blockchain are widely seen across pharmaceutical supply chains to combatthe falsification of possibly afflictive drugs. This system is more difficult to manage withblood, due to the quick perishable time, tracking/tracing of recycled components, and thenecessity of real-time metrics. Key attributes of private blockchains, such as digital identity,smart contracts, and authorized ledgers, may have the possibility of providing a significantpositive impact on the allocation and management functions of blood banks. Herein, we willidentify the economy and risks of the plasma ecosystem to extrapolate specific applications forthe use of blockchain technology. To understand tangible effects of blockchain, we developeda proof of concept application, aiming to emulate the business logic of modern plasma supplychain ecosystems adopting a blockchain data structure. The application testing simulates thesupply chain via agent-based modeling to analyze the scalability, benefits, and limitations ofblockchain for the plasma fractionation industry.

The product is a compact cooking set that keeps utensils safe from the common cross-contamination that can be life-threatening to a person with celiac disease. The application allows people to find new restaurants that will be safe for them to eat at, scan the barcode of common food products for 8 of the most common allergens, and provide the most up to date research about celiac disease. Within the app, they can also find a sense of community by connecting with other members.

The primary regulator of cellular hypoxic-responses is the hypoxia-inducible factor (HIF),

Hypoxia-responses help coordinate the growth of oxygen-transporting tissues with the growth of other tissues during development. In Drosophila, hypoxia strongly affects development with flies being reared in a low oxygen environment showing smaller body sizes and diminished tracheal growth. The primary regulator of cellular hypoxic-responses is the hypoxia-inducible factor (HIF), and under normoxic conditions, HIF-alpha is hydroxylated by prolyl hydroxylase domain (PHD) on a proline residue inside the alpha leading to the proteins proteasome degradation downstream. However, in response to reduced oxygen, cells accumulate HIF- alpha, which then joins with the constituent HIF-beta in the cytosol, forming a HIF- alpha/beta heterodimer. Which, in turn, enters the nucleus and binds to hypoxic response elements, activating the hypoxic response genes. Hyperoxia has recently been shown to stimulates metabolic rates only at the last stage Drosophila's larval development (L3), indicating oxygen limitation occurs towards the end of development. Green fluorescent protein (GFP) was added to the oxygen-dependent domain of Drosophila HIF- Alpha (Sima) and a monomeric red fluorescent protein with a nuclear localization signal (mRFP-nls) was added to a protein under the same ubiquitin-69E promoter but is not affected by changing O2 levels. Using a Leica SP5 AOBS Spectral Confocal, third instar larvae were analyzed at the cellular level with attention focused on HIF- signaling in the central nervous system (CNS). L3 Drosophila were divided into groups of 0-12h, 12-24h, 24-48h, and 48-60h corresponding to their development. In each group, flies were either treated for 10-12 hours in 5% O2 or were left normoxic before fixation. What was overwhelmingly found is that HIF-signaling was most prominent during their early development (0-12h), with a significant decline as age increased (P=<0.001). There was also an observed hypoxic effect as animals treated in lower oxygen concentrations had significantly higher HIF signaling (P=<0.001). However, this effect still declines as larvae continued developing. This data supports the idea that internal hypoxia does not become severe during late third instar growth but may occur during the actual molt of the flies.
