Following the implementation of federal immigration control measures in the 1990s, Arizona became the main point of entry for undocumented immigrants along the US border with Mexico in the early 2000s. Since then, reports have blamed human smuggling facilitators for the increase of undocumented immigration into the state and the apparent development of violent practices targeting the undocumented. However, little is known about the organization of the groups who work at facilitating the transit of undocumented immigrants along the US Mexico Border. Based on interviews and narratives present in legal files of smuggling cases prosecuted in Phoenix, Arizona, the present study provides an analysis of local human smuggling operations. It argues that far from being under the control of organized crime, smuggling is an income generating strategy of the poor that generates financial opportunities for community members in financial distress. The study, raises questions over smuggling's perceptions as violent and instead identifies smuggling-related violence as a reflection of the structural violence carried out by the state against immigrant communities through policing, surveillance and the consistent and systematic exercise of race-based policies.

Masculinity has been increasingly recognized as a critical and relatively unexplored area of inquiry in anthropological gender studies. This project seeks to expand anthropological research on masculinity to contemporary American society. Using the case study of a male-centered popular new sport, Mixed Martial Arts (also known as cagefighting) this project integrates theories of embodiment and feminist perspectives to explore how masculinity and masculine hegemony are shaped, contested, and perpetuated in the United States. Using a multi-level framework this project explores: 1) How is masculinity experienced and expressed by Mixed Martial Arts fighters as a form of self-identity? How do their bodies play a role in constructing masculinity? 2) What are the pervasive forms of masculinity associated with Mixed Martial Arts (MMA)? Are they truly representative of the sport? 3) Can these pervasive forms of masculinity be seen as hegemonic? How would hegemony operate in relation to individual experience? Using multiple methods to capture multiple points of view was critical to thoroughly examining the complex notion of masculinity. This study employed participant observation, semi-structured interviews, focus groups, surveys, photo elicitation, and media content analysis, as each presented particular benefits and allowed for the development a more well-rounded understanding of masculinity within the realm of MMA. This study also situates the rise of MMA and its representations of masculinity within the greater perspective of contemporary American society. By doing so reveals how ideologies of prescribed masculinity do not arise out of a vacuum but in relation to particular economic, social and political contexts. An emphasis of this study was to examine the daily lives of MMA fighters to understand how their participation in what may be regarded as a hypermasculine activity affects their own perceptions of masculinity. In looking at how masculinity is embodied, the gaps and often contradictions between representation and individual experiences are revealed. Ultimately, the goal of this research is to contribute to a better understanding of masculinity as both an embodied and relational construct.

How does it relate to others in the plant kingdom? How do other cultures use this plant? The most many of us know about them is that they are at the supermarket when we need them for dinner (Nabhan, 2009) (Vileisis, 2008).

Restraint stress is the most commonly used laboratory stressor. It is difficult to characterize as psychological or physical, because past studies show psychological features, but the nature of confinement adds a physical dimension. This was the first study to investigate how experience with restraint stress affects brain response to the next stress without a physical burden. Pair-housed adult male rats were transported to a novel context and restrained or left undisturbed (6hr). The next day, rats were returned to the same context and were either restrained or left undisturbed in the context (n=8/group). After 90min, rats were euthanized to determine functional activation in limbic structures using Fos immunohistochemistry and to measure HPA axis reactivity through blood serum corticosterone levels. Regardless of day 1 experience, context exposure on day 2 enhanced Fos expression in CA1 and CA3 of the hippocampus, basolateral amygdala, and central amygdala. Conversely, other regions and corticosterone levels demonstrated modulation from the previous day's experience. Specifically, rats that were placed back into the restraint context but not restrained on day 2 showed enhanced Fos expression in the dentate gyrus suprapyramidal blade (DGSup), and infralimbic cortex (IL). Also Fos expression was attenuated in rats that received two restraint exposures in the IL and medial amygdala (MEA), suggesting habituation. Only the DG infrapyramidal blade (DGInf) showed enhanced Fos expression to restraint on day 2 without influence of the previous day. While context predominately directed Fos activation, prior experience with restraint influenced Fos expression in the DGSup, IL, MEA and corticosterone levels to support restraint having psychological components.

Two-component regulatory systems (TCR systems) are the most ubiquitous signal transduction systems which regulate many genes required for virulence and survival of bacteria. In this study, I analyzed different TCR systems in two clinically-relevant Gram-negative bacteria, i.e.,

Pathogenic Gram-negative bacteria employ a variety of molecular mechanisms to combat host defenses. Two-component regulatory systems (TCR systems) are the most ubiquitous signal transduction systems which regulate many genes required for virulence and survival of bacteria. In this study, I analyzed different TCR systems in two clinically-relevant Gram-negative bacteria, i.e., oral pathogen Porphyromonas gingivalis and enterobacterial Escherichia coli. P. gingivalis is a major causative agent of periodontal disease as well as systemic illnesses, like cardiovascular disease. A microarray study found that the putative PorY-PorX TCR system controls the secretion and maturation of virulence factors, as well as loci involved in the PorSS secretion system, which secretes proteinases, i.e., gingipains, responsible for periodontal disease. Proteomic analysis (SILAC) was used to improve the microarray data, reverse-transcription PCR to verify the proteomic data, and primer extension assay to determine the promoter regions of specific PorX regulated loci. I was able to characterize multiple genetic loci regulated by this TCR system, many of which play an essential role in hemagglutination and host-cell adhesion, and likely contribute to virulence in this bacterium. Enteric Gram-negative bacteria must withstand many host defenses such as digestive enzymes, low pH, and antimicrobial peptides (AMPs). The CpxR-CpxA TCR system of E. coli has been extensively characterized and shown to be required for protection against AMPs. Most recently, this TCR system has been shown to up-regulate the rfe-rff operon which encodes genes involved in the production of enterobacterial common antigen (ECA), and confers protection against a variety of AMPs. In this study, I utilized primer extension and DNase I footprinting to determine how CpxR regulates the ECA operon. My findings suggest that CpxR modulates transcription by directly binding to the rfe promoter. Multiple genetic and biochemical approaches were used to demonstrate that specific TCR systems contribute to regulation of virulence factors and resistance to host defenses in P. gingivalis and E. coli, respectively. Understanding these genetic circuits provides insight into strategies for pathogenesis and resistance to host defenses in Gram negative bacterial pathogens. Finally, these data provide compelling potential molecular targets for therapeutics to treat P. gingivalis and E. coli infections.

The study of bacterial resistance to antimicrobial peptides (AMPs) is a significant area of interest as these peptides have the potential to be developed into alternative drug therapies to combat microbial pathogens. AMPs represent a class of host-mediated factors that function to prevent microbial infection of their host and serve as a first line of defense. To date, over 1,000 AMPs of various natures have been predicted or experimentally characterized. Their potent bactericidal activities and broad-based target repertoire make them a promising next-generation pharmaceutical therapy to combat bacterial pathogens. It is important to understand the molecular mechanisms, both genetic and physiological, that bacteria employ to circumvent the bactericidal activities of AMPs. These understandings will allow researchers to overcome challenges posed with the development of new drug therapies; as well as identify, at a fundamental level, how bacteria are able to adapt and survive within varied host environments. Here, results are presented from the first reported large scale, systematic screen in which the Keio collection of ~4,000 Escherichia coli deletion mutants were challenged against physiologically significant AMPs to identify genes required for resistance. Less than 3% of the total number of genes on the E. coli chromosome was determined to contribute to bacterial resistance to at least one AMP analyzed in the screen. Further, the screen implicated a single cellular component (enterobacterial common antigen, ECA) and a single transporter system (twin-arginine transporter, Tat) as being required for resistance to each AMP class. Using antimicrobial resistance as a tool to identify novel genetic mechanisms, subsequent analyses were able to identify a two-component system, CpxR/CpxA, as a global regulator in bacterial resistance to AMPs. Multiple previously characterized CpxR/A members, as well as members found in this study, were identified in the screen. Notably, CpxR/A was found to transcriptionally regulate the gene cluster responsible for the biosynthesis of the ECA. Thus, a novel genetic mechanism was uncovered that directly correlates with a physiologically significant cellular component that appears to globally contribute to bacterial resistance to AMPs.

"Culture talk" figures prominently in the discussions of and about Muslims, both locally and globally. Culture, in these discussions, is considered to be the underlying cause of gender and generational divides giving rise to an alleged "identity crisis." "Culture talk" figures prominently in the discussions of and about Muslims, both locally and globally. Culture, in these discussions, is considered to be the underlying cause of gender and generational divides giving rise to an alleged "identity crisis." Culture also presumably conceals and contaminates "pure/true Islam." Culture serves as the scaffold on which all that divides Muslim American immigrants and converts is built; furthermore, the fear of a Muslim cultural takeover underpins the "Islamization of America" narrative. This dissertation engages these generational and "immigrant"-"indigenous" fissures and the current narratives that dominate Muslim and public spheres. It does so through the perspectives of the offspring of converts and immigrants. As the children and grandchildren of immigrants and converts come of age, and distant as they are from historical processes and experiences that shaped the parents' generations while having shared a socialization process as both Muslim and American, what role do they play in the current chapter of Islam in post-9/11 America? Will the younger generation be able to cross the divides, mend the fissures, and play a pivotal role in an "American Muslim community"? Examining how younger generations of both backgrounds view each other and their respective roles in forging an American Muslim belonging, agenda and discourse is a timely and much needed inquiry. This project aims to contribute by shedding more light on the identities, perspectives and roles of these younger generations through the four dominant narratives of identity crisis, pure/true Islam vs. Cultural Islam, the Islamization of America, and creation of an American Muslim community/identity/culture. These narratives are both part of public discourse and themes generated from interviews, a questionnaire\survey, and personal observation. This ethnographic study examines how American born and/or raised offspring of both converts to Islam and immigrant Muslims in the Phoenix and Chicago metropolitan areas define self and community, how they negotiate fissures and fault lines (ethnicity, race, class, gender, and religious interpretation) within their communities, and how their faith informs daily life and envisions a future. I utilize participant observation, interviews, and surveys and examine digital, visual and published media to answer these questions.

Food system and health characteristics were evaluated across the last Waorani hunter-gatherer group in Amazonian Ecuador and a remote neighboring Kichwa indigenous subsistence agriculture community. Hunter-gatherer food systems like the Waorani foragers may not only be nutritionally, but also pharmaceutically beneficial because of high dietary intake of varied plant phytochemical compounds. A modern diet that reduces these dietary plant defense phytochemicals below levels typical in human evolutionary history may leave humans vulnerable to diseases that were controlled through a foraging diet. Few studies consider the health impact of the recent drastic reduction of plant phytochemical content in the modern global food system, which has eliminated essential components of food because they are not considered "nutrients". The antimicrobial and anti-inflammatory nature of the food system may not only regulate infectious pathogens and inflammatory disease, but also support beneficial microbes in human hosts, reducing vulnerability to chronic diseases. Waorani foragers seem immune to certain infections with very low rates of chronic disease. Does returning to certain characteristics of a foraging food system begin to restore the human body microbe balance and inflammatory response to evolutionary norms, and if so, what implication does this have for the treatment of disease? Several years of data on dietary and health differences across the foragers and the farmers was gathered. There were major differences in health outcomes across the board. In the Waorani forager group there were no signs of infection in serious wounds such as 3rd degree burns and spear wounds. The foragers had one-degree lower body temperature than the farmers. The Waorani had an absence of signs of chronic diseases including vision and blood pressure that did not change markedly with age while Kichwa farmers suffered from both chronic diseases and physiological indicators of aging. In the Waorani forager population, there was an absence of many common regional infectious diseases, from helminthes to staphylococcus. Study design helped control for confounders (exercise, environment, genetic factors, non-phytochemical dietary intake). This study provides evidence of the major role total phytochemical dietary intake plays in human health, often not considered by policymakers and nutritional and agricultural scientists.
