Infections caused by the Hepatitis C Virus (HCV) are very common worldwide, affecting up to 3% of the population. Chronic infection of HCV may develop into liver cirrhosis and liver cancer which is among the top five of the most common cancers. Therefore, vaccines against HCV are under intense study in order to prevent HCV from harming people's health. The envelope protein 2 (E2) of HCV is thought to be a promising vaccine candidate because it can directly bind to a human cell receptor and plays a role in viral entry. However, the E2 protein production in cells is inefficient due to its complicated matured structure. Folding of E2 in the endoplasmic reticulum (ER) is often error-prone, resulting in production of aggregates and misfolded proteins. These incorrect forms of E2 are not functional because they are not able to bind to human cells and stimulate antibody response to inhibit this binding. This study is aimed to overcome the difficulties of HCV E2 production in plant system. Protein folding in the ER requires great assistance from molecular chaperones. Thus, in this study, two molecular chaperones in the ER, calreticulin and calnexin, were transiently overexpressed in plant leaves in order to facilitate E2 folding and production. Both of them showed benefits in increasing the yield of E2 and improving the quality of E2. In addition, poorly folded E2 accumulated in the ER may cause stress in the ER and trigger transcriptional activation of ER molecular chaperones. Therefore, a transcription factor involved in this pathway, named bZIP60, was also overexpressed in plant leaves, aiming at up-regulating a major family of molecular chaperones called BiP to assist protein folding. However, our results showed that BiP mRNA levels were not up-regulated by bZIP60, but they increased in response to E2 expression. The Western blot analysis also showed that overexpression of bZIP60 had a small effect on promoting E2 folding. Overall, this study suggested that increasing the level of specific ER molecular chaperones was an effective way to promote HCV E2 protein production and maturation.

Diseases have been part of human life for generations and evolve within the population, sometimes dying out while other times becoming endemic or the cause of recurrent outbreaks. The long term influence of a disease stems from different dynamics within or between pathogen-host, that have been analyzed and studied by many researchers using mathematical models. Co-infection with different pathogens is common, yet little is known about how infection with one pathogen affects the host's immunological response to another. Moreover, no work has been found in the literature that considers the variability of the host immune health or that examines a disease at the population level and its corresponding interconnectedness with the host immune system. Knowing that the spread of the disease in the population starts at the individual level, this thesis explores how variability in immune system response within an endemic environment affects an individual's vulnerability, and how prone it is to co-infections. Immunology-based models of Malaria and Tuberculosis (TB) are constructed by extending and modifying existing mathematical models in the literature. The two are then combined to give a single nine-variable model of co-infection with Malaria and TB. Because these models are difficult to gain any insight analytically due to the large number of parameters, a phenomenological model of co-infection is proposed with subsystems corresponding to the individual immunology-based model of a single infection. Within this phenomenological model, the variability of the host immune health is also incorporated through three different pathogen response curves using nonlinear bounded Michaelis-Menten functions that describe the level or state of immune system (healthy, moderate and severely compromised). The immunology-based models of Malaria and TB give numerical results that agree with the biological observations. The Malaria--TB co-infection model gives reasonable results and these suggest that the order in which the two diseases are introduced have an impact on the behavior of both. The subsystems of the phenomenological models that correspond to a single infection (either of Malaria or TB) mimic much of the observed behavior of the immunology-based counterpart and can demonstrate different behavior depending on the chosen pathogen response curve. In addition, varying some of the parameters and initial conditions in the phenomenological model yields a range of topologically different mathematical behaviors, which suggests that this behavior may be able to be observed in the immunology-based models as well. The phenomenological models clearly replicate the qualitative behavior of primary and secondary infection as well as co-infection. The mathematical solutions of the models correspond to the fundamental states described by immunologists: virgin state, immune state and tolerance state. The phenomenological model of co-infection also demonstrates a range of parameter values and initial conditions in which the introduction of a second disease causes both diseases to grow without bound even though those same parameters and initial conditions did not yield unbounded growth in the corresponding subsystems. This results applies to all three states of the host immune system. In terms of the immunology-based system, this would suggest the following: there may be parameter values and initial conditions in which a person can clear Malaria or TB (separately) from their system but in which the presence of both can result in the person dying of one of the diseases. Finally, this thesis studies links between epidemiology (population level) and immunology in an effort to assess the impact of pathogen's spread within the population on the immune response of individuals. Models of Malaria and TB are proposed that incorporate the immune system of the host into a mathematical model of an epidemic at the population level.

Surveys have shown that several hundred billion weather forecasts are obtained by the United States public each year, and that weather news is one of the most consumed topics in the media. This indicates that the forecast provides information that is significant to the public, and that the public utilizes details associated with it to inform aspects of their life. Phoenix, Arizona is a dry, desert region that experiences a monsoon season and extreme heat. How then, does the weather forecast influence the way Phoenix residents make decisions? This paper aims to draw connections between the weather forecast, decision making, and people who live in a desert environment. To do this, a ten-minute survey was deployed through Amazon Mechanical Turk (MTurk) in which 379 respondents were targeted. The survey asks 45 multiple choice and ranking questions categorized into four sections: obtainment of the forecast, forecast variables of interest, informed decision making based on unique weather variables, and demographics. This research illuminates how residents in the Phoenix metropolitan area use the local weather forecast for decision-making on daily activities, and the main meteorological factors that drive those decisions.

My thesis project, "Reforming the NCAA: How the governing body of intercollegiate athletics should handle student-athletes into the future," sets out to create a working blueprint for how the NCAA should handle the relationship between student-athletes, universities, and the NCAA from this point forward. The NCAA has come under fire in the past 10 years for its failure to meet modern social constructs and provide student-athletes with the resources necessary to achieving a successful educational and athletic experience, and through my thesis, I formulated reforms the NCAA can adopt and enact to respond to the growing issues within intercollegiate athletics. I began the process last spring with my thesis director as we selected a topic together, and I researched a variety of topics relating to current NCAA issues throughout the summer and fall. In the fall, I outlined sources I hoped to interview, and I conducted interviews over winter break. I spent the first two months of 2015 writing and refining my thesis, and through March, I created a PowerPoint presentation I used to defend my thesis project. During the process, I met with my thesis director at critical junctures to discuss the direction of the project and to determine how to find a delicate balance between creating attainable goals for reform and overstepping my boundaries. After a successful thesis defense, I made small revisions to the thesis and had my project re-approved by my director and second reader. In the coming weeks after submitting my thesis, I plan on exploring the possibility of having my thesis published and received critiques from those in the industry who follow collegiate athletics closely.

Solution methods for certain linear and nonlinear evolution equations are presented in this dissertation. Emphasis is placed mainly on the analytical treatment of nonautonomous differential equations, which are challenging to solve despite the existent numerical and symbolic computational software programs available. Ideas from the transformation theory are adopted allowing one to solve the problems under consideration from a non-traditional perspective. First, the Cauchy initial value problem is considered for a class of nonautonomous and inhomogeneous linear diffusion-type equation on the entire real line. Explicit transformations are used to reduce the equations under study to their corresponding standard forms emphasizing on natural relations with certain Riccati(and/or Ermakov)-type systems. These relations give solvability results for the Cauchy problem of the parabolic equation considered. The superposition principle allows to solve formally this problem from an unconventional point of view. An eigenfunction expansion approach is also considered for this general evolution equation. Examples considered to corroborate the efficacy of the proposed solution methods include the Fokker-Planck equation, the Black-Scholes model and the one-factor Gaussian Hull-White model. The results obtained in the first part are used to solve the Cauchy initial value problem for certain inhomogeneous Burgers-type equation. The connection between linear (the Diffusion-type) and nonlinear (Burgers-type) parabolic equations is stress in order to establish a strong commutative relation. Traveling wave solutions of a nonautonomous Burgers equation are also investigated. Finally, it is constructed explicitly the minimum-uncertainty squeezed states for quantum harmonic oscillators. They are derived by the action of corresponding maximal kinematical invariance group on the standard ground state solution. It is shown that the product of the variances attains the required minimum value only at the instances that one variance is a minimum and the other is a maximum, when the squeezing of one of the variances occurs. Such explicit construction is possible due to the relation between the diffusion-type equation studied in the first part and the time-dependent Schrodinger equation. A modication of the radiation field operators for squeezed photons in a perfect cavity is also suggested with the help of a nonstandard solution of Heisenberg's equation of motion.

The relationship between mental illness (or "madness," as Socrates labeled psychosis, as well as a wide range of states of thought and emotion [Jamison, 1993]) and creativity has been noted since before the times of Socrates. The questions that this noted relationship pose are significant. Famous authors, poets, artists, and musicians who suffered from various forms of mental illness are littered throughout history. Edgar Allen Poe himself noted, "Men have called me mad but the question is not yet settled, whether madness is or is not the loftiest intelligence \u2014 whether much that is glorious \u2014 whether all that is profound \u2014 does not spring from disease of thought \u2014 from moods of mind exalted at the expense of the general intellect." Only in recent years have researchers begun to classify these disorders of past artists using modern diagnostic measures. While the concept of a relationship between creativity and mental illness is fascinating in and of itself, understanding the implications of a possible correlation is essential to the care and treatment that individuals with the diseases are given. If, indeed, creativity is linked with affective disorders, what does this mean for the treatment of these disorders? By eliminating the symptoms of mood disorders, are we also inadvertently eliminating creativity? It is important to acknowledge the possibility that mental illnesses like bipolar disorder include benefits to those afflicted. How can we treat the negative symptoms of mood disorders while enhancing the positive symptoms? In order to conclude that there is a link between creativity and mood disorders, it is first necessary to reexamine and reestablish the processes by which mood disorders are diagnosed. Although currently diagnosed on a categorical scale \u2014 meaning the patient is diagnosed under the black-and-white category of manic or depressive at a given point in time \u2014 this paper will argue that mood disorders can be better diagnosed and treated on a continuum of mood states. Furthermore, by viewing mood disorders on such a continuum, writers like Anne Sexton and Sylvia Plath, whose diagnoses are debated by historians and professionals, can be established as having a diagnosable affective disorder, since both experienced symptoms of depression and mania, if not in the precise timing and manner proscribed by the Diagnostic Manual. Finally, by determining these writers' diagnoses, a relationship can be explored between their creative states and their mood states.

We applied regressions to over 35,000 data points to estimate the basic reproduction number and excess mortality. Age-specific mortality curves show elevated mortality for all age groups, especially the young, and senior sparing effects. The low value for reproduction number indicates that transmissibility was moderately low.

Background: While research has quantified the mortality burden of the 1957 H2N2 influenza pandemic in the United States, little is known about how the virus spread locally in Arizona, an area where the dry climate was promoted as reducing respiratory illness transmission yet tuberculosis prevalence was high.
Methods: Using archival death certificates from 1954 to 1961, this study quantified the age-specific seasonal patterns, excess-mortality rates, and transmissibility patterns of the 1957 pandemic in Maricopa County, Arizona. By applying cyclical Serfling linear regression models to weekly mortality rates, the excess-mortality rates due to respiratory and all-causes were estimated for each age group during the pandemic period. The reproduction number was quantified from weekly data using a simple growth rate method and generation intervals of 3 and 4 days. Local newspaper articles from The Arizona Republic were analyzed from 1957-1958.
Results: Excess-mortality rates varied between waves, age groups, and causes of death, but overall remained low. From October 1959-June 1960, the most severe wave of the pandemic, the absolute excess-mortality rate based on respiratory deaths per 10,000 population was 17.85 in the elderly (≥65 years). All other age groups had extremely low excess-mortality and the typical U-shaped age-pattern was absent. However, relative risk was greatest (3.61) among children and young adolescents (5-14 years) from October 1957-March 1958, based on incidence rates of respiratory deaths. Transmissibility was greatest during the same 1957-1958 period, when the mean reproduction number was 1.08-1.11, assuming 3 or 4 day generation intervals and exponential or fixed distributions.
Conclusions: Maricopa County largely avoided pandemic influenza from 1957-1961. Understanding this historical pandemic and the absence of high excess-mortality rates and transmissibility in Maricopa County may help public health officials prepare for and mitigate future outbreaks of influenza.

Armenia is a radiant and unique country, known primarily for its vast history, delicious food, and traditional folklore dance. Although Armenia was constantly ravaged by invading armies, the nation was strong enough to retain its own identity and culture. The rich dance heritage remained a living tradition into the 20th century, when the Turks tried to destroy the dance heritage and left fragments of the dances. Even though many of these fragments were lost due to urbanization, the dance techniques are still continuously studied by scholars and people all around the world.  For my creative project, I chose to perform an Armenian Folk Dance, as dance is an immense part of Armenian’s lives, and is an essential part of the Armenian cultural heritage. Dance is one of the primary ways by which one can observe and gain understanding of the Armenian people, their identity and culture. Dance is a way for Armenians to express themselves, as it is a form of art which combines elements of their rich culture, event-filled past, and their spiritual nature. The folklore dance has always sustained a leading role throughout Armenian history, as it runs through the blood of Armenians, being passed down from generation to generation. The Armenian people have survived and endured various historical challenges, but they have been able to preserve their unique dance elements and forms. To this day, Armenians continue to use dance to keep their culture and identity alive. It is a way to express freedom, and celebrate that even though we as a nation have faced many hardships, us Armenians have survived, and will always continue to survive with our faith, passion, determination, and strong will.
