The ecological impact of energy production and consumption is often relegated in analytical accounts of the evolution of energy systems, where production and consumption patterns are analysed as the interaction of social, economic and technological factors. Ecological and social–ecological dynamics are, we argue, critical in the context of imperatives for access to modern energy services that are inadequate for significant sections of the world's population. The ecological impacts of energy use are often analysed as a set of externalities, many of which are uncertain or unquantifiable, particularly if they stem from earth system change such as anthropogenic climate change. Here we outline the benefits from analysing energy systems as social–ecological systems. We review the extensive literature from ecology and resilience theories, and compare the analytical domains, major findings and emphasis of social–ecological systems with socio-technical transition research. We illustrate these differences with the example of the multi-scale impacts of biofuel expansion. We show that social–ecological systems research combines analysis of interactions with ecological systems and power relations between actors in energy systems, and has the potential to do so across production, distribution and consumption domains whilst illustrating the dynamics of such energy systems, identifying potential trade-offs and regime shifts.

The purpose of the United Nations-guided process to establish Sustainable Development Goals is to galvanize governments and civil society to rise to the interlinked environmental, societal, and economic challenges we face in the Anthropocene. We argue that the process of setting Sustainable Development Goals should take three key aspects into consideration. First, it should embrace an integrated social-ecological system perspective and acknowledge the key dynamics that such systems entail, including the role of ecosystems in sustaining human wellbeing, multiple cross-scale interactions, and uncertain thresholds. Second, the process needs to address trade-offs between the ambition of goals and the feasibility in reaching them, recognizing biophysical, social, and political constraints. Third, the goal-setting exercise and the management of goal implementation need to be guided by existing knowledge about the principles, dynamics, and constraints of social change processes at all scales, from the individual to the global. Combining these three aspects will increase the chances of establishing and achieving effective Sustainable Development Goals.

The conventional theory based on selfish rational actors cannot explain empirical observations. A more comprehensive theoretical framework of human behavior is emerging that include concepts such as trust, conditional cooperation, other-regarding preferences, social norms, and reputation. The new behavioral perspective also demonstrates that behavioral responses depend on social and biophysical context.

Smart growth policy and planning have tended to emphasize urban centers and regions, yet rural communities can also be important sites of innovation. Recent work demonstrated that these communities had surprisingly high levels of current and potential nonmotorized travel. Legislation in California mandates reductions in greenhouse gas emissions across all of the state's metropolitan planning organization (MPO) regions, including the heavily rural San Joaquin Valley. Advocates for rural communities are finding common cause with more traditional environmental organizations around the vision of investing in and enhancing extant rural places as an alternative to leapfrog patterns of urban and suburban sprawl. Because of existing patterns of extreme disparity and legion underserved unincorporated communities, analyses that can help integrate social equity within regional planning are needed to serve and empower rural residents. This paper presents the results of several new analyses of the social equity dimensions of regional transportation plans in the San Joaquin Valley. Activity-based travel model data were used to analyze equity, with a particular focus placed on eight disadvantaged unincorporated communities identified by community advocates to be important demonstration sites. The investigators showed how improvements to traditional equity analysis could enhance the consideration of equity in the planning process and compared the results developed by innovative techniques with those obtained by use of their traditional counterparts. The methods outlined here can make substantial contributions to reduce disparities in rural communities, which would likely be overlooked in typical regional equity analyses because of their small size, and offer lessons for MPOs serving rural areas across the country.

Faced with numerous seemingly intractable social and environmental challenges, many scholars and practitioners are increasingly interested in understanding how to actively engage and transform the existing systems holding such problems in place. Although a variety of analytical models have emerged in recent years, most emphasize either the social or ecological elements of such transformations rather than their coupled nature. To address this, first we have presented a definition of the core elements of a social-ecological system (SES) that could potentially be altered in a transformation. Second, we drew on insights about transformation from three branches of literature focused on radical change, i.e., social movements, socio-technical transitions, and social innovation, and gave consideration to the similarities and differences with the current studies by resilience scholars. Drawing on these findings, we have proposed a framework that outlines the process and phases of transformative change in an SES. Future research will be able to utilize the framework as a tool for analyzing the alteration of social-ecological feedbacks, identifying critical barriers and leverage points and assessing the outcome of social-ecological transformations.

Recently, there has been an increased interest in using behavioral experiments to study hypotheses on the governance of social-ecological systems. A diversity of software tools are used to implement such experiments. We evaluated various publicly available platforms that could be used in research and education on the governance of social-ecological systems. The aims of the various platforms are distinct, and this is noticeable in the differences in their user-friendliness and their adaptability to novel research questions. The more easily accessible platforms are useful for prototyping experiments and for educational purposes to illustrate theoretical concepts. To advance novel research aims, more elaborate programming experience is required to either implement an experiment from scratch or adjust existing experimental software. There is no ideal platform best suited for all possible use cases, but we have provided a menu of options and their associated trade-offs.

Urbanization continues to be a transformative process globally, affecting ecosystem integrity and the health and well being of people around the world. Although cities tend to be centers for both the production and consumption of goods and services that degrade natural environments, there is also evidence that urban ecosystems can play a positive role in sustainability efforts. Despite the fact that most of the urbanization is now occurring in the developing countries of the Global South, much of what we know about urban ecosystems has been developed from studying cities in the United States and across Europe. We propose a conceptual framework to broaden the development of urban ecological research and its application to sustainability. Our framework describes four key contemporary urban features that should be accounted for in any attempt to build a unified theory of cities that contributes to urban sustainability efforts. We evaluated a range of examples from cities around the world, highlighting how urban areas are complex, connected, diffuse and diverse and what these interconnected features mean for the study of urban ecosystems and sustainability.

Most studies on the response of socioeconomic systems to a sudden shift focus on long-term equilibria or end points. Such narrow focus forgoes many valuable insights. Here we examine the transient dynamics of regime shift on a divided population, exemplified by societies divided ideologically, politically, economically, or technologically. Replicator dynamics is used to investigate the complex transient dynamics of the population response. Though simple, our modeling approach exhibits a surprisingly rich and diverse array of dynamics. Our results highlight the critical roles played by diversity in strategies and the magnitude of the shift. Importantly, it allows for a variety of strategies to arise organically as an integral part of the transient dynamics-as opposed to an independent process-of population response to a regime shift, providing a link between the population's past and future diversity patterns. Several combinations of different populations' strategy distributions and shifts were systematically investigated. Such rich dynamics highlight the challenges of anticipating the response of a divided population to a change. The findings in this paper can potentially improve our understanding of a wide range of socio-ecological and technological transitions.

We sought to evaluate the reproducibility of a liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based approach to measure the stable-isotope enrichment of in vivo-labeled muscle ATP synthase β subunit (β-F1-ATPase), a protein most directly involved in ATP production, and whose abundance is reduced under a variety of circumstances. Muscle was obtained from a rat infused with stable-isotope-labeled leucine. The muscle was homogenized, β-F1-ATPase immunoprecipitated, and the protein was resolved using 1D-SDS PAGE. Following trypsin digestion of the isolated protein, the resultant peptide mixtures were subjected to analysis by HPLC-ESI-MS/MS, which resulted in the detection of multiple β-F1-ATPase peptides. There were three β-F1-ATPase unique peptides with a leucine residue in the amino acid sequence, and which were detected with high intensity relative to other peptides and assigned with >95% probability to β-F1-ATPase. These peptides were specifically targeted for fragmentation to access their stable-isotope enrichment based on MS/MS peak areas calculated from extracted ion chromatographs for selected labeled and unlabeled fragment ions. Results showed best linearity (R[superscript 2] = 0.99) in the detection of MS/MS peak areas for both labeled and unlabeled fragment ions, over a wide range of amounts of injected protein, specifically for the β-F1-ATPase[subscript 134-143] peptide. Measured stable-isotope enrichment was highly reproducible for the β-F1-ATPase[subscript 134-143] peptide (CV = 2.9%). Further, using mixtures of synthetic labeled and unlabeled peptides we determined that there is an excellent linear relationship (R[superscript 2] = 0.99) between measured and predicted enrichment for percent enrichments ranging between 0.009% and 8.185% for the β-F1-ATPase[subscript 134-143] peptide. The described approach provides a reliable approach to measure the stable-isotope enrichment of in-vivo-labeled muscle β-F1-ATPase based on the determination of the enrichment of the β-F1-ATPase[subscript 134-143] peptide.

Background: Healthy individuals on the lower end of the insulin sensitivity spectrum also have a reduced gene expression response to exercise for specific genes. The goal of this study was to determine the relationship between insulin sensitivity and exercise-induced gene expression in an unbiased, global manner.

Methods and Findings: Euglycemic clamps were used to measure insulin sensitivity and muscle biopsies were done at rest and 30 minutes after a single acute exercise bout in 14 healthy participants. Changes in mRNA expression were assessed using microarrays, and miRNA analysis was performed in a subset of 6 of the participants using sequencing techniques. Following exercise, 215 mRNAs were changed at the probe level (Bonferroni-corrected P<0.00000115). Pathway and Gene Ontology analysis showed enrichment in MAP kinase signaling, transcriptional regulation and DNA binding. Changes in several transcription factor mRNAs were correlated with insulin sensitivity, including MYC, r=0.71; SNF1LK, r=0.69; and ATF3, r= 0.61 (5 corrected for false discovery rate). Enrichment in the 5’-UTRs of exercise-responsive genes suggested regulation by common transcription factors, especially EGR1. miRNA species of interest that changed after exercise included miR-378, which is located in an intron of the PPARGC1B gene.

Conclusions: These results indicate that transcription factor gene expression responses to exercise depend highly on insulin sensitivity in healthy people. The overall pattern suggests a coordinated cycle by which exercise and insulin sensitivity regulate gene expression in muscle.
