Transient Receptor Potential (TRP) ion channels are a diverse family of nonselective, polymodal sensors in uni- and multicellular eukaryotes that are implicated in an assortment of biological contexts and human disease. The cold-activated TRP Melastatin-8 (TRPM8) channel, also recognized as the human body's primary cold sensor, is among the few TRP channels responsible for thermosensing. Despite sustained interest in the channel, the mechanisms underlying TRPM8 activation, modulation, and gating have proved challenging to study and remain poorly understood. In this thesis, I offer data collected on various expression, extraction, and purification conditions tested in E. Coli expression systems with the aim to optimize the generation of a structurally stable and functional human TRPM8 pore domain (S5 and S6) construct for application in structural biology studies. These studies, including the biophysical technique nuclear magnetic spectroscopy (NMR), among others, will be essential for elucidating the role of the TRPM8 pore domain in in regulating ligand binding, channel gating, ion selectively, and thermal sensitivity. Moreover, in the second half of this thesis, I discuss the ligation-independent megaprimer PCR of whole-plasmids (MEGAWHOP PCR) cloning technique, and how it was used to generate chimeras between TRPM8 and its nearest analog TRPM2. I review steps taken to optimize the efficiency of MEGAWHOP PCR and the implications and unique applications of this novel methodology for advancing recombinant DNA technology. I lastly present preliminary electrophysiological data on the chimeras, employed to isolate and study the functional contributions of each individual transmembrane helix (S1-S6) to TRPM8 menthol activation. These studies show the utility of the TRPM8\u2014TRPM2 chimeras for dissecting function of TRP channels. The average current traces analyzed thus far indicate that the S2 and S3 helices appear to play an important role in TRPM8 menthol modulation because the TRPM8[M2S2] and TRPM8[M2S3] chimeras significantly reduce channel conductance in the presence of menthol. The TRPM8[M2S4] chimera, oppositely, increases channel conductance, implying that the S4 helix in native TRPM8 may suppress menthol modulation. Overall, these findings show that there is promise in the techniques chosen to identify specific regions of TRPM8 crucial to menthol activation, though the methods chosen to study the TRPM8 pore independent from the whole channel may need to be reevaluated. Further experiments will be necessary to refine TRPM8 pore solubilization and purification before structural studies can proceed, and the electrophysiology traces observed for the chimeras will need to be further verified and evaluated for consistency and physiological significance.

Netflix has positioned itself at the forefront of the future of television with its original programming, which has been rolled out in greater and more frequent amounts just in the last couple of years. The streaming service has already experimented with creativity in ways most other shows and creators haven't, playing with the pacing of overall seasons as well as the length of episodes. So, too, Netflix has been at the forefront of increasing visibility for minority characters on television. Many of its shows incorporate racially diverse casts and depict lots of LGBTQ characters, a refreshingly realistic view of the world that many of its viewers have always lived in but haven't yet witnessed on television. Visibility and representation are critical concepts for analyzing minority characters on television. It is important for diverse characters to be seen, first and foremost, but also to be seen in positive or at least realistic lights. Care must be taken to avoid fulfilling stereotypes or tropes, and attention must be paid to what has happened to other characters who have come before. However, many of Netflix's portrayals of these characters, particularly bisexual characters, leave much to be desired. With the original dramas House of Cards, Hemlock Grove, Orange is the New Black, and Sense8, all of which include characters who identify as or behave bisexually, Netflix has been reluctant to use the specific word bisexual to describe characters, and many don't even identify their sexuality with a synonym for the term. Many of the bisexual characters that I identified died or were killed on the shows, and nearly all of them fulfilled stereotypes or tropes in some way. There were multiple scenes of threesomes or other distinctly kinky sexual encounters, which served to exoticize bisexuality and distance it from the more normatively viewed identities of heterosexuality and homosexuality. Ultimately, while Netflix's original programming has offered increased visibility to bisexual characters, it has yet to reflect the real community it seeks to portray. In particular, Netflix's refusal to label characters as bisexual is frustrating and limiting. It can be argued that this is a progressive move toward more ideas of sexual fluidity and a post-modern lack of sexual labels, but there are not enough depictions of identified bisexual characters on television yet for this to make sense. Until bisexual characters and their identities are not invisibilized or stigmatized, more work has to be done to ensure that bisexual people are represented fairly and accurately on television and in all media.

This is a constructed language made for fictional creatures that will be featured in a novel. The creatures are feral goblins, and they are based on Irish mythology. The novel that the goblins and the language will be featured in is titled MaddConn, and it was written by Jon Bendera. It is an urban fantasy novel. The goblins have a language in the novel because there are a number of goblin characters throughout the story. One of the main characters, Maddi, has to survive amongst the feral goblins for a portion of the novel while another main character, Connor, works his way to her. She has multiple close encounters with them, and also has to find her way around their territory, so she comes into contact with the language many times. Although the language in the story was created to have developed prior to humanity, this constructed language was made to look somewhat similar to human language. As it will be involved in fiction that will be read by humans, it needs to be easily understood by humans. The language reveals features of the goblin culture and anatomy. For instance, it can be seen in the language that their shallow, wide mouths and thin lips influence their phonology. Their physiology influences much of their language. Likewise, they have three fingers on each hand, which influences their number system. As the goblins and their language developed before humans and because they are not human creatures, the typology and word order do not follow all of the correlations found in human languages. All in all, the lexicon includes many words that have to do with three of their favorite things: violence, food, and work.

My passion on the importance of oral health began when I was four years old with the traumatic experience of witnessing my grandmother, my idol, with a toothless smile without her dentures in. At present, I have been a dental assistant for five years, and have heard of similar stories of people struggling with maintaining oral hygiene and having access to oral health care, including: family members, friends, patients, coworkers, and even Arizona State University faculty. Since the abolishment of emergency dental services in Arizona's Health Care Cost Containment System, dental related oral health care rates have jumped incrementally. In addition Arizona has created a dental desert, because dentists are not setting up their practices in the rural areas of the Valley, due to wanting to generate the most amount of income. Studies have also shown that dentists feel inept at treating ethnically diverse patients as well as patients from lower socioeconomic classes. These problems create a myriad of mental effects and physical ailments on the patient that are not limited to the oral cavity. In this paper, these issues in oral health care were studied along with their effects, including recommendations for resolutions.

Unauthorized confronts the relationship between technology and personhood in the modern world. More specifically, it addresses the personal and social effects of border politics within the frame of cyber crime. The short film takes place in the near future where a hacker can create citizenship for anyone she wants, effectively turning anyone into a legal person in the United States. This parallels the real life struggles of unauthorized immigrants trying to gain a new life this side of the border despite the overwhelming backlash from the conservative and xenophobic population. The main character's ability to grant citizenship forces the viewer to confront what being a person really means. The film also alludes to the popularized antics of modern day hackers and whistleblowers who are often turned into heroes for fighting the establishment despite their character flaws. The protagonist of Unauthorized struggles through underhanded sexism and blatant racism as well as her own personal struggles with drug addiction and failed relationships. These are very real struggles women face in technology jobs and life in general. The main character's actions ultimately destroy every relationship that she's established, including the connection to her own family. This film, in short, is about the walls people build between each other, both physical and social.

This creative project consists of three short stories with a common theme of release, letting go, and exhalation. Nymphal Instar is a story about Tommy, a young boy, and his encounter with his uncle, a troubled man who has just returned from war. The story explores the idea of growth and maturation, and the ability to move past and let go of trauma. A Cat Goes Away is about a young man, Richard, who is required to simultaneously deal with the loss of his cat and the suicide attempts of his sister. He also runs into his sister's ex-husband and is forced to deal with him. The story explores the difficulty in recognizing one's own emotions and the importance of knowing the difference between what one can change and what one cannot. Since Diagnosis is a story about Kate, a woman who has just been diagnosed with cancer and who is unable to tell her loved ones. The story explores acceptance and the idea that letting go can allow one to live more fully. Though the three stories are disparate in their characters and events, they share a commonality in their endings and in the final realizations of the characters. There is a focus on the importance of breath and breathing, and the essentiality of acceptance and release.

This is an analysis of broad racial discourse through a critical race theory: Responsibility Avoidance Discourse (RAD). RAD is coded English that communicates meaning through connotation, avoidance, and implication as a means of securing its main purposes: enforcing white supremacy, obscuring inequality, and hindering significant racial progress. RAD is extremely effective at directing discussion away from arguments that might induce self-reflexivity or question white privilege. It focuses on discrediting others as a means of legitimizing whiteness. I analyze examples of it from a variety of sources—from political discourse to media coverage and social media trends—to demonstrate its manifestations throughout society.

For my creative project, I began an art press that produces small-run vinyl records and artist's books. For my creative project, I began an art press that produces small-run vinyl records and artist's books. Initially, the venture began as a means to circumvent record pressing facilities as a vinyl record-cutting service. By the end of this project, the focus shifted to encompass more visual art products than just vinyl records. The project began with vinyl records because I saw a need in the market; in the past decade, the industry has grown dramatically, but the dozen record pressing plants in the country cannot keep up with the demand. Because record pressing companies prioritize large orders, it is difficult for many small bands and independent record labels to produce work on this medium. This is due to the long lead times, high prices, and large minimum order sizes. I located a man in Germany, who invented a machine that makes high-quality, lathe-cut records. I named the project Blushing Soup, as homage to my father, who passed during my first semester of college. It is through his passing that I was able to secure funds to pursue this venture. I brought on a partner, who was more familiar with art and audio recording than myself. In the summer of 2015, we met with this inventor to learn how to use his machine. By October of 2015, a machine of our own had arrived. In early November, Blushing Soup won a grant from the Scottsdale Museum of Contemporary Art. During this time, we released two vinyl records for local bands. For a culminating project, I coordinated a Record Store Day compilation album consisting of six bands featuring. After securing all of the music, the machine started having problems, which forced me to cancel this release. Recognizing the delicacy of the machine, prompted a shift in the aim of Blushing Soup. During this process, I started learning printmaking processes, and I realized that Blushing Soup could function as more than a record cutting service; we could be an art press. In the last few month of this project, I started making artist's books. By the end of April 2016, Blushing Soup will have released vinyl records for two bands, as well as produced four handmade books. This creative project centered around the process of creating art through lathe cutting and printmaking; the objective was not to maximize profits but rather refocus the consumption of art (in a sustainable practice).

Over the course of my undergraduate experience, I have grown significantly as an artist - developing an assortment of strengths in a variety of cinematic disciplines ranging from screenwriting and producing to post-production and cinematography. All the while, I have been giving back to the Sun Devil community by serving

Over the course of my undergraduate experience, I have grown significantly as an artist - developing an assortment of strengths in a variety of cinematic disciplines ranging from screenwriting and producing to post-production and cinematography. All the while, I have been giving back to the Sun Devil community by serving in a number of leadership positions around campus which exposed me to a plethora of communities differ from my own. The combination of these experiences allows me to continuously explore new passions in synergy with my art. Two of these standing as the concept of live performance and the work of William Shakespeare. Through this exploration of artistic synergy, I have experimented with integrating the works of the Bard of Avon into the realm of cinema. From the beginning of 2105, I have been drafting a feature-length screenplay which serves as a quasi-prequel to Shakespeare's The Tempest. Under the title of A Kingdom or a Cure, it tells the story of the revolutionary war-hero Miguel Prosperiti as he struggles to save his daughter form a mysterious disease which has baffled the best medical minds while the country he has rebuilt comes crumbling down in post-apocalyptic Italy. Deposed and left to die at the hands of his brother, Miguel must defend his child from the evil witch Sycorax who attempts to kill the pair in order to feed off of their suffering and prolong her own life. Serving to fill the requirements for the Film and Media Production Capstone, A Kingdom or a Cure reimagines the world of Shakespeare's play and creates a new context for the words and actions of its leading characters. Such stands as the foundation of what I have created for what I have created as my applied project - a stylistic re-imagining of William Shakespeare's The Tempest which draws from multiple interpretations of the narrative to be performed as a piece within a larger theatrical presentation staged with only the classical techniques which stand contemporary to the Bard of Avon. The remainder of this document shall lie in six primary sections. The first two establish the project and detail its evolution over the course of the thesis process. Next stands as the production log which chronicles my journey over the Classical and Poetic Drama course as well as the rehearsal process for Mythfest and the Chaucer Festival. Fourth shall consist of a bibliography of all the texts which I have worked with over the course of this thesis experience. Fifth rests A Kingdom or a Cure - the screenplay which inspired me to embark on the grand journey which this thesis has taken me. Sixth shall assume the form of the PowerPoint Presentation which I presented at my thesis defense which contains a collection of images which have provided me with artistic inspiration throughout the thesis process. In conjunction with one another, these pieces serve as the written elements of my applied project.

A theatrical prompt book is a compilation of all blocking notes, meeting reports, designs, show cues, and any other relevant information necessary to ensure that the production runs smoothly.
