Described is a study investigating the feasibility and predictive value of the Teacher Feedback Coding System, a novel observational measure of teachers’ feedback provided to students in third grade classrooms. This measure assessed individual feedback events across three domains: feedback type, level of specificity and affect of the teacher. Exploratory and confirmatory factor analysis revealed five factors indicating separate types of feedback: positive and negative academic-informative feedback, positive and negative behavioral-informative feedback, and an overall factor representing supportive feedback. Multilevel models revealed direct relations between teachers’ negative academic-informative feedback and students’ spring math achievement, as well as between teachers’ negative behavioral-informative feedback and students’ behavior patterns. Additionally, a fall math-by-feedback interaction was detected in the case of teachers’ positive academic-informative feedback; students who began the year struggling in math benefitted from more of this type of feedback. Finally, teachers’ feedback was investigated as a potential mediator in a previously established relation between teachers’ self-reported depressive symptoms and the observed quality of the classroom environment. Partial mediation was detected in the case of teachers’ positive academic-informative feedback, such that this type of feedback was accountable for a portion of the variance observed in the relation between teachers’ depressive symptoms and the quality of the classroom environment.

Prior research has established associations between sleep duration and body mass index (BMI) scores and risk for obesity in middle childhood, but it is less clear whether other objectively- and subjectively-measured sleep indicators may be associated with BMI scores, weight status (e.g., obesity), and other estimates of weight and body fat such as waist circumference (WC) and percent body fat. Empirical studies have also demonstrated independent associations between broad self-regulation and sleep indicators and BMI scores, but no study to date has tested these factors in a model together and the extent to which associations between normative sleep problems, weight indicators, and effortful control (EC) may be explained by shared genetic or environmental influences. Data from a large longitudinal study of twins was used to test phenotypic associations between sleep problems at eight years and weight indicators at nine years, including whether EC at eight years moderates these associations. Additionally, multiple quantitative behavior genetic models were used to estimate unique and shared genetic and environmental covariances among normative sleep problems, weight indicators, and EC at eight years of age and whether additive genetic influence on weight in middle childhood differs by child weight status group. Phenotypic findings showed that greater sleep duration at eight years predicted greater decreases BMI at nine years of age for children with low levels of EC at eight years. Greater sleep midpoint variability at eight years predicted greater increases in percent body fat from eight to nine years of age for children with low EC at eight years. Behavior genetic findings showed greater environmental influences on parent-reported sleep duration and quality, as well as objective sleep midpoint variability. Similarly, associations between parent-reported sleep duration and sleep midpoint variability and other sleep indicators and EC were primarily accounted for by shared environmental factors. In contrast, there was high additive genetic influence on objective sleep quantity and quality, all weight indicators, and EC. Many of the associations between sleep indicators, sleep and weight indicators, and among weight indicators were entirely accounted for by shared additive genetic factors, suggesting that common, underlying sets of genes explain these relations.

The following study was developed to investigate the development of writing skills in second and third grade students. The recent emphasis on writing, specifically writing in multiple genres, made in the Common Core State Standards (CCSS, 2010) has increased the need to further understand how students write. The NAEP (2002) reports that approximately 77% of fourth grade students have only a general grasp of writing. Despite this poor performance, the Common Core State Standards (CCSS, 2010) have increased the expectations for student writing. The goal of this proposed dissertation, using an holistic literacy perspective, is to shed light on differences in how students write in informative and opinion genres, which skills predict writing outcomes, the extent to which reciprocal effects between writing and literacy are present, and what type of student profiles exist within the classroom. It was found that students received lower scores on opinion writing compared to their informative compositions. It was also found that better reading comprehension was associated with better writing performance in both genres. High vocabulary ability predicted higher opinion essay scores and better performance on a behavioral regulation task predicted better informative essay outcomes. Reciprocal effects between writing outcomes and literacy skill were found, with higher opinion writing scores predicted higher vocabulary outcomes. Finally, students appeared to fall into four latent profiles: high achievers, average achievers, struggling students, and a group of students who have average literacy skill but scored extremely poorly on the opinion essay task.

Clinically meaningful emotional and behavioral problems are thought to be present beginning in infancy, and may be reliably assessed in children as young as 12 months old. However, few studies have investigated early correlates of emotional and behavioral problems assessed in infancy. The current study investigates the direct and interactive contributions of early infant and caregiver characteristics thought to play an important role in the ontogeny of behavior problems. Specifically, the study examines: (1) the links between temperamental reactivity across the first year of life and behavior problems at 18 months, (2) whether children high in temperamental reactivity are differentially susceptible to variations in maternal sensitivity, (3) the extent to which child temperamental risk or susceptibility may further be explained by mothers’ experiences of stressful life events (SLEs) during and before pregnancy. Data were collected from 322 Mexican American families during prenatal, 6-, 12-, 18-, and 24-week home interviews, as well as during 12- and 18-month lab interviews. Mother reports of SLEs were obtained between 23-40 weeks gestation; temperamental negativity and surgency at 6 weeks and 12 months; and internalizing and externalizing behaviors at 18 months. Maternal sensitivity during structured mother-infant interaction tasks at the 6-, 12-, 18-, and 24-week visits was assessed by objective observer ratings. Study findings indicated that maternal SLEs before birth were associated with more infant negativity across the first year of life, and that negativity in turn was associated with more internalizing problems at 18 months. Ecological stressors thought to be associated with sociodemographic risk factors such as low-income and ethnic minority status may begin to exert cascades of influence on children’s developmental outcomes even before birth.

This dissertation examined how anxiety levels and social competence change across the course of early elementary school, as well as how individual differences at the transition to kindergarten may influence these trajectories. Previous research has supported unidirectional relations among anxiety and social competence, but few studies explore how inter- and intra-individual changes in social competence and anxiety may be related across time. From a developmental perspective, studying these trajectories following the transition to kindergarten is important, as cognitive and emotion regulation capacities increase markedly across kindergarten, and the relative success with which children navigate this transition can have a bearing on future social and emotional functioning across elementary school. In addition, given gender differences in anxiety manifestation and social competence development broadly, gender differences were also examined in an exploratory manner. Data from parent and teacher reports of a community sample of 291 children across kindergarten, 1st, and 2nd grades were analyzed. Results from bivariate growth models revealed steeper increases in anxiety, relative to peers in the sample, were associated with steeper decreases in social competence across time. This finding held after controlling for externalizing behavior problems at each time point, which suggests that relations among anxiety and social competence may be independent of other behavior problems commonly associated with poor social adjustment. Temperament variables were associated with changes in social competence, such that purportedly "risky" temperament traits of higher negative emotionality and lower attention control were associated with concurrently lower social competence in kindergarten, but with relatively steeper increases in social competence across time. Temperament variables in kindergarten were unrelated with changes in anxiety across time. Gender differences in relations among anxiety in kindergarten and growth in social competence also were revealed. Findings for teacher and parent reports of child behavior varied. Results are discussed with respect to contexts that may drive differences between parent and teacher reports of child behavior, as well as key developmental considerations that may help to explain why kindergarten temperament variables examined herein appear to predict changes in social competence but not changes in anxiety levels.

This study examined whether social support available to parents moderated the heritability of parent-reported social approach at 12 months (N = 286 twin pairs, 52.00% female) and social competence at 30 months (N = 259 twin pairs, 53.30% female). Genetic and environmental covariance across age is also reported. Social support consistently moderated genetic influences on children’s social approach and competence, such that heritability was highest when parents reported low social support. Shared environment was not moderated by social support and explained continuity across age. Findings provide further evidence that genetic and environmental influences on development vary across context. When parents are supported, environmental influences on children’s social competence are larger, perhaps because support helps parents provide a broadly promotive environment.

Reducing the amount of error and introduced data variability increases the accuracy of Western blot results. In this study, different methods of normalization for loading differences and data alignment were explored with respect to their impact on Western blot results. GAPDH was compared to the LI-COR Revert total protein stain as a loading control. The impact of normalizing data to a control condition, which is commonly done to align Western blot data distributed over several immunoblots, was also investigated. Specifically, this study addressed whether normalization to a small subset of distinct controls on each immunoblot increases pooled data variability compared to a larger set of controls. Protein expression data for NOX-2 and SOD-2 from a study investigating the protective role of the bradykinin type 1 receptor in angiotensin-II induced left ventricle remodeling were used to address these questions but are also discussed in the context of the original study. The comparison of GAPDH and Revert total protein stain as a loading control was done by assessing their correlation and comparing how they affected protein expression results. Additionally, the impact of treatment on GAPDH was investigated. To assess how normalization to different combinations of controls influences data variability, protein data were normalized to the average of 5 controls, the average of 2 controls, or an average vehicle and the results by treatment were compared. The results of this study demonstrated that GAPDH expression is not affected by angiotensin-II or bradykinin type 1 receptor antagonist R-954 and is a less sensitive loading control compared to Revert total protein stain. Normalization to the average of 5 controls tended to reduce pooled data variability compared to 2 controls. Lastly, the results of this study provided preliminary evidence that R-954 does not alter the expression of NOX-2 or SOD-2 to an expression profile that would be expected to explain the protection it confers against Ang-II induced left ventricle remodeling.

Premature babies are at risk of death from immature lung development. For this reason, pregnant mothers at risk for preterm delivery are administered dexamethasone (DEX), a synthetic glucocorticoid that promotes fetal lung development. However, exposure to DEX in utero is associated with low birth weight and cardiovascular development pathologies. Moreover, our lab found that DEX administration in-utero leads to a sex-specific increase in stress-induced tachycardia in female, but not male offspring. This project seeks to expand on this preliminary finding of the heart by examining local effectors of activity from the sympathetic system (tyrosine hydroxylase and catechol-o-methyltransferase). Tyrosine hydroxylase was measured as it catalyzes the rate limiting step of norepinephrine synthesis while catechol-O- methyltransferase was studied as it catalyzes the degradation of norepinephrine. Acetylcholinesterase was used to measure parasympathetic activity as it catalyzes the degradation of the primary neurotransmitter of the parasympathetic nervous system, acetylcholine. Analyses of sympathetic as well as parasympathetic activity were done to determine influences of in-utero DEX exposure on autonomic regulation in adulthood. Pregnant rats were administered DEX (0.4 mg/kg, i.p.) or vehicle (20% w/v 2-hydroxypropyl ß- cyclodextran) at gestation days 18-21, with euthanasia of offspring occurring at around the time the offspring reached 13-15 weeks of age. Left ventricles and right atria were pulverized, processed and subjected to western blot analysis to determine expression of proteins of interest. Males exposed to DEX in-utero saw a decrease in tyrosine hydroxylase expression in left ventricle and right atrium when compared to vehicle control, a difference not seen with females. In addition, catechol-o-methyltransferase expression was increased in right atria from male, but not female rats. Acetylcholinesterase expression was reduced in the right atria of female, but not male rats. The present findings suggest reduced norepinephrine signaling in the heart of male, but not female DEX-exposed offspring. Given that we have previously found that female, but not male rats exhibit exaggerated stress-induced tachycardia, our current findings suggest that males possess a sex-specific compensatory mechanism allowing the heart to resist increased sympathetic signaling from the brain, one that females do not possess. The underlying mechanics of this proposed mechanism are unclear, and further investigation is needed in this subject to determine the significance of the findings from our study.

Guided by the Risky Families model and Daily Process methods, the present study examined how daily stressors are related to emotional well-being at the between- and within-person levels among adolescent grandchildren raised by grandmothers. This study also examined whether risk (i.e., adverse childhood experiences/ACES) and resilience (i.e., socio-emotional skills) factors were linked to differences in daily well-being, stressor exposure, and emotional reactivity, and evaluated the efficacy of an online social intelligence training (SIT) program on daily stressor-emotion dynamics. Data came from a subsample (n = 188) of custodial adolescents who participated in an attention-controlled randomized clinical trial and completed 14-day daily surveys prior to and following intervention. Analyses were conducted with dynamic structural equation modeling. Daily stressors, on average, and experiencing above average stressors, were associated with higher negative emotions and lower positive emotions and social connection. Those with more ACEs, on average, reported higher daily stressors and worse well-being, whereas those with higher socio-emotional skills, on average, reported lower daily stressors and better well-being. At the within-person level, more ACEs were associated with higher daily negative emotions. Nonverbal processing was linked to higher daily positive emotions and social connection. Conversational skills were associated with higher daily positive emotions and social connection, and lower, more inert daily negative emotions. Neither ACEs nor socio-emotional skills were associated with within-person reactivity to stressors. Also, the SIT program did not demonstrate efficacy for any outcome. My discussion focused on how findings extend the literature on custodial adolescents by showing that daily stressors impact well-being, offer knowledge of how ACEs and socio-emotional skills shape daily stressor-emotion dynamics, and considers reasons why the online, self-guided SIT program failed to show efficacy on key outcomes.

Elevated rates of exposure to multi-level chronic stressors (e.g., poverty, discrimination, acculturative stress) place low-income, Mexican-origin individuals in the United States at elevated risk for adverse psychological and physical health across the lifespan. Despite exposure to contextual risk factors, many individuals maintain positive biobehavioral health. In particular, despite greater exposure to sociodemographic risk factors, more recently immigrated Mexican-origin individuals in the U.S. may demonstrate more positive biobehavioral health, warranting consideration of specific cultural values and practices that confer and maintain positive health across generations. Parental cultural socialization is an understudied mechanism in promotive pathways of parent-child processes and child biobehavioral health. Across three generations of Mexican-origin families in the United States – maternal grandmothers, mothers, children – the current study (1) identified a multidimensional measure of child biobehavioral health across psychological and biological indicators, (2) evaluated the intergenerational transmission of grandmother-mother cultural socialization, (3) evaluated the effect of maternal cultural socialization on child-perceived parenting and child biobehavioral health, and (4) evaluated child cultural orientation as a moderator of the effect of maternal cultural socialization on child-perceived parenting and child biobehavioral health. Findings highlight the complex and nuanced relations among parental cultural socialization, individual cultural orientation, child perceptions of parenting, and child biobehavioral health among low-income, Mexican-origin families in the United States.
