This thesis critically examines the scientific and ethical dimensions of the novel and controversial technology of mitochondrial replacement therapy (MRT). The first portion provides a background on mitochondrial diseases and examines how these organelles (mitochondria) have left humans vulnerable to disease. The second portion of the thesis examines the technology of mitochondrial replacement therapy and what research contributed to the development of MRT. Finally, the third section responds to the ethical and legal concerns proposed and outlined by the Nuffield Council of Bioethics. After careful consideration of all legal and ethical concerns, it is concluded that the technology of MRT should be pursued under careful foresight with the idea that the technology is already happening and we must move forward in a way that provides the greatest information.

Binaural beat therapy is one acoustics technique used in music; a technique utilized in the music most people listen to when they have trouble sleeping or concentrating. The binaural beats technique, along with other therapeutic methods in music, have always fascinated me. This topic is something that I’ve wanted to research for some time. So for this creative project I want to challenge myself. I am going to learn how to produce music intended for musical therapy. I’ll be implementing methods based on my research. I hope to produce a piece of music that can help others. My goal is to create a song that has the potential to be added to the vast online library of therapeutic music.

While growing up, I was placed into dance classes, dance started out as a hobby, but as I grew up it became a way for me to escape from the struggles life itself brings. While I was taking a dance culture class at Arizona State University, I stumbled across research that revealed that dance does not just help people like myself, but it also has the ability to help those with more difficult life-altering situations like Parkinson’s Disease. With having about 970 million adults aged 65 years old and up (United Nations), around 10-million of these individuals have Parkinson’s Disease (PD) (Parkinson’s New Today). With these large numbers, Parkinson’s is the second leading neurodegenerative disease worldwide (Parkinson’s News Today) behind Alzheimer’s. Parkinson’s is a motor system disorder that affects the production of dopamine in one’s brain (Harvard). With the current treatment of PD being medication as well as surgical therapy based on the severity of each patient (Parkinson’s Foundation), there is one form of treatment that has been tested but not certified, partnership dancing. The way that partnership dance benefits those with Parkinson’s Disease is by using many areas of the brain to facilitate dopamine production. The four main areas used are the motor cortex, the somatosensory, the basal ganglia, and lastly the cerebellum (Harvard). With the vast amount of existing research, as well as the information gained through secondary research, I feel as though there needs to be a study to open the development of partnership dance as a therapy modality for those with many of the forms of degenerative mental diseases. Although unable to put on this research, I have outlined what this study could look like to be continued in the hopes of having partnership dance become a certified form of therapy for those with Parkinson’s Disease.

The fashion industry dubs couture as high fashion, yet couture never reaches the finish line when it comes to comfort. Most of the brand name high heels on the market are too painful to wear for long periods of time. For this project, I have developed 3D printed high heels with detachable insoles that will relieve tired feet based on the principle of reflexology. The product integrates traditional flexible insoles with Arduino computing and the result is a functional surface that can ease the pain of the wearer. This paper introduces the product and with it, under-explored opportunities to customize your own high heels at home. Essentially, each consumer will have the ability to personalize and switch out their style without sacrificing comfort. Soon, a consumer will be a designer.

Development of a Blood Brain Barrier Permeability Database for Investigational Anti-Cancer Agents for Use in Glioblastoma Research

Glioblastoma (GBM) is an extremely malignant form of brain cancer characterized by rapid progression and poor patient survival. Even after standard of care treatments, less than ten percent of patients with this disease survive five years. More effective therapeutic options are urgently needed to improve outcomes for patients with GBM. Adequate drug delivery is a critical challenge in GBM treatment, as drugs delivered systemically must be able to penetrate the blood brain barrier (BBB) and reach the tumor at therapeutic levels. To address this, we developed a resource to catalog BBB penetration information for investigational agents currently in clinical trials in cancer. Using an in silico prediction model and manual annotation to capture existing knowledge from the published literature, BBB content for ~500 investigational drugs was added to the investigational database tool. In addition to BBB content, the database also includes information on the gene targets of the included therapies. The investigational database tool was used to identify investigational agents that (1) may have increased activity against GBM based on the presence of a specific mutation in the tumor sample and (2) have evidence suggesting the compounds may penetrate the BBB. By prioritizing investigational agents for further study based on evidence for BBB penetration, this resource can help the GBM research community pursue more effective treatments for GBM.

Ultrasound Imaging of Swallowing Subsequent to Feeding and Myofunctional Intervention

The purpose of this study was to examine swallowing patterns using ultrasound technology subsequent to the implementation of two therapeutic interventions. Baseline swallow patterns were compared to swallows after implementation of therapeutic interventions common in both feeding therapy (FT) and orofacial myofunctional therapy (OMT). The interventions consist of stimulation of the tongue by z-vibe and tongue pops. Changes in swallowing patterns are described, and similarities of interventions across the two professions are discussed. Ultrasound research in the realm of swallowing is sparse despite having potential clinical application in both professions. In using ultrasound, this study outlines a protocol for utilization of a hand-held probe and reinforces a particular protocol described in the literature. Real-time ultrasound recordings of swallows for 19 adult female subjects were made. Participants with orofacial myofunctional disorder are compared to a group with typical swallowing and differences in swallowing patterns are described. Three stages of the oral phase of the swallow were assigned based on ultrasonic observation of the tongue shape. Analysis involves total duration of the swallow, duration of the three stages in relation to the total duration of the swallow, and the number of swallows required for the bolus to be cleared from the oral cavity. No significant effects of either intervention were found. Swallowing patterns showed a general trend to become faster in total duration subsequent to each intervention. An unexpected finding showed significant changes in the relationship between the bolus preparation stage and the bolus transportation stage when comparing the group classified as having a single swallow and the group classified as having multiple swallows.

The purpose of this project was to research the expression of Autism Spectrum Disorder in children and create a children’s book that can help the peers of individuals with a diagnosis to understand what the disorder entails and potentially gain a new sense of empathy for peers of all levels of physical and mental abilities. The research component includes interviews with individuals deemed knowledgeable about ASD, including occupational therapists, behavioral analysts, and parents, as well as a literature review of research studies on the expression of Autism in children. This written portion of the project may also serve as a manual for individuals who have little to no knowledge of ASD, as it dives deeper into the content of the book and research, while remaining easily understandable and clear to those without any prior knowledge or experience with ASD. It could prove especially useful for those in professions that come into contact with individuals with Autism, but do not necessarily require psychology courses or training as a prerequisite for the role, such as teachers and some health professionals.

Robotic rehabilitation for upper limb post-stroke recovery is a developing technology. However, there are major issues in the implementation of this type of rehabilitation, issues which decrease efficacy. Two of the major solutions currently being explored to the upper limb post-stroke rehabilitation problem are the use of socially assistive rehabilitative robots, robots which directly interact with patients, and the use of exoskeleton-based systems of rehabilitation. While there is great promise in both of these techniques, they currently lack sufficient efficacy to objectively justify their costs. The overall efficacy to both of these techniques is about the same as conventional therapy, yet each has higher overhead costs that conventional therapy does. However there are associated long-term cost savings in each case, meaning that the actual current viability of either of these techniques is somewhat nebulous. In both cases, the problems which decrease technique viability are largely related to joint action, the interaction between robot and human in completing specific tasks, and issues in robot adaptability that make joint action difficult. As such, the largest part of current research into rehabilitative robotics aims to make robots behave in more "human-like" manners or to bypass the joint action problem entirely.

Although music therapy has grown in recent years as a treatment method for a variety of mental health and wellness problems, it has yet to be comprehensively applied to the specific issue of test anxiety. Some studies have examined the use of music in testing situations in order to reduce anxiety or improve academic performance. However, more in-depth music therapy interventions are a promising, largely untried treatment possibility for students suffering from this type of anxiety.

Human genetic engineering, the manipulation of genomic DNA, is an up and coming field in which soon we will no longer be asking if we can do it, but should we do it? It raises ethical questions regarding autonomy, the concept that an individual is capable of self-governance and making informed decisions, as well as consent of those who undergo genetic manipulation. Along with the ethical questions come issues of equality, equal opportunity of usage, and responsibility of those who use such technologies. The benefits and consequences of genetically engineering humans must be examined, such as the eradication of genetic diseases and the ability to alter ourselves as we desire, as well as the possible unintended outcomes, such as a two-class society and insurmountable inequality. Eugenics and the Transhumanism movement will also be examined, and how they could affect genetic engineering. Regulations will be needed, to limit parents on how they can modify their children and on how adults can modify themselves, to ensure the safe future of genetic engineering.
