The depiction of mental illness, schizophrenia in particular, within film is a unique phenomenon that film directors have decided to undertake more so in the last 20 years than ever before in cinematic history (Wedding & Niemic, 2014; Robinson, 2004; Gabbard & Gabbard, 1999; Wahl, 1997). Countless filmmakers have taken on the challenge of depicting this complex, yet degenerative condition that entails auditory and visual hallucinations, disorganized thought and speech, and delusions. Its portrayals are usually exaggerated and romanticized, and convey a sense of separate "Otherness" with those who have a mental disorder. And while filmmakers try to encapsulate the schizophrenic experience, it is not without psychiatric error and regarding the person who has schizophrenia as a spectacle. This unfair and ostracizing view of people who have schizophrenia is fueled by films like A Beautiful Mind and The Shining where the film either creates impossibly high standards for schizophrenics to perform at, or the film paints the character as a violent savage. In either case, the end result is the marking and, usually, denouncement of the schizophrenic for their illness. What filmmakers tend to overlook is how much the public learns from the cinematic portrayals of these disorders, and that their films are contributing to an overarching issue of public presumptions of actual schizophrenia and how it is perceived. While the Hollywood approach offers a depiction that is usually more tangible and enjoyable for masses of audiences, spectators should recognize that these are artistic interpretations that take liberties in their depictions of schizophrenia. Viewing these films with an objective mindset to better understand the inner workings of schizophrenia is absolutely crucial in arriving anything close to the truth behind this mental illness that has been demonized long enough.

Vulnerability research is a fairly new field of study and has yet to be applied to fields such as improvisation or online content creation. Making vulnerability public in a way that necessitates improvisation is fundamental to YouTube content creation. This Creative Project focuses on drawing connections between Vulnerability and Improvisation as it relates to creating content for YouTube. I did this through creating my own content which consisted of four YouTube videos centering around the theme of embracing fear. I found that in order to create content, I had to practice vulnerability myself and embrace improvisation if I wished to communicate those same themes to my audience. This project revealed that there are many ways which this powerful vulnerability research is yet to be applied, specifically in the realm of vastly public arenas such as YouTube. This research shows that vulnerability is not only applicable to interpersonal relationships, but has the potential to influenced thousands when used in public spheres.

My name is Adriana Becerra and I am a student at the Walter Cronkite School of Journalism and Mass Communication at Arizona State University. In hoping to combine my two passions of journalism and film, for my Honors Undergraduate Thesis project I created my own film review website. My website is a complete review of the films that were nominated for the 2015 Oscars in the following categories: Best Picture, Animated Feature, Documentary Feature, Foreign Language, and Short Film Live Action. In all, I watched and reviewed a total of twenty-eight films based on acting, lighting, music, cinematography, costume/makeup/set design, writing, and visual effects. Over the course of nine months, I have watched, reviewed, and talked extensively about each film that I have reviewed. Though tedious at times, I thoroughly enjoyed completing my Undergraduate Thesis Project. I hope to continue critically looking at films, and possibly even incorporating film in my journalistic career.

Duchenne Muscular Dystrophy (DMD) is an X-linked recessive disease characterized by progressive muscle loss and weakness. This disease arises from a mutation that occurs on a gene that encodes for dystrophin, which results in observable muscle death and inflammation; however, the genetic changes that result from dystrophin's dysfunctionality remain unknown.

Duchenne Muscular Dystrophy (DMD) is an X-linked recessive disease characterized by progressive muscle loss and weakness. This disease arises from a mutation that occurs on a gene that encodes for dystrophin, which results in observable muscle death and inflammation; however, the genetic changes that result from dystrophin's dysfunctionality remain unknown. Current DMD research uses mdx mice as a model, and while very useful, does not allow the study of cell-autonomous transcriptome changes during the progression of DMD due to the strong inflammatory response, perhaps hiding important therapeutic targets. C. elegans, which has a very weak inflammatory response compared to mdx mice and humans, has been used in the past to study DMD with some success. The worm ortholog of the dystrophin gene has been identified as dys-1 since its mutation phenocopies the progression of the disease and a portion of the human dystrophin gene alleviates symptoms. Importantly, the extracted RNA transcriptome from dys-1 worms showed significant change in gene expression, which needs to be further investigated with the development of a more robust model. Our lab previously published a method to isolate high-quality muscle-specific RNA from worms, which could be used to study such changes at higher resolution. We crossed the dys-1 worms with our muscle-specific strain and demonstrated that the chimeric strain exhibits similar behavioral symptoms as DMD patients as characterized by a shortened lifespan, difficulty in movement, and a decrease in speed. The presence of dys-1 and other members of the dystrophin complex in the body muscle were supported by the development of a resulting phenotype due to RNAi knockdown of each component in the body muscle; however, further experimentation is needed to reinforce this conclusion. Thus, the constructed chimeric C. elegans strain possesses unique characteristics that will allow the study of genetic changes, such as transcriptome rearrangements and dysregulation of miRNA, and how they affect the progression of DMD.

The Walt Disney Company has been a worldwide phenomenon for over half a century. Disney's animated films in particular impact a large number of individuals around the world. The fact that they rerelease popular films every few years lends to the lasting influence these movies will hold in the lives of children to come. It is important to examine the messages Disney animated films can teach children in regards to women's roles, United States history, and racial difference. This essay examines these topics as they appear in Disney's Snow White and the Seven Dwarves, The Little Mermaid, Pocahontas, and The Lion King. Lastly, it examines the potential impact these films can leave on children and suggests ways in which adults can help children analyze what they see in the media.

This thesis aims to analyze and explain the resurgence of the superhero genre, particularly in recent cinema, directly following the terrorist attacks on September 11, 2001. It will also deconstruct the current American political landscape and define how popular culture has historically reflected real-world issues. The study draws heavily on the political ideology of neoliberalism and Henry Jenkins' media theory of convergence culture. I ultimately argue in the course of the analysis that viewers of these superhero films, regardless of their interest in comic books, cathartically release their fears and post-9/11 anxiety through cinematic escapism. It will also relay the evolution of the superhero in the last seventy years as a way to show the effects current events have on popular culture and history, using Captain America and Iron Man as examples of shifting American values.

It is important to consider factors that contribute to successful fertilization and the development of viable offspring. Better understanding the factors that contribute to infertility can be used to assist in the development of viable offspring, especially for human beings looking to successfully reproduce. Identifying paternal effect genes, genes that come from the father, introduces more targets that can be manipulated to produce specific reproductive effects. Use of Drosophila melanogaster as a model to study reproduction has increased, in part, due to the use of the GAL4 system. In this system, the GAL4 gene encodes an 881 amino acid protein that binds to the 4-site Upstream Activating Sequence (UAS) to induce transcription of the gene of interest. These sequences constitute the two components of the system: the driver (GAL4) and the responder (gene of interest) \u2014 each of which is maintained as a separate parental line. Effects of the GAL4 driver line "driving" transcription of the responder can be assessed by examining the offspring. One of the more common uses of the GAL4 system involves analyzing phenotypic effects of reducing or eliminating expression of a target gene through the induction of RNAi transcription, which often results in toxicity, lethality, or reduced viability. Utilizing these principles, we strove to demonstrate the effect of knocking down the expression of testis-specific sperm-leucyl-aminopeptidases gene CG13340 on progeny by inducing expression of RNAi with two distinct GAL4 driver lines - one with a nonspecific actin-binding activation sequence and the other with a testis-specific activation sequence. Comparison of both GAL4 driver lines to crosses using N01 wild type ("wt") flies verify that inducing RNAi transcription using the GAL4 system results in reduction of proper offspring development. Further studies using D. melanogaster and the GAL4 system can improve knowledge of factors contributing to male fertility and also be applied to better understand mammalian, specifically human, fertility.

Alternative polyadenylation (APA) is the biological mechanism in which the same gene can have multiple 3'untranslated region (3'UTR) isoforms due to the presence of multiple polyadenylation signal (PAS) elements within the pre mRNAs. Because APA produces mRNA transcripts that have different 3'UTR isoforms, certain transcripts may be subject to post-transcriptional regulation by regulatory non-coding RNAs, such as microRNAs or RNA binding proteins defects of which have been implicated in diseases such as cancer. Despite the increasing level of information, functional understanding of the molecular mechanisms involved in transcription is still poorly understood, nor is it clear why APA is necessary at a cell or tissue-specific level. To address these questions I wanted to develop a set of sensor strain plasmids capable of detecting cleavage and polyadenylation in vivo, inject the complete sensor strain plasmid into C. elegans and prepare stable transgenic lines, and perform proof-of-principle RNAi feeding experiments targeting genes associated with the cleavage and polyadenylation complex machinery. I demonstrated that it was possible to create a plasmid capable of detecting cleavage and polyadenylation in C. elegans; however, issues arose during the RNAi assays indicating the sensor strain plasmid was not sensitive enough to the RNAi to effectively detect in the worms. Once the problems involved with sensitivity and variability in the RNAi effects are resolved, the plasmid would be able to better address questions regarding the functional understanding of molecular mechanisms involved in transcription termination.
