This thesis investigated two different thermal flow sensors for intravascular shear stress analysis. They were based on heat transfer principle, which heat convection from the resistively heated element to the flowing fluid was measured as a function of the changes in voltage. For both sensors, the resistively heated elements were made of Ti/Pt strips with the thickness 0.12 µm and 0.02 µm. The resistance of the sensing element was measured at approximately 1.6-1.7 kohms;. A linear relation between the resistance and temperature was established over the temperature ranging from 22 degree Celsius to 80 degree Celsius and the temperature coefficient of resistance (TCR) was at approximately 0.12 %/degree Celsius. The first thermal flow sensor was one-dimensional (1-D) flexible shear stress sensor. The structure was sensing element sandwiched by a biocompatible polymer "poly-para-xylylene", also known as Parylene, which provided both insulation of electrodes and flexibility of the sensors. A constant-temperature (CT) circuit was designed as the read out circuit based on 0.6 µm CMOS (Complementary metal-oxide-semiconductor) process. The 1-D shear stress sensor suffered from a large measurement error. Because when the sensor was inserted into blood vessels, it was impossible to mount the sensor to the wall as calibrated in micro fluidic channels. According to the previous simulation work, the shear stress was varying and the sensor itself changed the shear stress distribution. We proposed a three-dimensional (3-D) thermal flow sensor, with three-axis of sensing elements integrated in one sensor. It was in the similar shape as a hexagonal prism with diagonal of 1000 µm. On the top of the sensor, there were five bond pads for external wires over 500 µm thick silicon substrate. In each nonadjacent side surface, there was a bended parylene branch with one sensing element. Based on the unique 3-D structure, the sensor was able to obtain data along three axes. With computational fluid dynamics (CFD) model, it is possible to locate the sensor in the blood vessels and give us a better understanding of shear stress distribution in the presence of time-varying component of blood flow and realize more accurate assessment of intravascular convective heat transfer.

Continuous monitoring in the adequate temporal and spatial scale is necessary for a better understanding of environmental variations. But field deployments of molecular biological analysis platforms in that scale are currently hindered because of issues with power, throughput and automation. Currently, such analysis is performed by the collection of large sample volumes from over a wide area and transporting them to laboratory testing facilities, which fail to provide any real-time information. This dissertation evaluates the systems currently utilized for in-situ field analyses and the issues hampering the successful deployment of such bioanalytial instruments for environmental applications. The design and development of high throughput, low power, and autonomous Polymerase Chain Reaction (PCR) instruments, amenable for portable field operations capable of providing quantitative results is presented here as part of this dissertation. A number of novel innovations have been reported here as part of this work in microfluidic design, PCR thermocycler design, optical design and systems integration. Emulsion microfluidics in conjunction with fluorinated oils and Teflon tubing have been used for the fluidic module that reduces cross-contamination eliminating the need for disposable components or constant cleaning. A cylindrical heater has been designed with the tubing wrapped around fixed temperature zones enabling continuous operation. Fluorescence excitation and detection have been achieved by using a light emitting diode (LED) as the excitation source and a photomultiplier tube (PMT) as the detector. Real-time quantitative PCR results were obtained by using multi-channel fluorescence excitation and detection using LED, optical fibers and a 64-channel multi-anode PMT for measuring continuous real-time fluorescence. The instrument was evaluated by comparing the results obtained with those obtained from a commercial instrument and found to be comparable. To further improve the design and enhance its field portability, this dissertation also presents a framework for the instrumentation necessary for a portable digital PCR platform to achieve higher throughputs with lower power. Both systems were designed such that it can easily couple with any upstream platform capable of providing nucleic acid for analysis using standard fluidic connections. Consequently, these instruments can be used not only in environmental applications, but portable diagnostics applications as well.

Over the past fifty years, the development of sensors for biological applications has increased dramatically. This rapid growth can be attributed in part to the reduction in feature size, which the electronics industry has pioneered over the same period. The decrease in feature size has led to the production of microscale sensors that are used for sensing applications, ranging from whole-body monitoring down to molecular sensing. Unfortunately, sensors are often developed without regard to how they will be integrated into biological systems. The complexities of integration are underappreciated. Integration involves more than simply making electrical connections. Interfacing microscale sensors with biological environments requires numerous considerations with respect to the creation of compatible packaging, the management of biological reagents, and the act of combining technologies with different dimensions and material properties. Recent advances in microfluidics, especially the proliferation of soft lithography manufacturing methods, have established the groundwork for creating systems that may solve many of the problems inherent to sensor-fluidic interaction. The adaptation of microelectronics manufacturing methods, such as Complementary Metal-Oxide-Semiconductor (CMOS) and Microelectromechanical Systems (MEMS) processes, allows the creation of a complete biological sensing system with integrated sensors and readout circuits. Combining these technologies is an obstacle to forming complete sensor systems. This dissertation presents new approaches for the design, fabrication, and integration of microscale sensors and microelectronics with microfluidics. The work addresses specific challenges, such as combining commercial manufacturing processes into biological systems and developing microscale sensors in these processes. This work is exemplified through a feedback-controlled microfluidic pH system to demonstrate the integration capabilities of microscale sensors for autonomous microenvironment control.

The design and development of analog/mixed-signal (AMS) integrated circuits (ICs) is becoming increasingly expensive, complex, and lengthy. Rapid prototyping and emulation of analog ICs will be significant in the design and testing of complex analog systems. A new approach, Programmable ANalog Device Array (PANDA) that maps any AMS design problem to a transistor-level programmable hardware, is proposed. This approach enables fast system level validation and a reduction in post-Silicon bugs, minimizing design risk and cost. The unique features of the approach include 1) transistor-level programmability that emulates each transistor behavior in an analog design, achieving very fine granularity of reconfiguration; 2) programmable switches that are treated as a design component during analog transistor emulating, and optimized with the reconfiguration matrix; 3) compensation of AC performance degradation through boosting the bias current. Based on these principles, a digitally controlled PANDA platform is designed at 45nm node that can map AMS modules across 22nm to 90nm technology nodes. A systematic emulation approach to map any analog transistor to PANDA cell is proposed, which achieves transistor level matching accuracy of less than 5% for ID and less than 10% for Rout and Gm. Circuit level analog metrics of a voltage-controlled oscillator (VCO) emulated by PANDA, match to those of the original designs in 90nm nodes with less than a 5% error. Voltage-controlled delay lines at 65nm and 90nm are emulated by 32nm PANDA, which successfully match important analog metrics. And at-speed emulation is achieved as well. Several other 90nm analog blocks are successfully emulated by the 45nm PANDA platform, including a folded-cascode operational amplifier and a sample-and-hold module (S/H)

The fluorescence enhancement by a single Noble metal sphere is separated into excitation/absorption enhancement and the emission quantum yield enhancement. Incorporating the classical model of molecular spontaneous emission into the excitation/absorption transition, the excitation enhancement is calculated rigorously by electrodynamics in the frequency domain. The final formula for the excitation enhancement contains two parts: the primary field enhancement calculated from the Mie theory, and a derating factor due to the backscattering field from the molecule. When compared against a simplified model that only involves the primary Mie theory field calculation, this more rigorous model indicates that the excitation enhancement near the surface of the sphere is quenched severely due to the back-scattering field from the molecule. The degree of quenching depends in part on the bandwidth of the illumination because the presence of the sphere induces a red-shift in the absorption frequency of the molecule and at the same time broadens its spectrum. Monochromatic narrow band illumination at the molecule's original (unperturbed) resonant frequency yields large quenching. For the more realistic broadband illumination scenario, we calculate the final enhancement by integrating over the excitation/absorption spectrum. The numerical results indicate that the resonant illumination scenario overestimates the quenching and therefore would underestimate the total excitation enhancement if the illumination has a broader bandwidth than the molecule. Combining the excitation model with the exact Electrodynamical theory for emission, the complete realistic model demonstrates that there is a potential for significant fluorescence enhancement only for the case of a low quantum yield molecule close to the surface of the sphere. General expressions of the fluorescence enhancement for arbitrarily-shaped metal antennas are derived. The finite difference time domain method is utilized for analyzing these complicated antenna structures. We calculate the total excitation enhancement for the two-sphere dimer. Although the enhancement is greater in this case than for the single sphere, because of the derating effects the total enhancement can never reach the local field enhancement. In general, placing molecules very close to a plasmonic antenna surface yields poor enhancement because the local field is strongly affected by the molecular self-interaction with the metal antenna.

This dissertation proposes a miniature FIR filter that works at microwave frequencies, whose filter response can ideally be digitally programmed. Such a frequency agile device can find applications in cellular communications and wireless networking. The basic concept of the FIR filter utilizes a low loss acoustic waveguide of appropriate geometry that acts as a traveling wave tapped-delay line. The input RF signal is applied by an array of capacitive transducers at various locations on the acoustic waveguide at one end that excites waves of a propagating acoustic mode with varying spatial delays and amplitudes which interfere as they propagate. The output RF signal is picked up at the other end of the waveguide by another array of capacitive transducers. Tuning of the FIR filter coefficients is realized by controlling the DC voltage profile applied to the individual transducers which essentially shapes the overall filter response. Equivalent circuit modeling of the capacitive transducer, acoustic waveguide and transducer-line coupling is presented in this dissertation. A theoretical model for the filter is developed from a general theory of an array of transducers exciting a waveguide and is used to obtain a set of filter design equations. A MATLAB based circuit simulator is developed to simulate the filter responses. Design parameters and simulation results obtained for an example waveguide structure are presented and compared to the values estimated by the theoretical model. A waveguide structure utilizing the Rayleigh-like mode of a ridge is then introduced. A semi-analytical method to obtain propagating elastic modes of such a ridge waveguide etched in an anisotropic crystal is presented. Microfabrication of a filter based on ridges etched in single crystal Silicon is discussed along with details of the challenges faced. Finally, future work and a few alternative designs are presented that can have a better chance of success. Analysis and modeling work to this point has given a good understanding of the working principles, performance tradeoffs and fabrication pitfalls of the proposed device. With the appropriate acoustic waveguide structure, the proposed device could make it possible to realize miniature programmable FIR filters in the GHz range.

When ferrite materials are used in antenna designs, they introduce some interesting and unique performance characteristics. One of the attractive features, for example, is the ability to reconfigure the center frequency of the antenna. In addition, ferrite materials also introduce a number of challenges in the modeling and simulation of such antennas. In order for the ferrite material to be useful in an antenna design, it usually is subjected to an external magnetic field. This field induces the internal magnetic field inside the ferrite material. The internal field plays a pivotal role in the radiation characteristics of the antenna. Thus, from the numerical point of view, accurate computation of this field is critical to the overall accuracy of the analysis. Usually the internal field is non-uniform and its computation is often a rather complex and non-trivial task. Therefore, to facilitate the modeling, simplifying assumptions, which introduce some kind of averaging, are often made. In this study, ferrite-loaded cavity-backed slot antennas are used to demonstrate that averaging procedures can lead to very unsatisfactory results. For instance, it is common practice to assume that the external field is uniform by averaging its distribution. One of the pivotal points in this study is the demonstration that the external magnetic field plays a very significant role and should be included in the modeling without averaging, if the accurate results are to be attained. Results presented in this study clearly support this argument. A procedure which avoids such averaging is presented and verified by comparing simulations with measurements. In contrast to the previous formulations, the modeling methodology developed in this dissertation leads to accurate results which compare very well with measurements for both uniform and non-uniform field distributions. The utility of this methodology is especially evident for the case when the magnetic field is severely non-uniform.

The purpose of the solar powered quadcopter is to join together the growing technologies of photovoltaics and quadcopters, creating a single unified device where the technologies harmonize to produce a new product with abilities beyond those of a traditional battery powered drone. Specifically, the goal is to take the battery-only flight time of a quadcopter loaded with a solar array and increase that flight time by 33% with additional power provided by solar cells. The major concepts explored throughout this project are quadcopter functionality and capability and solar cell power production. In order to combine these technologies, the solar power and quadcopter components were developed and analyzed individually before connecting the solar array to the quadcopter circuit and testing the design as a whole. Several solar copter models were initially developed, resulting in multiple unique quadcopter and solar cell array designs which underwent preliminary testing before settling on a finalized design which proved to be the most effective and underwent final timed flight tests. Results of these tests are showing that the technologies complement each other as anticipated and highlight promising results for future development in this area, in particular the development of a drone running on solar power alone. Applications for a product such as this are very promising in many fields, including the industries of power, defense, consumer goods and services, entertainment, marketing, and medical. Also, becoming a more popular device for UAV hobbyists, such developments would be very appealing for leisure flying and personal photography purposes as well.

A hybrid PV/T module was built, consisting of a thermal liquid heating system and a photovoltaic module system that combine in a hybrid format. This report will discuss the work on the project from Fall 2012 to Spring 2013. Three stages of experiments were completed. Stage 1 showed our project was functional as we were able to verify our panel produced electricity and increased the temperature of water flowing in the system by 0.65°C. Stage 2 testing included “gluing” the flow system to the back of the panel resulting in an average increase of 4.76°C in the temperature of the water in the system. Stage 3 testing included adding insulating foam to the module which resulted in increasing the average temperature of the water in our flow system by 6.95°C.

In this thesis two methodologies have been proposed for evaluating the fault response of analog/RF circuits. These proposed approaches are used to evaluate the response of the faulty circuit in terms of specifications/measurements. Faulty response can be used to evaluate important test metrics like fail probability, fault coverage and yield coverage of given measurements under process variations. Once the models for faulty and fault free circuit are generated, one needs to perform Monte Carlo sampling (as opposed to Monte Carlo simulations) to compute these statistical parameters with high accuracy. The first method is based on adaptively determining the order of the model based on the error budget in terms of computing the statistical metrics and position of the threshold(s) to decide how precisely necessary models need to be extracted. In the second method, using hierarchy in process variations a hybrid of heuristics and localized linear models have been proposed. Experiments on LNA and Mixer using the adaptive model order selection procedure can reduce the number of necessary simulations by 7.54x and 7.03x respectively in the computation of fail probability for an error budget of 2%. Experiments on LNA using the hybrid approach can reduce the number of necessary simulations by 21.9x and 17x for four and six output parameters cases for improved accuracy in test statistics estimation.
