Separation from a loved one is a highly stressful event. The range and intensity of emotions accompanying such a separation arguably are amplified when one's spouse deploys. This thesis examines at-home spouses (AHSs) of deployed military and how emotion, marital satisfaction, and communication are impacted throughout the deployment cycle. Additionally, I explore technology as a possible coping mechanism to help AHSs adapt and overcome stressfulness of deployment. One hundred sixty-six married females with a partner currently deployed, anticipating deployment, or recently returned from deployment completed an on-line survey. It was predicted AHSs would experience specific emotions during each phase, categorized as "anticipatory," (e.g., anger, worry) "absence" (e.g., lonely, sad) or "post" (e.g., happiness, relief); marital satisfaction also was predicted to be higher among spouses whose partner recently returned from deployment versus was deployed or anticipating deployment. Data showed AHSs whose partner was anticipating or currently deployed reported more "anticipatory" and "absence" emotions than AHSs with a recently returned partner. The former two groups did not differ in these emotions. AHSs with a recently returned partner reported more "post" emotions than the other two groups. Marital satisfaction did not differ based on deployment status. It was also predicted that among AHSs with a currently deployed partner, less negative emotion upon deployment would be associated with more frequent communication during deployment. Data showed AHSs who reported less negative emotion upon deployment engaged in more frequent communication with their deployed partner. Lastly, I predicted AHSs whose partners are currently deployed and who prefer modes of communication allowing direct contact (e.g., Skype) will experience less negative emotions than AHSs who prefer indirect contact (e.g., e-mail). Data showed reports of negative emotion did not differ based on preference for direct versus indirect communication. Therefore, negative emotions may develop and persist before and during deployment, but when the partner returns home, spouses do experience a rebound of positive emotions. Additionally, emotions at the time of deployment may be useful in predicting spouses' communication frequency during deployment. Findings aim to provide knowledge of family life during separation and explore technology as a possible coping mechanism for AHSs.

Anti-retroviral drugs and AIDS prevention programs have helped to decrease the rate of new HIV-1 infections in some communities, however, a prophylactic vaccine is still needed to control the epidemic world-wide. Despite over two decades of research, a vaccine against HIV-1 remains elusive, although recent clinical trials have shown promising results. Recent successes have focused on highly conserved, mucosally-targeted antigens within HIV-1 such as the membrane proximal external region (MPER) of the envelope protein, gp41. MPER has been shown to play critical roles in the viral mucosal transmission, though this peptide is not immunogenic on its own. Gag is a structural protein configuring the enveloped virus particles, and has been suggested to constitute a target of the cellular immunity potentially controlling the viral load. It was hypothesized that HIV-1 enveloped virus-like particles (VLPs) consisting of Gag and a deconstructed form of gp41 comprising the MPER, transmembrane, and cytoplasmic domains (dgp41) could be expressed in plants. Plant-optimized HIV-1 genes were constructed and expressed in Nicotiana benthamiana by stable transformation, or transiently using a tobacco mosaic virus-based expression system or a combination of both. Results of biophysical, biochemical and electron microscopy characterization demonstrated that plant cells could support not only the formation of HIV-1 Gag VLPs, but also the accumulation of VLPs that incorporated dgp41. These particles were purified and utilized in mice immunization experiments. Prime-boost strategies combining systemic and mucosal priming with systemic boosting using two different vaccine candidates (VLPs and CTB-MPR - a fusion of MPER and the B-subunit of cholera toxin) were administered to BALB/c mice. Serum antibody responses against both the Gag and gp41 antigens could be elicited in mice systemically primed with VLPs and these responses could be recalled following systemic boosting with VLPs. In addition, mucosal priming with VLPs allowed for a robust boosting response against Gag and gp41 when boosted with either candidate. Functional assays of these antibodies are in progress to test the antibodies' effectiveness in neutralizing and preventing mucosal transmission of HIV-1. This immunogenicity of plant-based Gag/dgp41 VLPs represents an important milestone on the road towards a broadly-efficacious and inexpensive subunit vaccine against HIV-1.

Dangerous drinking on college campuses is a significant public health issue. Over the last decade, the National Institute on Alcohol Abuse and Alcoholism and the U.S. Department of Health and Human Services have called on universities, community leaders, policymakers, parents and students to work together to develop effective, research based alcohol prevention and/or intervention programs. Despite such calls, parent-based prevention programs are relatively rare on college campuses, and there is a paucity of research on the ways in which parents influence their emerging adult children's drinking behaviors. The present project is designed to help address this need. Grounded in social cognitive theory, this exploratory study focuses on alcohol communication and poses numerous questions regarding the alcohol messages exchanged between college students and their parents, as well as how such messages associate with college students' dangerous drinking. Undergraduate students ages 18 to 25 who were enrolled in communication classes were recruited for the study and asked to recruit a parent. The sample included 198 students and 188 parents, all of whom completed an online survey. Results indicated the majority of college students have had alcohol conversations with a parent since the student graduated from high school. Parents viewed such conversations as significantly more open, direct, and ongoing than did students; though both generally agreed on the content of their alcohol communication, reporting an emphasis on the negative aspects of drinking, particularly the dangers of drinking and driving and the academic consequences of too much partying. Frequent discussions of drinking risks had significant, positive associations with students' dangerous drinking, whereas parents' reports of discussing rules about alcohol had a significant negative association with students' alcohol consumption. There were strong significant associations between the types alcohol topics discussed and students' perception that their parents approved of their drinking, as well as parents' actual approval. Perceived approval had a significant, positive association with students' dangerous drinking; however, actual parental approval was not a significant predictor of students' drinking outcomes. Parents' alcohol consumption had a significant positive association with students' alcohol consumption. Implications for parents, public health practitioners, and future research are discussed.

ABSTRACT The goal of this study is to use neoclassical realist methodology to add to the growing body of literature explaining why America is failing so horribly in its media war with militant Islamists. The general argument being conveyed is that inconsistencies in America's ostensibly liberal diplomacy strategy leaves it open to criticism and deprives it of the credibility necessary to muster an adequate rebuttal. To accomplish its aim, the analysis begins with an investigation into the origins of America's current liberal rhetorical approach. It is believed that with this sort look beneath the surface of the idealistic romanticism U.S. citizens have been continually conditioned to embrace, it becomes apparent that the grandiose pronouncements made by America's national political elite are actually based on rather dubious foundations. The evaluation then turns to a more focused rhetorical examination, which spans from the start of the so-called Arab Spring uprisings on December 18, 2010 to the delivery of President Obama's highly publicized State Department address regarding these demonstrations on May 19, 2011, in order to go behind the White House's official statements and uncover what truly motivated its policy decision making. The belief here is that a close review of the administration's abysmal performance during this historic period assists in making the inadequacy of America's current rhetorical narrative all the more evident. Finally, once the contradictory nature of contemporary American liberalism has been fully demonstrated, the last section concludes with an effort to explain why replacing America's liberal strategy with a straightforward realist stance is best for both American's relations with the Muslim world and America's overall security.

Every communication system has a receiver and a transmitter. Irrespective if it is wired or wireless.The future of wireless communication consists of a massive number of transmitters and receivers. The question arises, can we use computer vision to help wireless communication? To satisfy the high data requirement, a large number of antennas are required. The devices that employ large-antenna arrays have other sensors such as RGB camera, depth camera, or LiDAR sensors.These vision sensors help us overcome the non-trivial wireless communication challenges, such as beam blockage prediction and hand-over prediction.This is further motivated by the recent advances in deep learning and computer vision that can extract high-level semantics from complex visual scenes, and the increasing interest of leveraging machine/deep learning tools in wireless communication problems.[1] <br/><br/>The research was focused solely based on technology like 3D cameras,object detection and object tracking using Computer vision and compression techniques. The main objective of using computer vision was to make Milli-meter Wave communication more robust, and to collect more data for the machine learning algorithms. Pre-build lossless and lossy compression algorithms, such as FFMPEG, were used in the research. An algorithm was developed that could use 3D cameras and machine learning models such as YOLOV3, to track moving objects using servo motors and low powered computers like the raspberry pi or the Jetson Nano. In other words, the receiver could track the highly mobile transmitter in 1 dimension using a 3D camera. Not only that, during the research, the transmitter was loaded on a DJI M600 pro drone, and then machine learning and object tracking was used to track the highly mobile drone. In order to build this machine learning model and object tracker, collecting data like depth, RGB images and position coordinates were the first yet the most important step. GPS coordinates from the DJI M600 were also pulled and were successfully plotted on google earth. This proved to be very useful during data collection using a drone and for the future applications of position estimation for a drone using machine learning. <br/><br/>Initially, images were taken from transmitter camera every second,and those frames were then converted to a text file containing hex-decimal values. Each text file was then transmitted from the transmitter to receiver, and on the receiver side, a python code converted the hex-decimal to JPG. This would give an efect of real time video transmission. However, towards the end of the research, an industry standard, real time video was streamed using pre-built FFMPEG modules, GNU radio and Universal Software Radio Peripheral (USRP). The transmitter camera was a PI-camera. More details will be discussed as we further dive deep into this research report.

As digital technology promises immediacy and interactivity in communication, sight and sound in motion graphics has expanded the range of design possibilities in advertising, social networking, and telecommunication beyond the visual realm. The experience of seeing has been greatly enriched by sound as visual solutions become dynamic and multi-dimensional. The ability to record and transfer sight and sound with new media has granted the designer more control in manipulating a viewer's experience of time and space. This control allows time-based form to become the foundation that establishes many interactive, multisensory and interdisciplinary applications. Is conventional design theory for print media adequate to effectively approach time-based form? If not, what is the core element that is required to balance the static and dynamic aspects of time in new media? Should time-related theories and methodologies from other disciplines be adopted into our design principles? If so, how would this knowledge be integrated? How can this experience in time be effectively transferred to paper? Unless the role of the time dimension in sight is operationally deconstructed and retained with sound, it is very challenging to control the design in this fugitive form. Time activation refers to how time and the perception of time can be manipulated for design and communication purposes. Sound, as a shortcut to the active time design element, not only encapsulates the structure of its "invisible" time-based form, but also makes changes in time conspicuously measurable and comparable. Two experiments reflect the influence of sound on imagery, a slideshow and video, as well as how the dynamics in time are represented across all design media. A cyclical time-based model is established to reconnect the conventional design principles learned in print media with time-based media. This knowledge helps expand static images to motion and encapsulate motion in stasis. The findings provide creative methods for approaching visualization, interactivity, and design education.

The cyanobacterium Synechocystis sp. PCC 6803 performs oxygenic photosynthesis. Light energy conversion in photosynthesis takes place in photosystem I (PSI) and photosystem II (PSII) that contain chlorophyll, which absorbs light energy that is utilized as a driving force for photosynthesis. However, excess light energy may lead to formation of reactive oxygen species that cause damage to photosynthetic complexes, which subsequently need repair or replacement. To gain insight in the degradation/biogenesis dynamics of the photosystems, the lifetimes of photosynthetic proteins and chlorophyll were determined by a combined stable-isotope (15N) and mass spectrometry method. The lifetimes of PSII and PSI proteins ranged from 1-33 and 30-75 hours, respectively. Interestingly, chlorophyll had longer lifetimes than the chlorophyll-binding proteins in these photosystems. Therefore, photosynthetic proteins turn over and are replaced independently from each other, and chlorophyll is recycled from the damaged chlorophyll-binding proteins. In Synechocystis, there are five small Cab-like proteins (SCPs: ScpA-E) that share chlorophyll a/b-binding motifs with LHC proteins in plants. SCPs appear to transiently bind chlorophyll and to regulate chlorophyll biosynthesis. In this study, the association of ScpB, ScpC, and ScpD with damaged and repaired PSII was demonstrated. Moreover, in a mutant lacking SCPs, most PSII protein lifetimes were unaffected but the lifetime of chlorophyll was decreased, and one of the nascent PSII complexes was missing. SCPs appear to bind PSII chlorophyll while PSII is repaired, and SCPs stabilize nascent PSII complexes. Furthermore, aminolevulinic acid biosynthesis, an early step of chlorophyll biosynthesis, was impaired in the absence of SCPs, so that the amount of chlorophyll in the cells was reduced. Finally, a deletion mutation was introduced into the sll1906 gene, encoding a member of the putative bacteriochlorophyll delivery (BCD) protein family. The Sll1906 sequence contains possible chlorophyll-binding sites, and its homolog in purple bacteria functions in proper assembly of light-harvesting complexes. However, the sll1906 deletion did not affect chlorophyll degradation/biosynthesis and photosystem assembly. Other (parallel) pathways may exist that may fully compensate for the lack of Sll1906. This study has highlighted the dynamics of photosynthetic complexes in their biogenesis and turnover and the coordination between synthesis of chlorophyll and photosynthetic proteins.

The goal of this research was to contribute to the understanding of how the physical design of Intensive Care Unit (ICU) environments may be improved to enhance nursing communication, and in turn, the quality and safety of patient outcomes. This study was guided by two research questions: (1) What are the major characteristics of nurse communication in a hybrid ICU nurse station design? (2) What are the factors in the built environment that enhance or hinder nurse communication in a hybrid ICU nurse station design? The research design was exploratory and qualitative. Observations were conducted in two ICUs with hybrid nurse station layouts. Participant observation was used to systematically observe and document nurse communication and the physical attributes of the ICU nurse work environment that affect communication. Literature, observations, and information regarding staffing and design about the selected ICUs were analyzed for the generation of concepts and the exploration of significant themes. Results show that nurse interactions with other staff members varied within the different zones of the ICU pod. A biaxial map illustrates four key types of core nurse communication interactions: At ease, On guard, In motion, and On the edge. The quadrants representing barriers to nurse communication are On guard and On the edge, and included interactions with other staff members in the pod. The quadrants representing facilitators to nurse communication are At ease and In motion. The hybrid nurse station layout supported nurse-nurse communication, but not communication interactions with other staff members present on the pod. The results provide a broad understanding of how nurse communication is affected by the environment in which nurses work, and allows for the emergence of design opportunities to enhance nurse communication.

Non-native saltcedar (Tamarix spp.) has invaded many riparian communities and is the third most abundant tree in Southwestern riparian areas. I evaluated lizard populations and microhabitat selection during 2009 and 2010 along the Virgin River in Nevada and Arizona to determine the impact of saltcedar. Along the riparian corridor, I observed common side-blotched lizards (Uta stansburiana) within two vegetation types: monotypic non-native saltcedar stands or mixed stands of cottonwood (Populus fremontii), willow (Salix spp.), mesquite (Prosopis spp.) and saltcedar. I predicted that population parameters such as body condition, adult to hatchling ratio, abundance, and persistence would vary among vegetation types. Also, I predicted the presence of saltcedar influences how lizards utilize available habitat. Lizard population parameters were obtained from a mark-recapture study in which I captured 233 individual lizards. I examined habitat selection and habitat availability using visual encounter surveys (VES) for lizards and recorded 11 microhabitat variables where 16 lizards were found. I found no significant difference in population parameters between mixed and non-native saltcedar communities. However, population parameters were negatively correlated with canopy cover. I found that lizards selected habitat with low understory and canopy cover regardless of vegetation type. My results indicate that lizards utilize similar structural characteristics in both mixed and non-native vegetation. Understanding impacts of saltcedar on native fauna is important for managers who are tasked with control and management of this non-native species.

There is increasing evidence that ovarian status influcences behavioral phenotype in workers of the honey bee Apis mellifera. Honey bee workers demonstrate a complex division of labor. Young workers perform in-hive tasks (e.g. brood care), while older bees perform outside tasks (e.g. foraging for food). This age correlated division of labor is known as temporal polyethism. Foragers demonstrate further division of labor with some bees biasing collection towards protein (pollen) and others towards carbohydrates (nectar). The Reproductive Ground-plan Hypothesis proposes that the ovary plays a regulatory role in foraging division of labor. European honey bee workers that have been selectively bred to store larger amounts of pollen (High strain) also have a higher number of ovarioles per ovary than workers from strains bred to store less pollen (Low strain). High strain bees also initiate foraging earlier than Low strain bees. The relationship between ovariole number and foraging behavior is also observed in wild-type Apis mellifera and Apis cerana: pollen-biased foragers have more ovarioles than nectar-biased foragers. In my first study, I investigated the pre-foraging behavioral patterns of the High and Low strain bees. I found that High strain bees progress through the temporal polyethism at a faster rate than Low strain bees. To ensure that the observed relationship between the ovary and foraging bias is not due to associated separate genes for ovary size and foraging behavior, I investigated foraging behavior of African-European backcross bees. The backcross breeding program was designed to break potential gene associations. The results from this study demonstrated the relationship between the ovary and foraging behavior, supporting the proposed causal linkage between reproductive development and behavioral phenotype. The final study was designed to elucidate a regulatory mechanism that links ovariole number with sucrose sensitivity, and loading decisions. I measured ovariole number, sucrose sensitivity and sucrose solution load size using a rate-controlled sucrose delivery system. I found an interaction effect between ovariole number and sucrose sensitivity for sucrose solution load size. This suggests that the ovary impacts carbohydrate collection through modulation of sucrose sensitivity. Because nectar and pollen collection are not independent, this would also impact protein collection.
