White-nose syndrome (WNS) is a fungal infection devastating bat populations throughout eastern North America. WNS is caused by a fungus, Pseudogymnoascus destructans (Pd), that invades the skin of hibernating bats. While there are a number of treatments being researched, there is currently no effective treatment for WNS that is deployed in the field, except a few being tested on a limited scale. Bats have lowered immune function and response during hibernation, which may increase susceptibility to infection during the winter months. Antimicrobial peptides (AMPs) are a crucial component of the innate immune system and serve as barriers against infection. AMPs are constitutively expressed on skin and facilitate wound healing, stimulate other immune responses, and may also stay active on bat skin during hibernation. AMPs are expressed by all tissues, have direct killing abilities against microbes, and are a potential treatment for bats infected with Pd. In this investigation, the fungicidal activity of several readily available commercial AMPs were compared, and killing assay protocols previously investigated by Frasier and Lake were replicated to establish a control trial for use in future killing assays. Another aim of this investigation was to synthesize a bat-derived AMP for use in the killing assay. Sequences of bat-derived AMPs have been identified in bat skin samples obtained from a large geographic sampling of susceptible and resistant species. Contact was made with GenScript Inc., the company from which commercially available AMPs were purchased, to determine the characteristics of peptide sequences needed to synthesize an AMP for lab use. Based on recommendations from GenScript Inc., peptide sequences need to have a hydrophobicity of less than 50% and a sequence length of less than 50 amino acids. These criteria serve as a potential barrier because none of the known bat-derived sequences analyzed satisfy both of these requirements. The final aim of this study was to generate a conceptual model of the immune response molecules activated when bats are exposed to a fungal pathogen such as Pd. Overall, this work investigated sources of variability between trials of the killing assay, analyzed known bat-derived peptide sequences, and generated a conceptual model that will serve as a guideline for identification of immune response molecules on the skin of bats in future proteomics work.

Conservation is a complicated entity consisting of a multitude of professional fields including social issues, cultural issues, and physical science. This thesis evaluates the positive and negative aspects of two broad types of conservation: top down fortress conservation and bottom up community-based conservation. Fortress conservation has many negative aspects, such as displacing human communities and preventing utilization of resources. However, it also has positive aspects, such as preventing the destruction of delicate ecosystems and slowing down extinctions. Community-based conservation is more inclusive and focuses on including the indigenous populations located within the proposed conservation site in the decision-making process. Its negatives include having an anthropocentric goal instead of valuing nature's intrinsic values. Understanding the differences inherent in these two methods is necessary in order to implement a conservation network with the highest chance for success.

The purpose of my thesis is to illustrate the story of the black-footed ferret's conservation, and to provide my own suggestions for how to eventually get the species removed from the Endangered Species List, marking a successful rebound in population numbers. The purpose of my thesis is to illustrate the story of the black-footed ferret's conservation, and to provide my own suggestions for how to eventually get the species removed from the Endangered Species List, marking a successful rebound in population numbers. I highlight my personal experience working at the Phoenix Zoo's black-footed ferret breeding center. In the first chapter, I present the species by describing its morphology, diet, reproduction, behaviors, range, and habitat. In the second chapter, I recount the chronological history of the conservation of the species, starting with its rediscovery following its putative "extinction", and ending with its present status. In the third chapter, I discuss the threats that have led to the species' overall decline and continue to affect its persistence today. In the fourth and final chapter, I conclude by making recommendations regarding what needs to occur in order to eventually get the species delisted.

The setting of the story is Phoenix, Arizona, a city that is made up of a plethora of natural wonders and a diverse, rich culture. However, our city also has a large population of undocumented citizens, gang violence, social segregation, drug trade activity, and political polarization. Recently, immigrants and minorities have been blamed for most of these problems. The origins of crime and the drug trade must be addressed, so that incoming immigrants have a fair opportunity at the American Dream. Americans must understand this, as we are a nation of immigrants, and knowing our history will ground us in our identity, as well as redirect us to our core values. I decided to write this rap musical for the people in the ghettoes all around the world, but especially for the people in my barrio. To know where we are headed, it helps to know where we come from.

Various forms of media have contributed to and shaped negative perception of immigrants and refugees in the United States. These perceptions may contribute to dangerous stereotyping, which may impact policy and the daily life of immigrants in the United States. Various factors must be considered when analyzing media, such as the type of media, language used, consumption, and the reaction of viewers. This thesis focuses on AZcentral, an Arizona news source, and how it portrays immigrants versus the commentary from an immigrant here in Arizona. First it is important to look at time periodization of the portrayal of immigrants and how it has changed through technology mediums. The information obtained by looking at these periods provide specific terms and content to look for when collecting sources for the podcast. In the podcast, AZcentral and the interviewee stories are compared to show how different or similar they may be. Through this, the audience should be making their own conclusions to show accurate AZcentral is portraying the immigrant experience.

The use of genetic management in conservation has sparked much debate around the ethical and environmental impacts of the plans. A case study on the conservation of leopard frogs in Arizona was analyzed to better understand the benefits and issues surrounding genetic management plans. The first part of the case focuses on the recent management plan for Chiricahua Leopard Frogs implemented by the Arizona Game and Fish Department. The goal of the plan is to better understand the genetic dynamics of the established Chiricahua Leopard Frog populations to develop a more effective management plan. The second part of the case focuses on the Arizona Game and Fish Department’s management of the Northern Leopard Frog. There was little success with the initial breed and release program of the native species, however a nonnative subspecies of Northern Leopard Frog was able to establish a thriving population. This case study exemplifies the many complications with genetic management plans and the importance of careful assessment of options when deciding on a genetic management plan. Despite the complexity of genetic management plans, it is an important method to consider when discussing the conservation of a species.

The population of undocumented immigrants includes over a million individuals known as “Dreamers,” who are for all intents and purposes naturalized Americans but are not counted among the American citizens. Dreamers have grown up believing in the American dream of one day being able to develop themselves into hard-working, accomplished and fully recognized Americans. Unfortunately, however, Dreamers are caught in a limbo of hope: their undocumented status forces them to work and live in the “underground,” or risk being deported to an unfamiliar country. After years of activism, the Dreamers found limited protection under an executive action introduced by former-President Barack Obama, known as the Deferred Action for Childhood Arrivals (DACA). DACA deferred deportation and gave the Dreamer the ability to legally work within the United States. Unfortunately, DACA’s provisional protection of hundreds of thousands of Dreamers also created gaps of its own: many other naturalized residents have been excluded and those covered by the DACA program remain limited in education and work. DACA’s protection is also limited in practice and time; it lasts for only two years before needing to be renewed and it can be revoked at any time. Only through legislation creating a “path to citizenship” and/or through formal naturalization can the Dreamers’ nightmare end and normalcy be instated. But precisely this idea of a “path to citizenship” has become the object of invective by anti-immigrant politics of Trump and the Republican Party. The Republican Party has worked to define itself as the party of White America by fomenting fear and anxiety over immigrant populations, whose mere existence or growth it presents as a threat. With continuing Republican government control, the Dreamer will not be able to legalize their status and will remain to be limited with their American dreams.

The Tale of the Tigers is a children’s picture book in which a tiger named Guava is transported from his world of Serenia to our world. In an attempt to find someone to help him with this situation, Guava meets another tiger named Papaya. After having his prey scared away by Guava, Papaya begins to explain the different hardships and dangers that tigers face in their natural habitat. Papaya also teaches Guava about the different programs and activities that humans have been doing to help increase and restore tiger populations. At the end of the story, Guava returns to his world and spreads awareness to those that live in Serenia about how tigers are threatened in other places and what’s being done to help them. Papaya uses basic ecological concepts to explain the importance of tigers in their ecosystem. These concepts include habitat loss, trophic levels, landscape fragmentation, and poaching. The story also incorporates different conservation methods, including captive breeding and the use of camera traps.
