ABSTRACT The experience of grief and loss is a process that can be extremely distressing to anyone, regardless of age. This may be especially true for youth. This study was designed and conducted to determine the effects of a therapy dog as a therapeutic adjunct in Child Life interventions with adolescents experiencing grief and loss. Subjects were randomly assigned to one of two groups. The intervention consisted of 3 sessions with a Certified Child Life Specialist (CCLS) to address grief. Group 1 (N=14) was the control group, meeting only with the CCLS. Group 2 (N=13) was the experimental group and met with the CCLS with a therapy dog present during the sessions. Participants completed a pre-test and post-test of the Children's Mood Questionnaire. At the end of each session, subjects completed a Therapeutic Engagement Questionnaire. The pet therapy group experienced a significant improvement in mood scores on the Children's Mood Questionnaire following the intervention. However, there were no significant differences between groups on the Therapeutic Engagement Questionnaire during any of the 3 sessions. The data collected from this study indicate that the addition of a therapy dog in grief interventions with adolescents may improve mood outcomes.

In species with highly heteromorphic sex chromosomes, the degradation of one of the sex chromosomes can result in unequal gene expression between the sexes (e.g., between XX females and XY males) and between the sex chromosomes and the autosomes. Dosage compensation is a process whereby genes on the sex chromosomes achieve equal gene expression which prevents deleterious side effects from having too much or too little expression of genes on sex chromsomes. The green anole is part of a group of species that recently underwent an adaptive radiation. The green anole has XX/XY sex determination, but the content of the X chromosome and its evolution have not been described. Given its status as a model species, better understanding the green anole genome could reveal insights into other species. Genomic analyses are crucial for a comprehensive picture of sex chromosome differentiation and dosage compensation, in addition to understanding speciation.

In order to address this, multiple comparative genomics and bioinformatics analyses were conducted to elucidate patterns of evolution in the green anole and across multiple anole species. Comparative genomics analyses were used to infer additional X-linked loci in the green anole, RNAseq data from male and female samples were anayzed to quantify patterns of sex-biased gene expression across the genome, and the extent of dosage compensation on the anole X chromosome was characterized, providing evidence that the sex chromosomes in the green anole are dosage compensated.

In addition, X-linked genes have a lower ratio of nonsynonymous to synonymous substitution rates than the autosomes when compared to other Anolis species, and pairwise rates of evolution in genes across the anole genome were analyzed. To conduct this analysis a new pipeline was created for filtering alignments and performing batch calculations for whole genome coding sequences. This pipeline has been made publicly available.

This dissertation explores discourses in the contemporary United States surrounding the creation, coding, sterilization, and general keeping of canines in order to interrogate how sex, gender, race, class, sexuality, and species together serve biopolitical formations of social control, patriarchal white supremacy, and heteronormativity. Interrogating these socially constructed and oftentimes stereotypical narratives through an interspecies lens demonstrates how taxonomies of power and systems of oppression and privilege become situated across species. This project utilizes interviews and ethnography, as well as analysis of popular culture, legislation and news media.

Interspeciesism is informed by feminist influences, functioning as a framing paradigm that engages with a politicized question of the animal that explicitly acknowledges human-animal entanglements across sites that are shaped by imperialism and colonialism. This interspecies project considers the political nature of relationships between humans and canines. It suggests that people situate their own identities and power not only in relation to other humans but also to other species. Simultaneously, the interspeciesm I engage with extends analyses of biopolitics, or the regulations of living bodies, beyond humans to all species. It interrogates how contemporary U.S. society has organized and identified itself in part through the ways in which it controls and monitors canines, often in relationship to the multiple ways dogs in the U.S. are racialized, classed and gendered by specific breeds. This coding of canine bodies with various taxonomies of power is not about dog breeds’ in-and-of themselves, but instead indicates that dominant U.S. society seeks to assert control over certain populations that are constructed as undesirable and unproductive.

Canines exist in a unique space in the U.S. cultural imaginary where they have multiple and oftentimes contradictory meanings that are influenced by a variety of power relations that transcend species. At stake is a critical concern regarding how interspecies bodies are made, controlled, formed, and refigured together under heteropatriarchal white supremacist modes of power. It draws attention to what these corporeal un(makings) imply for an ethics of being with, and thinking of, the other—human and animal.

Objective: To explore the dimensions of the human animal bond and provide a community needs assessment to inform the community stake holders such as the Arizona Humane Society and Nina Mason Pulliam Charitable Trust Foundation how many animals are in need of veterinary services within the homeless population of Phoenix, Arizona. In addition to this, pets of the homeless individuals will be able to gain access to veterinary services for eight consecutive weeks. Background: Pets have an important impact on human mental, physical, social, and emotional health. It has been reported that about one third of the homeless population in Arizona has pets that are not able to gain access to veterinary care (Wang, 2015). Most homeless shelters will not allow people to access services with pets. As a consequence people will sleep out in the streets. Animals as Lifechangers and Lifesavers: Pets in the Redemption Narratives of Homeless People (Irvine, 2013) contains interviews of homeless people based on their life stories. A common theme among interviewees was that they felt they had a responsibility to their pets that served as a motivating purpose for giving up horrible personal habits because they had a sense of responsibility. Methods/Materials: Wandering Paws was launched in February 2015, but did not officially start as an eight-week study until March 2016. This pilot program serves the homeless populations' dogs and cats with veterinary care. The Arizona Humane Society was approached to acquire their services for this project including a veterinarian, a technician, and usage of their seventy-one foot mobile unit. Homeless individuals who wanted veterinary services were recruited and asked to fill out a twenty-three-question survey. Secondary data was procured from the Arizona Humane Society about the animal and services rendered for that pet. Results: Over the course of the first four weeks 22 surveys have been completed. 86% of the surveys completed indicate a strong bond between the owner and animal. The remaining 14% of the surveys completed indicate a weaker bond between the animal and owner. Conclusion/Implications: The research indicates a strong connection between most people and their animals. The veterinary services provided for the homeless population should be continued on a monthly basis as a wellness clinic in the future, as these services are in great demand.

Domestic dogs have assisted humans for millennia. However, the extent to which these helpful behaviors are prosocially motivated remains unclear. To assess the propensity of pet dogs to spontaneously and actively rescue distressed humans, this study tested whether sixty pet dogs would release their seemingly trapped owners from a large box. To examine the causal mechanisms that shaped this behavior, the readiness of each dog to open the box was tested in three conditions: 1) the owner sat in the box and called for help (“Distress” test), 2) an experimenter placed high-value food rewards in the box (“Food” test), and 3) the owner sat in the box and calmly read aloud (“Reading” test).

Dogs were as likely to release their distressed owner as to retrieve treats from inside the box, indicating that rescuing an owner may be a highly rewarding action for dogs. After accounting for ability, dogs released the owner more often when the owner called for help than when the owner read aloud calmly. In addition, opening latencies decreased with test number in the Distress test but not the Reading test. Thus, rescuing the owner could not be attributed solely to social facilitation, stimulus enhancement, or social contact-seeking behavior.

Dogs displayed more stress behaviors in the Distress test than in the Reading test, and stress scores decreased with test number in the Reading test but not in the Distress test. This evidence of emotional contagion supports the hypothesis that rescuing the distressed owner was an empathetically-motivated prosocial behavior. Success in the Food task and previous (in-home) experience opening objects were both strong predictors of releasing the owner. Thus, prosocial behavior tests for dogs should control for physical ability and previous experience.

A handheld metal noisemaker known as a “clicker” is widely used to train new behaviors in dogs; however, evidence for the superior efficacy of clickers as opposed to providing solely primary reinforcement or other secondary reinforcers in the acquisition of novel behavior in dogs is almost entirely anecdotal. Three experiments were conducted to determine under what circumstances a clicker may result in acquisition of a novel behavior more rapidly or to a higher level compared to other readily available reinforcement methods. In Experiment 1, three groups of 30 dogs each were trained to emit a novel sit and stay behavior of increasing duration with either the delivery of food alone, a verbal stimulus paired with food, or a clicker with food. The group that received only a primary reinforcer reached a significantly higher criterion of training success than the group trained with a verbal secondary reinforcer. Performance of the group experiencing a clicker secondary reinforcer was intermediate between the other two groups, but not significantly different from either. In Experiment 2, three different groups of 25 dogs each were shaped to emit a nose targeting behavior and then perform that behavior at increasing distances from the experimenter using the same three methods of positive reinforcement as in Experiment 1. No statistically significant differences between the groups were found. In Experiment 3, three groups of 30 dogs each were shaped to emit a nose-targeting behavior upon an array of wooden blocks with task difficulty increasing throughout testing using the same three methods of positive reinforcement as previously. No statistically significant differences between the groups were found. Overall, the findings suggest that both clickers and other forms of positive reinforcement can be used successfully in training a dog to perform a novel behavior, but that no positive reinforcement method has significantly greater efficacy than any other.

The regulation of gene expression, timing, location, and amount of a given project, ultimately affects the cellular structure and function. More broadly, gene regulation is the basis for cellular differentiation and development. However, gene expression is not uniform among individuals and varies greatly between genetic males and females. Males are hemizygous for the X chromosome, whereas females have two X chromosome copies. Contributing to the sex differences in gene expression between males and females are the sex chromosomes, X and Y. Gene expression differences on the autosomes and the X chromosome between males (46, XY) and females (46, XX) may help inform on the mechanisms of sex differences in human health and disease. For example, XX females are more likely to suffer from autoimmune diseases, and genetic XY males are more likely to develop cancer. Characterizing sex-specific gene expression among human tissues will help inform the molecular mechanisms driving sex differences in human health and disease. This dissertation covers a range of critical aspects in gene expression. In chapter 1, I will introduce a method to align RNA-Seq reads to a sex chromosome complement informed reference genome that considers the X and Y chromosomes' shared evolutionary history. Using this approach, I show that more genes are called as sex differentially expressed in several human adult tissues compared to a default reference alignment. In chapter 2, I characterize gene expression in an early formed tissue, the human placenta. The placenta is the DNA of the developing fetus and is typically XY male or XX female. There are well-documented sex differences in pregnancy complications, yet, surprisingly, there is no observable sex difference in expression of innate immune genes, suggesting expression of these genes is conserved. In chapter 3, I investigate gene expression in breast cancer cell lines. Cancer arises in part due to the disruption of gene expression. Here I show 19 tumor suppressor genes become upregulated in response to a synthetic protein treatment. In chapter 4, I discuss gene and allele-specific expression in Nasonia jewel wasp. Chapter 4 is a replication and extension study and discusses the importance of reproducibility.

To date, there have been few, if any, studies evaluating the venom toxin levels in dogs that have been naturally envenomated by pit vipers. Understanding venom toxin pharmacokinetics in a clinical setting is important for a variety of reasons, including the potential to better elucidate treatment options, prognosis, and other factors associated with pit viper envenomation. In addition, dogs serve as a comparative species to humans for evaluating pit viper envenomations. This pilot study’s primary objective was to address the question of “What do we see?” in dogs presenting for rattlesnake envenomation. To answer this question, we obtained serum from envenomated dogs presenting at three veterinary clinics, then used enzyme-linked immunosorbent assay (ELISA) and western blot analysis to measure total venom and key toxins in sera. Phospholipase A2, a primary venom toxin, was identified in a few samples by the western blot, and contributed to the positive correlation between percent echinocytes in the blood and venom concentration. Medical data records were compared to venom concentrations measured using ELISA to determine whether there were any significant correlations. First, the hematological results were compared. Clotting times showed a strong positive correlation, clotting times and platelets showed a negative correlation, while echinocytes and platelets showed no correlation. When compared to venom concentration, clotting times showed a negative correlation, while age showed a positive correlation. Weight and platelets were also compared to venom concentration, but no significant correlations were found. The logistics of this study provided a real-world model where time elapsed between envenomation and hospital admission, thus giving a realistic look at what occurs in both animal and human medicine.
