The focus of this project was to look at alternative treatments for endocrine resistant breast cancer (ERBC), which are breast cancers that have become resistant to hormone therapies such as Tamoxifen or aromatase inhibitors. The first part of this project involves investigating the relationship between histone de-acetylase inhibitor Vorinostat and Tamoxifen in MCF7 G11 cells, Tamoxifen resistant sub-clones, according to the PSOC Time grant. The second part involves targeting the androgen receptor (AR) in MCF7 sub-clones with AR antagonists, Bicalutamide and MDV3100, and investigating the possible usage of AR as a biomarker, due to over-expression of AR in ERBC, in accordance with the Mayo ASU Seed Grant.
The synergistic effects between Vorinostat and Tamoxifen observed through a phase II study on breast cancer patients resistant to hormone therapy may involve more than the modulation of ER-alpha to reverse Tamoxifen resistance in ERBC cells. RT-qPCR of genes expressed in Tamoxifen resistant cells, trefoil factor 1(TFF1) and v-myc avian myelocytomatosis viral oncogene homolog (MYC), were evaluated along with ESR1 and Diablo as a control. MYC was observed to have increased expression in the treated cells, whereas the other genes had a decrease in their expression levels after the cells were treated for 3 days with Vorinostat IC30 of 1 µM. As for targeting the AR, MCF7 Tamoxifen sensitive and resistant cells were not affected by the AR antagonists to determine an IC50. The cell viability for all MCF7 sub-clones only decreased for high concentrations of 5.56 µM - 50 µM in Bicalutamide and 16.67 µM – 50 µM of MDV1300. Furthermore, hormone depletion of MCF7 G11 Tamoxifen resistant sub-clones did not show a great response to DHT stimulation or the AR antagonists. In the RT-qPCR, the MCF7 G11 cells showed an increase in mRNA expression for ER, AR, and PR after 4 hours of treatment with estradiol. As for the DHT treatment, ER, AR, PR, and PSA had a minimal increase in the fold change, but the fold change in AR was less than in the estradiol treatment. The Mayo Clinic will investigate the possible usage of AR as a biomarker through immunohistochemistry.

The purpose of S.T.O.M.P. is to serve as an additional aid to support a number of adolescent mothers participating in a group-based support program at Chandler Regional Medical Center, but to initiate core values, skills, and social networks for the young mothers. A collection of current literature in support of one-on-one mentorship programs, comprehensive needs as identified by the adolescent pregnant population, personal experience and findings, as well as collaborative discussions amongst health educators has supported the identification of the core objectives of this creative project; the development of a one-on-one mentorship program.

The pathogenesis of type 1 diabetes (T1D) is still not fully understood in the scientific community. Evidence has shown that viral infections are one of the important environmental factors associated with the disease development. Seven of the top T1D related viruses were selected to study the prevalence of viral humoral response in T1D patients using our innovative protein array platform called Nucleic Acid Programmable Protein Array (NAPPA). In this study, each viral gene was individually captured using various PCR based techniques, cloned into a protein expression vector, and assembled as the first version of T1D viral protein array. Humoral responses of IgG, IgA, and IgM were examined. Although each class of immunoglobulin generated a wide-range of reactivity, responses to various viral proteins from different proteins were observed. In summary, we captured most of the T1D related viral genes, established viral protein expression on the protein array, and displayed the serum response on the viral protein array. The successful progress will help to fulfill the long term goal of testing the viral infection hypothesis in T1D development.

Patients receive electroconvulsive therapy every day in the United States but negative media representations of the therapy put doubts in the minds of potential patients as to whether or not they should go through with the therapy or not. The purpose of this study was to explore how health care providers perceive the influence of media on patient decision-making regarding whether or not to receive electroconvulsive therapy (ECT) and how they might address the perceived effects of said media. A total of 16 articles were analyzed in a literature review regarding the media's portrayal of ECT and/or how it affected patients and one interview with a physician was conducted. It was found that while the current literature still largely focuses on the negative media and its implications on patient decision-making, the physician had a more positive viewpoint. As a whole, the literature and physician agreed that media played a role in patients' decisions, but the media's portrayal of the therapy was not seen as negatively by the physician.

The purpose of this study was to explore how the phenomenon of familismo effects behavioral change within the Mexican-American family if one member of the family participates in an obesity prevention curriculum. The qualitative approach findings indicate that the principle of familismo regarding perceived responsibility to provide emotional support to family members regarding changes in physical activity and dietary habits. Participants reported that their families are eating healthier, since, they started the obesity prevention curriculum. The findings regarding physical activity were inconclusive. This study can help nurses, because it emphasizes the importance of promoting family involvement as a motivator for behavioral change, in terms of physical activity and healthy diet eating, within Mexican-American populations.

Family planning educational programs offer a list of artificial contraceptive methods to couples wishing to avoid a pregnancy; however, many of these methods have disadvantages: many lead to negative individual and environmental health outcomes, do not promote a sense of fertility awareness in women and men, may be culturally incompatible with certain religious beliefs and worldviews, and do not take into account the full extent of family planning, which includes the ability to achieve as well as to avoid a pregnancy. Natural Family Planning (NFP) is a true method of family planning in that it offers to the couple the option to achieve as well as avoid a pregnancy. NFP methods holistically approach fertility by taking into account the woman's unique fertility cycle and patterns, the need for the couple to understand complex fertility issues, and the needs and family planning intentions of the couple as a whole. This thesis utilizes Evidence Based Practice in an effort to search the best literature regarding the effectiveness of Natural Family Planning methods to avoid a pregnancy, in comparison to the effectiveness of artificial contraceptive methods to avoid a pregnancy. If effectiveness rates are similar, it is reasonable for the health care profession to consider Natural Family Planning as a valid and reliable family planning method, to move towards further research on its effectiveness and benefits, and to implement a practice change by including it in family planning client education programs.

Protein AMPylation is a recently discovered and relatively unstudied post-translational modification (PTM). AMPylation has previously been shown to play an important role in metabolic regulation and host pathogenesis in bacteria, but the recent identification of potential AMPylators across many species in every domain of life has supported the possibility that AMPylation could be a more fundamental and physiologically significant regulatory PTM. For the first time, we characterized the auto-AMPylation capability of the human protein SOS1 through in vitro AMPylation experiments using full-length protein and whole-domain truncation mutants. We found that SOS1 can become AMPylated at a tyrosine residue possibly within the Cdc25 domain of the protein, the Dbl homology domain is vital for efficient auto-AMPylation activity, and the C-terminal proline-rich domain exhibits a complex regulatory function. The proline-rich domain alone also appears to be capable of catalyzing a separate, unidentified covalent self-modification using a fluorescent ATP analogue. Finally, SOS1 was shown to be capable of catalyzing the AMPylation of two endogenous human protein substrates: a ubiquitous, unidentified protein of ~49kDa and another breast-cancer specific, unidentified protein of ~28kDa.

Abstract
Objective: The purpose of this literature review is to examine the most psychometrically sound (e.g., valid and reliable) instruments measuring physical activity (PA) so that reflection of preschool children’s activity and documentation of intervention effects on preschool child PA is accurate.
Methods: Rigorous validation and calibration studies and those studies designed to test the psychometric properties of PA measurement instruments were specifically sought out to include in this review. Articles were excluded if they did not include specific information about the validity and reliability of the PA measures used with preschool children.
Discussion: Of the six articles reviewed, the systematic review used the most rigorous protocols to conduct its review, yielding the highest level of evidence appraising several validation studies. Because Pfeiffer et al. (2006) utilized the most valid and reliable criterion measure (Cosmed® portable metabolic system), the validation study this research team conducted is deemed to have identified the most valid and reliable cut points to utilize when reducing accelerometry data.
Conclusion: Current cut-points vary widely and greatly affect the reported results of a study. Therefore, it is critical that authors reference validation studies used to support the cut-points that were employed in the data-reduction phase. Currently, validation methods have been identified using high quality criterion measures in rigorous validation studies and thus it is strongly recommended that those cut points be used in data reduction processes.

etworking and a tool for stress coping methods.
With this blog, it is my objective to aid my peers who might need help recognizing and coping with stress by the following methods:
a) Actualize the burden of Stress—Chronic stress is a burden and can be overwhelming if not managed. By disclosing my own stressors, it is my hope that peers will identify with me, so that I can then change the way they view and handle the stress.
b) Discuss the psychological and physical effects of stress on the body—It is my intent to clarify how unmanaged chronic stress can affect the physical and mental health and how acute stress is normal and healthy.
c) Share my coping methods that I have found effective in five minute or less videos with blurbs about how and why they are effective. I believe showing them to you in these mostly raw and unedited videos help maintain the current theme I am going for—keep things as real and raw as possible. Hopefully, these raw videos will help peers visualize working coping methods!
