The goal of my study is to test the overarching hypothesis that art therapy is effective because it targets emotional dysregulation that often accompanies significant health stressors. By reducing the salience of illness-related stressors, art therapy may improve overall mood and recovery, particularly in patients with cancer. After consulting the primary literature and review papers to develop psychological and neural mechanisms at work in art therapy, I created a hypothetical experimental procedure to test these hypotheses to explain why art therapy is helpful to patients with chronic illness. Studies found that art therapy stimulates activity of multiple brain regions involved in memory retrieval and the arousal of emotions. I hypothesize that patients with chronic illness have a reduced capacity for emotion regulation, or difficulty recognizing, expressing or altering illness-related emotions (Gross & Barrett, 2011). Further I hypothesize that art therapy improves mood and therapeutic outcomes by acting on the emotion-processing regions of the limbic system, and thereby facilitating the healthy expression of emotion, emotional processing, and reappraisal. More mechanistically, I propose art therapy reduces the perception or salience of stressors by reducing amygdala activity leading to decreased activation of the hypothalamic-pituitary-adrenal (HPA) axis. The art therapy literature and my hypothesis about its mechanisms of action became the basis of my proposed study. To assess the effectiveness of art therapy in alleviating symptoms of chronic disease, I am specifically targeting patients with cancer who exhibit a lack of emotional regulation. Saliva is collected 3 times a week on the day of intervention: morning after waking, afternoon, and evening. Stress levels are tested using one-hour art therapy sessions over the course of 3 months. The Perceived Stress Scale (PSS) assesses an individual's perceived stress and feelings in past and present situations, for the control and intervention group. To measure improvement in overall mood, 10 one-hour art sessions are performed on patients over 10 weeks. A one-hour discussion analyzing the participants' artwork follows each art session. The Spielberger State-Trait Anxiety Inventory (STAI) assesses overall mood for the intervention and control groups. I created rationale and predictions based on the intended results of each experiment.

While a number of vertebrates, including fishes, salamanders, frogs, and lizards, display regenerative capacity, the process is not necessarily the same. It has been proposed that regeneration, while evolutionarily conserved, has diverged during evolution. However, the extent to which the mechanisms of regeneration have changed between taxa still remains elusive.

While a number of vertebrates, including fishes, salamanders, frogs, and lizards, display regenerative capacity, the process is not necessarily the same. It has been proposed that regeneration, while evolutionarily conserved, has diverged during evolution. However, the extent to which the mechanisms of regeneration have changed between taxa still remains elusive. In the salamander limb, cells dedifferentiate to a more plastic state and aggregate in the distal portion of the appendage to form a blastema, which is responsible for outgrowth and tissue development. In contrast, no such mechanism has been identified in lizards, and it is unclear to what extent evolutionary divergence between amniotes and anamniotes has altered this mechanism. Anolis carolinensis lizards are capable of regenerating their tails after stress-induced autotomy or self-amputation. In this investigation, the distribution of proliferating cells in early A. carolinensis tail regeneration was visualized by immunohistochemistry to examine the location and quantity of proliferating cells. An aggregate of proliferating cells at the distal region of the regenerate is considered indicative of blastema formation. Proliferating cell nuclear antigen (PCNA) and minichromosome maintenance complex component 2 (MCM2) were utilized as proliferation markers. Positive cells were counted for each tail (n=9, n=8 respectively). The percent of proliferating cells at the tip and base of the regenerating tail were compared with a one-way ANOVA statistical test. Both markers showed no significant difference (P=0.585, P=0.603 respectively) indicating absence of a blastema-like structure. These results suggest an alternative mechanism of regeneration in lizards and potentially other amniotes.

The development of skeletal muscle during embryogenesis and repair in adults is dependent on the intricate balance between the proliferation of myogenic progenitor cells and the differentiation of those cells into functional muscle fibers. Recent studies demonstrate that the Drosophila melanogaster transcription factor CG9650 is expressed in muscle progenitor cells, where it maintains myoblast numbers. We are interested in the Mus musculus orthologs Bcl11a and Bcl11b (C2H2 zinc finger transcription factors), and understanding their role as molecular switches that control proliferation/differentiation decisions in muscle progenitor cells. Expression analysis revealed that Bcl11b, but not Bcl11a, is expressed in the region of the mouse embryo populated with myogenic progenitor cells; gene expression studies in muscle cell culture confirmed Bcl11b is also selectively transcribed in muscle. Furthermore, Bcl11b is down-regulated with differentiation, which is consistent with the belief that the gene plays a role in cell proliferation.

Sarcopenia, a disease defined by age-related muscle loss and function, impacts each and every one of us as we age. Medical research over the past 40 years has identified dozens of factors that contribute to Sarcopenia, including, hormonal changes, deficiencies in nutrition, denervation, changes in physical activity and diseases. Developing

Sarcopenia, a disease defined by age-related muscle loss and function, impacts each and every one of us as we age. Medical research over the past 40 years has identified dozens of factors that contribute to Sarcopenia, including, hormonal changes, deficiencies in nutrition, denervation, changes in physical activity and diseases. Developing effective therapeutic treatments for Sarcopenia is dependent on identifying the mechanisms by which these factors affect muscle loss and understanding the interrelationship of these mechanisms. I conducted my research by compiling and analyzing several previous studies on many different mechanisms that contribute to Sarcopenia. Of these mechanisms, I determined the most significant mechanisms and mapped them out on a visual presentation. In addition to the contributing factors listed above, I found that dysregulated cell signaling, mitochondrial abnormalities, impaired autophagy/protein regulation, altered nitric oxide production, and systemic inflammation all contribute to Sarcopenia. Their impact on skeletal muscle is manifested by reduced satellite function, reduced regenerative capacity, loss of muscle mass, accumulation of damaged products, and fibrosis. My research clearly demonstrated that there was not a one-to-one correlation between factors and specific pathological characteristics of Sarcopenia. Instead, factors funneled into a discrete number of cellular processes, including cell proliferation, protein synthesis, and autophagy and apoptosis. Based on my findings, the overall cause of Sarcopenia appears to be a loss of balance between these pathways. The results of my thesis indicate that Sarcopenia is a multifactorial disorder, and therefore, effective therapy should consist of those that prevent necrosis associated with autophagy and apoptosis.

This paper explores the idea of xenophilia and the circumstances under which it may occur. Xenophilia is the preference for an outgroup member over an ingroup member. This preference does not have to be amicable, and in fact can be exploitative under certain circumstances. Previous research indicates that xenophobia is much more common, but a few researchers have found support for the existence of xenophilia. To experimentally test the circumstances under which xenophilia might occur, I conducted a survey-based experiment on Amazon’s Mechanical Turk. This consisted of directed visualizations that manipulated participant goal (self-protection vs. mate acquisition) and the resources offered by both a fictitious outgroup and the hometown ingroup, followed by measures of ingroup/outgroup preference. I hypothesized that when the resource offered by the group addressed the participants’ goal, they would prefer the group with the “matched” resource—even if it was the outgroup providing that resource. My hypothesis was not supported, as the univariate analysis of variance for preference for the outgroup was not significant, F (2, 423) = .723, p = .486. This may have occurred because the goal manipulations were not strong enough to counteract the strong natural preference for ingroup members.

Duchenne Muscular Dystrophy (DMD) is a muscular degenerative disease characterized by striated membrane instability that stimulates continuous cycles of muscle repair. Chronic activation of the innate immune response necessary for muscle repair leads to a pathological accumulation of fibrotic materials that disrupt muscle function. During healthy tissue repair, a balance between pro-inflammatory macrophage (M1) and anti-inflammatory macrophage (M2) promotes clearance of necrotic fibers (myolysis) followed by tissue repair. This is regulated by an intricate feedback loop between muscle, neutrophils and macrophages mediated by Th1 and Th2 cytokines and chemokines. During chronic inflammation, there is an imbalance in an M2 species that produces high levels of extracellular matrix that leads to fibrosis. Finding treatments that ameliorate fibrosis are essential to limiting the muscle pathology that reduces ambulation of DMD patients. Previous studies have shown that Mohawk, (Mkx) a homeobox transcription factor, is essential for the initiation of the inflammation response during acute muscle injury. This study aims to examine whether Mkx regulates inflammation during chronic damage associated with muscular dystrophy. The mdx mouse is a well-studied mouse model that recapitulates muscle necrosis, chronic inflammatory response and fibrosis associated with muscular dystrophy. Utilizing quantitative RT-PCR and histological analysis, the diaphragms and Quadriceps of adult Mkx-/-/mdx and Mkx+/+/mdx mice were analyzed at three critical time points (4 weeks, 3 months and 7 months). In contrast to what was anticipated, there was evidence of increased muscle damage in the absence of Mkx. There was a consistent reduction in the diameter of muscle fibers found in both types of tissue in Mkx-/-/mdx versus Mkx+/+/mdx mice without a difference in the number of fibers with centralized nuclei at 4 weeks and 1 year between the two genotypes, suggesting that the Mkx mutation influences the maturation of fibers forming in response to muscle damage. Fibrosis was higher in the diaphragm of the Mkx-/-/mdx mice at 4 weeks and 3 months, while at1 year there did not appear to be a difference. Overall, the results predict that the absence of Mkx exacerbates the instability of muscle fibers in the mdx mouse. Future studies will be needed to understand the relationship between Mkx and the dystrophin gene.

Hand-coding systems of measuring facial expressions were developed to study and analyze human emotions, but they are time-intensive and thus seldom used. As technology has advanced, new computer software programs, such as Affectiva, were developed to code facial expressions automatically using artificial intelligence and machine learning. Since this technology is still new, Affectiva and its validity remain understudied, and no psychological research has been conducted to compare Affectiva computer coding and hand coding of children’s emotions. The purpose of this study was to compare hand and computer coding of children’s expressions of emotion during a videotaped parent-child interaction. The study answered the following questions: 1) Do hand and computer coding agree?; and 2) Are hand and computer coding in higher agreement for some emotions than others? The sample included 25 pairs of twins from the Arizona Twin Project. Facial expressions were coded from videotape by a trained and reliable human coder and using the software Affectiva. The results showed that hand and computer coded emotion were in agreement for positive, but not negative emotions. Changing the context of the interaction to elicit more negative emotion, and using the same indicators of each emotion in computer and hand coding are suggested to improve the comparison of computer and hand coding.

In vitro gametogenesis (IVG) research has been growing in countries like Japan, US, and China after the development of stem cell research and other scientific advancements as well as because of the perception of infertility as a domestic and international problem. IVG research’s progress has been deliberated internationally, with discussion of questions, challenges, and possibilities that have arisen and may arise in the future as the technology is adopted by different countries. The first section introduces the meaning of IVG, explains the importance of review by scientists and citizens for IVG, and describes a rise in infertility reported in multiple developed countries that could be addressed by IVG. The second section discusses IVG’s applications and implications using 5 ethical categories articulated by Obama’s Presidential Commission for the Study of Bioethical Issues: Public Beneficence, Responsible Stewardship, Intellectual Freedom and Responsibility, Democratic Deliberation, and Justice and Fairness. These five ethical principles were intended for analysis of emerging technologies, and IVG is an emerging technology with possible integration into clinical settings. Among the principles, it seemed that a major weak point of inquiry concerns LGBT+ and disability inclusion, especially of gender dysphoric and transgender people who may experience higher rates of infertility and have a harder time conceiving due to a mix of discrimination, gender dysphoria, and infertility due to hormone replacement therapy (HRT) treatment or gender/sex reassignment surgeries (GRSs/SRSs) that may impair or remove reproductive body parts. A number of other ethical considerations arise about this technology.
