The use of microelectrode arrays (MEA) to electroporate cells is now a reliable way of transfecting RNA interfering substances with high viability and efficiency. However, as the 50-200 micron electrodes are coated with many cells, there are differences in both viability and efficiency between the outside and inside of the electrode. This is due to the field created by the electrode, which has higher intensities toward the outside and lower intensities toward the middle. In order to get the electric field to spread in a more even manner, an "Anodisc" inorganic membrane seeded with cells was placed on the MEA to act as a buffer to the electric fields. One hundred percent transfection efficiency on live cells was found on one sample, though there were problems encountered along the experimental process that introduced error into the results, some of which included the inability for cells to grow to high levels of confluency on the Anodisc as well as the inverted imaging technique used on the opaque disc.

The effect of three different drug modulators on the electrophysiological response of Aplysia neurons was observed through the use of extracellular and intracellular recordings. Extracellular recordings captured the effects of magnesium chloride and glutamate at a variety of concentrations for each. Intracellular recordings displayed the effects of magnesium chloride, glutamate, and GABA for two concentrations each. For extracellular recordings, the average firing rate, average peak-to-peak voltage, average SNR, and sorted units were considered. For intracellular recordings, average firing rate, average peak voltage, and average resting potential were considered. Significance of data could not be determined using statistical analysis due to having a sample size of 1 for every experiment.

It is unknown which regions of the brain are most or least active for golfers during a peak performance state (Flow State or "The Zone") on the putting green. To address this issue, electroencephalographic (EEG) recordings were taken on 10 elite golfers while they performed a putting drill consisting of hitting nine putts spaced uniformly around a hole each five feet away. Data was collected at three time periods, before, during and after the putt. Galvanic Skin Response (GSR) measurements were also recorded on each subject. Three of the subjects performed a visualization of the same putting drill and their brain waves and GSR were recorded and then compared with their actual performance of the drill. EEG data in the Theta (4 \u2014 7 Hz) bandwidth and Alpha (7 \u2014 13 Hz) bandwidth in 11 different locations across the head were analyzed. Relative power spectrum was used to quantify the data. From the results, it was found that there is a higher magnitude of power in both the theta and alpha bandwidths for a missed putt in comparison to a made putt (p<0.05). It was also found that there is a higher average power in the right hemisphere for made putts. There was not a higher power in the occipital region of the brain nor was there a lower power level in the frontal cortical region during made putts. The hypothesis that there would be a difference between the means of the power level in performance compared to visualization techniques was also supported.

Optogenetics presents the ability to control membrane dynamics through the usage of transfected proteins (opsins) and light stimulation. However, as the field continues to grow, the original biological and stimulation tools used have become dated or limited in their uses. The usage of Organic Light Emitting Diodes (OLEDs) in optical stimulation offers greater resolution, finer control of pixel arrays, and the increased functionality of a flexible display at the cost of lower irradiance power density. This study was done to simulate methods using genetic and optical tools towards decreasing the threshold irradiance needed to initiate an action potential in a ChR2 expressing neuron. Simulations show that pulsatile stimulation can decrease threshold irradiances by increasing the overall duration of stimulus while keeping individual pulse durations below 5 ms. Furthermore, the redistribution of Channelrhodopsin-2 (ChR2) to the apical dendrites and a change in wavelength to 625 nm both result in lower threshold irradiances. However, the model used has many discrepancies and has room for improvement in areas such as the light distribution model and ChR2 dynamics. The simulations run with this model however still present valuable insight and knowledge towards the usage of new stimulation methods and revisions on existing protocols.

This thesis is a proposition for an addition to an engineering project that involves creating a heads up display for a scuba diving mask which displays important safety information. The premise of this thesis includes three different features: distress, distance, and direction. The distress feature is to alert a diver that their “buddy diver” is having an emergency and is requiring attention. Distance and direction are intended to be included on the heads up display, informing the diver of the relative location of their “buddy diver” in case they have lost sight of them. A set of requirements was created to find the most practical solutions. From these requirements and extensive research, three different methods of underwater communication were found, but only one, acoustics, was feasible for the scope of this project. Using modems and transducers, an acoustic signal is able to be sent from one diver to another in order to detect relative location as well as send a message of distress. Ultimately, two possible concepts were designed, with one deemed as most advantageous. This concept engages the use of four transponders that have the ability to transmit and receive high frequencies, minimizes blind spots, and is small enough to not cause discomfort or be obstructive to the divers experience.

Battle Royale Games as Esports: Where Fortnite and PUBG Land as Successful Esports

This thesis examines two electronic games, PlayerUnknown’s Battlegrounds (PUBG) and Fortnite, which rose to popularity as battle royale-style videogames, to question whether they are viable candidates for success as esports. In his 2018 Barrett Honors Thesis, Noah Hilliker developed a model for assessing electronic games as successful esports. His thesis, titled “Ambassador Communities, Hybrid Spectatorship, and Other Indicators of Success in eSports” applied his model of indicators of success to League of Legends, Overwatch and Hearthstone. In this thesis, I have applied his indicators to (PUBG) and Fortnite, which differ significantly from the games he previously analyzed, precisely as I argue here, because they are battle royale games. These indicators fall under three categories: the nature of fans, the overall design of the game and the relationship between game developers and fans. The data that was examined for this investigation included academic literature on esports, posts on online platforms, news articles, YouTube content and forums. The following indicators were analyzed in their application to the collected data for both games: self-organization, spectator-player hybridity, ambassadors, clear center of action, demonstration of skill, mechanical variance, game balance, character of fan/developer relationship and feedback to fans. Through this project, I apply a previous approach to analyzing esports success to two games, Fortnite and PUBG, in order to analyze how well the model fits battle royale-style games.

Effects of rooftop photovoltaics on building cooling demand and ambient air

Photovoltaic panels are commonly used for their versatility in on-site generation of clean electricity in urban environments, specifically on rooftops. However, their implementation on rooftops poses potential (positive and negative) impacts on the energy use of buildings, and urban climates. The negative impacts are compounded if PV is installed on top of a high-albedo rooftop. This study quantitively investigates these impacts from PV installation on top of a building with a white roof in Phoenix, AZ. We supplemented our measurements with EnergyPlus simulations to model the energy implications for archetypical residential and retail buildings and calculated the energy penalty to generation ratio as well as sensible heat flux for each combination of panel height and building type. Results indicate that the daily cooling energy penalty to due blockage of outgoing longwave radiation can be 4.9—11.2% of the PV generation. In addition, while we observed a small decrease in nighttime sensible heat flux to the ambient, PV cases increased the daytime heat flux by more than a factor of 10. This study highlights the potential unintended consequences of rooftop PV under certain conditions and provides a broader perspective for building designers and urban planners.

International refugees have continuously shaped the identity of modern London, England, creating a diverse cityscape. However, the referendum in June 2016 indicated a perceived desire of the majority of United Kingdom (UK) citizens to leave the European Union (EU) and the domination of far-right, anti-immigrant rhetoric in British politics. These elements have given rise to the question of how refugees will find belonging in a geographical space that continues to create borders at both a national and borough level. As the Brexit vote still stands, barriers to applying for refugee status and successful resettlement could increase - complicating the lives of refugees wanting to resettle in the UK. Urban spaces such as London, Manchester, and Birmingham have transformed into places where the lives of the British cross daily with the lives of those forcibly removed from their home state. With minimal current research on the relationship between international refugees in London and the current social and political identity of the city, post-Brexit vote, I believe there is a gap in understanding to be filled. This gap includes defining the relationship between place, people, and politics in the context of the city of London as well as the boroughs that comprise the city. In addition, this research explores the future of London as a place at a borough-level and aims to understand how the idea of borders and nationalism have been uncovered and subsequently amplified through the referendum. The following paper includes data collected from British refugee agencies and inhabitants of five London boroughs that will add to existing research in the form of academic and professional journals and published reports produced by refugee agencies and the British government in hopes to identify the current nature of the relationship between international refugees and Londoners and how this relationship might shift in the future.
