For my senior thesis, I explored the topic of toxic masculinity in America, specifically on college campuses and cultivated through aspects of Greek Life. My exploration of the topic culminated in an art showcase that was intended to create a space for candid dialogue about the impact and effects of a persistent culture of toxic masculinity perpetuated through Greek Life. It is important to note that I do not believe that masculinity in and of itself is toxic; rather, often the ways in which it is conceptualized and enacted is harmful to men, women, and society as a whole. Thus, my goal is not to disparage men or masculinity, but rather to open a dialogue exploring how Greek Life reproduces a culture that pressures men to conform to a rigid and narrow conception of masculinity.

The idea for this thesis emerged from my senior design capstone project, A Wearable Threat Awareness System. A TFmini-S LiDAR sensor is used as one component of this system; the functionality of and signal processing behind this type of sensor are elucidated in this document. Conceptual implementations of the optical and digital stages of the signal processing is described in some detail. Following an introduction in which some general background knowledge about LiDAR is set forth, the body of the thesis is organized into two main sections. The first section focuses on optical processing to demodulate the received signal backscattered from the target object. This section describes the key steps in demodulation and illustrates them with computer simulation. A series of graphs capture the mathematical form of the signal as it progresses through the optical processing stages, ultimately yielding the baseband envelope which is converted to digital form for estimation of the leading edge of the pulse waveform using a digital algorithm. The next section is on range estimation. It describes the digital algorithm designed to estimate the arrival time of the leading edge of the optical pulse signal. This enables the pulse’s time of flight to be estimated, thus determining the distance between the LiDAR and the target. Performance of this algorithm is assessed with four different levels of noise. A calculation of the error in the leading-edge detection in terms of distance is also included to provide more insight into the algorithm’s accuracy.

Speculative fiction and fantasy media have abundant power to portray alternative realities or imagined futures for minority groups. Buffy the Vampire Slayer, from the late 1990s-early 2000s, and Wynonna Earp, from the late 2010s, are two fantasy television shows that produce this often-progressive, impactful representation, particularly for lesbians and bisexual people. Drawing on Queer and Monster Theories from Susan Stryker, Marilee Lindemann, Harry Benshoff, and Alexis Lothian, this thesis examines queer representation in these TV shows and how it contributes to the normalization of LGBTQ+ individuals whilst simultaneously honoring the shows’ queer fans. Normalizing non-cishetero genders and sexualities helps rewrite the narrative of LGBTQ+ people being considered “deviant” and threatening societal order; and holding true to queer roots of challenging social norms prevents the power of the queer community from being influenced by the pressures of compulsory heterosexuality.

A Skunkworks project is the name given to a small team of individuals leading an innovative undertaking, and conducting research and development outside of the normal scope of an organization. With this concept in mind, our team of six individuals was tasked with finding and conceptualizing innovative solutions within varying business markets of interest. Our team started off with five markets that we identified issues in and were passionate about solving. These included Sports Engagement, Education, Student Debt, Digital Literacy, and Viral Health. From extensive research, trial and error, and endless conversations we settled on creating business models in two final areas: Student Debt and Viral Health. Our research in Student Debt led us to the discovery that the average Arizona State student, takes out $21,237 in loans for their four year degree and in the whole state of Arizona, a student takes on an average of $22,253. Our solution to this problem was to create a student financial app that served as an efficient debt tracker that provided important information about finances, investing, and student loan information. Additionally, our team also wanted the address the issue of sexually transmitted diseases, just a small scope of Viral Health, within Arizona State University. Our research led us to discover that 50% of people report not getting tested, and from this population most reported it was due to anxiety and financial issues. From our research the StayInformed app was created to provide students with better accessibility to both at-home and clinic testing services, and updated education on sexual health. With this project model we hope to increase the rate of students testing and allow students more agency over their sexual health. Although these two services are addressing very different markets, they both utilize forward thinking technology to create much needed solutions and better the lives of students.

The 5-year survival rate for late-stage metastatic melanoma is only ~30%. The 5-year survival rate for late-stage metastatic melanoma is only ~30%. A major reason for this low survival rate is that one of the most commonly mutated genes in melanoma, NRAS, has no FDA-approved targeted therapies. Because the RAS protein does not have any targeted therapies, patients with RAS mutant tumors have an ongoing need for treatments that indirectly target RAS. This thesis project aims to identify expression and phosphorylation levels of proteins downstream of RAS in melanoma cell lines with the most common driver mutations. By analyzing the protein-level differences between these genetic mutants, we hope to identify additional indirect RAS protein targets for the treatment of NRAS mutant melanoma. RAS has several downstream effector proteins involved in oncogenic signaling pathways including FAK, Paxillin, AKT, and ERK. 5 melanoma cell lines (2 BRAF mutant, 2 NRAS mutant, and 1 designated wildtype) were analyzed using western bloting for FAK, Paxillin, AKT, and ERK phosphorylation and total expression levels. The results of western blot analysis showed that NRAS mutant cell lines had increased expression of phosphorylated Paxillin. Increased Paxillin phosphorylation corresponds to increased Paxillin binding at the FAT domain of FAK. Therefore, cell lines with increased FAK FAT – Paxillin interaction would be more sensitive to FAK FAT domain inhibition. The data presented provide an an explanation for the reduction in cell viability in NRAS mutant cell lines infected with Ad-FRNK. This information also has significant clinical relevance as researchers work to develop synthetic FAK FAT domain inhibitors, such as cyclic peptides. Additionally, cell lines with high levels of phosphorylated AKT showed a significant reduction in the amount of phosphorylated ERK. The identification of this inverse relationship may help to explain why BRAF and NRAS mutations are mutually exclusive. To conclude, NRAS mutant cell lines have increased expression of phosphorylated Paxillin and AKT which may explain why NRAS mutant cell lines are more sensitive to FAK FAT domain inhibition.
