Every day we pass people without thinking everyone has a story. If an individual looks “normal,” any struggles faced living with an invisible disability are left without words or thoughts due to the dominant norm—ableism. Conversely, a more visible disability may not be dismissed as quickly. Who are unseen, ignored, and misunderstood are those who live with invisible disabilities not only in a dominant able-bodied society, but also within academic scholarship as well, because they do not fit into the dominant definition of disability. In turn, binaries form between power relations and within knowledge production that create exclusion. This thesis is an intersectional analysis on expanding the definition of disability, specifically invisible disability, in order to deconstruct, challenge, and transform the hegemonic conceptualization of disability and break binaries in order to give voice to ignored and misunderstood narratives of invisible disabilities as well as foster and create nuanced understanding within knowledge production and power itself. I particularly use an autoethnographic approach to conduct this analysis of my own everyday, lived experience as a young, mixed race woman living with an invisible disability, or chronic illness, on how ableism operates in the medical sphere and at the academy, further exploring what it means to be a “good” or “bad” chronic illness patient and categorized and labeled by the stigmas attached to the definition of disability.

The purpose of this study was to examine the overall maintenance of behavior during the 12 to 24 month period of the ​Stand&Move@Work​ study and the impact of implementation factors (i.e., facilitators, advocate activity, and the amount of strategies used) on behavior change. The design of the study was a cluster randomized trial which was facilitated by researchers for the first 12 months of the study. The primary aim of the study was to examine the maintenance of behavior change (i.e., sitting time) at the 12 month and 24 month marks using objectively measured sedentary behavior (activPAL micro). The secondary aim of the study was to examine the impact of implementation factors (i.e., facilitators, advocate activity, and the amount of strategies used) on behavior change during the 12 through 24 months maintenance period. Participants (N=630) included full-time, caucasian, middle-aged office workers. For the primary aim, descriptive means were used to cluster for observations within-persons and were adjusted for age, gender, race, job-type, and ordering effects.. For the secondary aim, descriptive means adjusted for workplace culture and environment were computed. At the 24 month mark, participants spent 280.67 ± 87.67 min/8hr workday sitting and 161.94 ± 85.87 min/8hr workday standing. The top performing worksites displayed reductions in sitting time which largely translated into standing time by about 30 minutes per 8 hour workday at 24 months. Feasibility findings indicated that implementation strategies do not show differences between the top 25% and bottom 25% performing worksites. This study provides insight to implementation strategies for interventions in the workplace.

Past studies have shown that auditory feedback plays an important role in maintaining the speech production system. Typically, speakers compensate for auditory feedback alterations when the alterations persist over time (auditory motor adaptation). Our study focused on how to increase the rate of adaptation by using different auditory feedback conditions.

Past studies have shown that auditory feedback plays an important role in maintaining the speech production system. Typically, speakers compensate for auditory feedback alterations when the alterations persist over time (auditory motor adaptation). Our study focused on how to increase the rate of adaptation by using different auditory feedback conditions. For the present study, we recruited a total of 30 participants. We examined auditory motor adaptation after participants completed three conditions: Normal speaking, noise-masked speaking, and silent reading. The normal condition was used as a control condition. In the noise-masked condition, noise was added to the auditory feedback to completely mask speech outputs. In the silent reading condition, participants were instructed to silently read target words in their heads, then read the words out loud. We found that the learning rate in the noise-masked condition was lower than that in the normal condition. In contrast, participants adapted at a faster rate after they experience the silent reading condition. Overall, this study demonstrated that adaptation rate can be modified through pre-exposing participants to different types auditory-motor manipulations.

To date, there have been few, if any, studies evaluating the venom toxin levels in dogs that have been naturally envenomated by pit vipers. Understanding venom toxin pharmacokinetics in a clinical setting is important for a variety of reasons, including the potential to better elucidate treatment options, prognosis, and other factors associated with pit viper envenomation. In addition, dogs serve as a comparative species to humans for evaluating pit viper envenomations. This pilot study’s primary objective was to address the question of “What do we see?” in dogs presenting for rattlesnake envenomation. To answer this question, we obtained serum from envenomated dogs presenting at three veterinary clinics, then used enzyme-linked immunosorbent assay (ELISA) and western blot analysis to measure total venom and key toxins in sera. Phospholipase A2, a primary venom toxin, was identified in a few samples by the western blot, and contributed to the positive correlation between percent echinocytes in the blood and venom concentration. Medical data records were compared to venom concentrations measured using ELISA to determine whether there were any significant correlations. First, the hematological results were compared. Clotting times showed a strong positive correlation, clotting times and platelets showed a negative correlation, while echinocytes and platelets showed no correlation. When compared to venom concentration, clotting times showed a negative correlation, while age showed a positive correlation. Weight and platelets were also compared to venom concentration, but no significant correlations were found. The logistics of this study provided a real-world model where time elapsed between envenomation and hospital admission, thus giving a realistic look at what occurs in both animal and human medicine.

Oncolytic virotherapy (OV) is the use of viruses that do not target normal human cells to infect and destroy cancer cells; some also stimulate the immune system against the tumors. Myxoma virus (VMYX) is a candidate for use as an oncolytic agent, as it is not pathogenic to humans and can infect a variety of human cancer cells. VMYX also can initiate immune responses against the virus-infected tumor. Thus, we investigated the oncolytic efficacy of a few recombinant constructs of VMYX on triple-negative breast cancer (TNBC), a highly aggressive subtype of breast cancer with limited treatment options. TNBC lacks an estrogen receptor, progesterone receptor, and HER2, which render hormone-based therapies useless. Further challenges of TNBC include early metastasis and recurrence, as well as poor prognosis due to a lack of molecular targets. Here, we utilized 4T1-Luc2 cells, an in vitro mouse model of TNBC, to examine the oncolytic potential of recombinant viral constructs of VMYX. Ability to infect was measured by fluorescence intensity, while ability to induce cytotoxicity was measured through MTS and SYTOX assays. Further characterization of cell death was performed using Caspase 3/7 activity assay, immunofluorescent staining and confocal microscopy to detect ecto-expression of calreticulin, and ATP release assays. We demonstrated the ability of recombinant VMYX constructs to infect and induce cell death in 4T1-Luc2 cells. VMYX-p14-FAST-GFP induced the most cell death, while VMYX-M011LKO-GFP best activated Caspase 3/7. Through ATP release assays and examination of ecto-expression of calreticulin, preliminary data indicated that VYX-135KO-GFP is unable to stimulate immunogenic cell death, a form of cell death that stimulates an adaptive immune response, in these cells. Future directions include further characterization of cell death in vitro, as well as in vivo studies.

Checklists have become a vital aspect of aviation, regardless of skill level. From general aviation pilots going through flight training to commercial airline pilots responsible for hundreds of lives every day, checklists are used from the moment you step into the cockpit until the last light that is turned off at the end of the flight. Checklists are such a significant part of aviation, and several different ways to run a checklist have been created (such as the challenge-response and do-and-tell methods). Despite these variations in checklist usage and procedures, all methods are restricted in terms of user involvement; in other words, pilots are not easily engaged or invested in the checklists that they use in day to day operations. Theorized through exposure to this issue as a student and as a Certified Flight Instructor, Know Go™ has been created as a long term tool to replace conventional checklists with a resourceful one that acts as both a normal checklist for daily use and a learning tool for long term retention. The purpose of this text is to introduce the capabilities of the application, as well as discuss the theories behind the effectiveness of the application. The developmental processes and the challenges associated with application production will also be analyzed.

Bexarotene is a synthetic analog of 9-cis-retinoic acid and ligand for the retinoid X receptor which has a history of clinical success in the treatment of T-cell lymphoma. Bexarotene has also shown potential for treating a variety of other cancers, which we seek to explore in this project. The potential of bexarotene lies in its unique mechanisms and wide application, however, it has shown limited effectiveness thus far in the treatment of breast and lung cancer, with moderate levels of efficacy and symptoms such as cutaneous toxicity, hyperlipidemia, and hypothyroidism. For this project several analogs of bexarotene were synthesized with the intentions of making a more potent ligand that can be used to treat these carcinomas while minimizing harmful side effects. We were successful in synthesizing a large variety of analogs over the span of roughly two years, including iso-chroman derivatives of bexarotene and NEt-TMN, in addition to a new series of analogs of the reported NEt-TMN derivative. These analogs were analyzed via melting point determination and nuclear magnetic resonance (NMR) spectroscopy to confirm the molecular structure and determine purity, and it is our intent to continue with further testing of these compounds to determine their effectiveness as well as the side effects they are likely to cause with levels of toxicity. Recent studies suggest that continuing the analysis of these compounds and other rexinoids like the ones described herein is a worthwhile endeavor as similar rexinoids have shown in numerous assays to be more potent and less toxic in the treatment of cancers when compared with bexarotene.

Titanium dioxide is an essential material under research for energy and environmental applications, chiefly through its photocatalytic properties. These properties allow it to be used for water-splitting, detoxification, and photovoltaics, in addition to its conventional uses in pigmentation and sunscreen. Titanium dioxide exists in several polymorphic structures, of which the most common are rutile and anatase. We focused on anatase for the purposes of this research, due to its promising results for hydrolysis.

Anatase exists often in its reduced form (TiO2-x), enabling it to perform redox reactions through the absorption and release of oxygen into/from the crystal lattice. These processes result in structural changes, induced by defects in the material, which can theoretically be observed using advanced characterization methods. In situ electron microscopy is one of such methods, and can provide a window into these structural changes. However, in order to interpret the structural evolution caused by defects in materials, it is often necessary and pertinent to use atomistic simulations to compare the experimental images with models.

In this thesis project, we modeled the defect structures in anatase, around oxygen vacancies and at surfaces, using molecular dynamics, benchmarked with density functional theory. Using a “reactive” forcefield designed for the simulation of interactions between anatase and water that can model and treat bonding through the use of bond orders, different vacancy structures were analyzed and simulated. To compare these theoretical, generated models with experimental data, the “multislice approach” to TEM image simulation was used. We investigated a series of different vacancy configurations and surfaces and generated fingerprints for comparison with TEM experiments. This comparison demonstrated a proof of concept for a technique suggesting the possibility for the identification of oxygen vacancy structures directly from TEM images. This research aims to improve our atomic-level understanding of oxide materials, by providing a methodology for the analysis of vacancy formation from very subtle phenomena in TEM images.

Analog to Digital Converters (ADCs) are a critical component in modern circuit applications. ADCs are used in virtually every application in which a digital circuit is interacting with data from the real world, ranging from commercial applications to crucial military and aerospace applications, and are especially important when interacting with sensors that observe environmental factors. Due to the critical nature of these converters, as well as the vast range of environments in which they are used, it is important that they accurately sample data regardless of environmental factors. These environmental factors range from input noise and power supply variations to temperature and radiation, and it is important to know how each may affect the accuracy of the resulting data when designing circuits that depend upon the data from these ADCs. These environmental factors are considered hostile environments, as they each generally have a negative effect on the operation of an ADC. This thesis seeks to investigate the effects of several of these hostile environmental variables on the performance of analog to digital converters. Three different analog to digital converters with similar specifications were selected and analyzed under common hostile environments. Data was collected on multiple copies of an ADC and averaged together to analyze the results using multiple characteristics of converter performance. Performance metrics were obtained across a range of frequencies, input noise, input signal offsets, power supply voltages, and temperatures. The obtained results showed a clear decrease in performance farther from a room temperature environment, but the results for several other environmental variables showed either no significant correlation or resulted in inconclusive data.

Abstract
This study investigates the effects of Bharatanatyam dance on stress, mood, and anxiety. I have danced Bharatanatyam since I was 8 years old, it has offered me a way to release stress and anxiety. This study provides empirical data to support the claim that Bharatanatyam has therapeutic effects that release stress and reduce anxiety. This investigation was conducted through self-reports and interviews. A Positive Affect and Negative Affect Schedule (PANAS) scale was used to determine positive and negative effects. The average positive affect during the “dance weeks” (DW) was 46.6 and the average negative affect was 12.2. During the “no dance weeks” (NDW), the average positive effect was 23.7 and the average negative affect was 31. The participant’s interview PANAS results had an average positive effect of 39.8 and an average negative effect of 12.8. Analyzing the self-report journaling highlighted a more prevalent use of positive words during the DW and a more significant use of negative words during the NDW. The Bharatanatyam dancers who were probed to enter post-performance environment for an interview also used positive words to describe Bharatanatyam dancing. In conclusion, practicing Bharatanatyam had an overall positive effect on mood, and can reduce stress and anxiety.
