ABSTRACT

This study intended to provide people diagnosed with autism spectrum disorder a creative outlet to experience dance and other art forms as a way of expressing themselves. Other potential benefits were observed throughout the exploration, including social interaction, coordination, and confidence. An interpretive phenomenological research model analyzed participant and parent verbal reflections, written feedback, and video recorded movement sessions to understand and interpret the participant's experience and the potential value of creative movement. The study was conducted over a seven-week period, which included 13, 30-minute movement sessions held biweekly along with interviews, discussions, surveys, and journaling. The research revealed dance empowered each participant to explore his/her creativity and exercise personal expression. The feedback received from the participants and parents through interviews and reflections revealed the participants did exercise and discover social, physical, emotional, and creative expression throughout the study.

The life of Jean-Michel Basquiat is often misinterpreted in artistic discourse. From a social justice perspective, Basquiat's work is not merely art. Despite the symbolism and subject matter open for analysis, Basquiat articulated the self in relation to nuances of race, socio-economy, and historical scripts based upon real relations and conditions. Of the genre of Neo-Expressionism without a disciplined schooling in art, Jean-Michel is categorized as 'primitive' in style and form, labeled the "first black artist." Beyond the art world's possessive confines and according to post-colonial aesthetics, Jean-Michel articulates the existence of a learning self. With a pedagogical lens, a process of becoming an "artist" deepened Basquiat's expressions of self in relation to a “white” art world, which typically restricted the artist to specific categories and definitional parameters.

While recognition of the "artist" highlights the limitations of 'public' and 'self' in pedagogy, learning of the self through Neo-Expressionism is contingent upon articulating a situated existence among particular "publics," with regard to time and place. Variable dimensions of recognition create a fragmented self with transitional 'stages' and a series of acute shifts re-establish the definitional boundaries of art, definers, and ultimately the self and “Other”. These shifts continuously create new margins of the avant-garde and the self is redefined by art and discourse to sustain capital inflow, thereby replicating the colonial nature of capitalism with regard to communication, material and discovery, and “Other”. The process eschews a realized finality while expression as a relational communication of the situated persona redefines one's identity and demarcates a value of the self.

Art is a form of spiritual expression that is thriving in many Indigenous cultures. It can take many forms, meanings and have a multitude of emotional, mental, physical and spiritual effects on its creator as well as its audience. Amongst American Indians, art has been a method for maintaining holistic well-being intended to heal and cope with traumatic experiences. In this thesis, I examine the western societal and cultural influences that have led to the loss of cultural identity and examine approaches and practices that aim to re-establish a resilient connection to identity and well-being using art as a spiritual catalyst. Literary research and articles were reviewed related to the issue of art as a form of spiritual expression in Indigenous cultures. An autoethnography was conducted with the intent to record and reflect on the well-being of the researcher in relation to her artistic expression. Journaling and vlogging were used as research methods and painting, sketching, and beading was used as artistic methods. Over the course of six months, over 50 videos with 30 hours of raw footage were recorded; averaging 2 hours per day. The results are reflected in the researchers free-flowing and emotionally driven reflection of experiences that have driven her artwork. This thesis supports the establishment of art as a form of spiritual expression for transforming the current western focused health care paradigm to one that recognizes, values and employs Indigenous insight, methodologies, worldviews, culture and spirituality.

Recently, the Redding and Jones labs created a microbial photoelectrosynthesis cell that utilized these properties to produce molecular hydrogen using electrons provided by a cathode via a chemical mediator. Although this light-driven

Heliobacterium modesticaldum (H. modesticaldum) is an anaerobic photoheterotroph that can fix nitrogen (N2) and produce molecular hydrogen (H2). Recently, the Redding and Jones labs created a microbial photoelectrosynthesis cell that utilized these properties to produce molecular hydrogen using electrons provided by a cathode via a chemical mediator. Although this light-driven creation of fuel within a microbial electrochemical cell was the first of its kind, its production rate of hydrogen was low. It was hypothesized that the injection of electrons into H. modesticaldum was a rate-limiting step in H2 production. Within the H. modesticaldum genome, there is a gene (HM1_0653) that encodes a multi-heme cytochrome c that may be directly involved in this step. From past transcriptomic experiments, this gene is known to be very poorly expressed in H. modesticaldum. Our hypothesis was that increasing its expression with a strong promoter could result in faster electron transfer, and thus, increased H2 production in the photoelectrosynthesis cell. In order to test this hypothesis, different promoters that could lead to high expression in H. modesticaldum were included with a copy of HM1_0653 in various plasmid constructs that were first cloned into E. coli before being conjugated with H. modesticaldum. Cloning in E. coli was possible with the newly derived transformation system and by reducing the copy-number of the vector system. When overexpressed in E. coli, the protein appeared to be expressed, but its purification proved to be difficult. Moreover, conjugation with H. modesticaldum was not achieved. Our results are consistent with the idea that high level overexpression in H. modesticaldum was toxic. An inducible promoter may circumvent these issues and prove more successful in future experiments.

This correlational study investigated how adolescent's artistic style changed depending on their level of interest in art. The participants were 28 students from the 7th and 8th grade of Zuni Hills Elementary School. A survey was administered which measured, in regards to their artwork, the categories of interest, experimentation in subject, experimentation in material, abstract style, literal style, imagination, identity, emotion and storytelling. It was predicted that interest would have a significant positive relationship with abstract style and experimentation with subject. It was also predicted that interest would have a significant negative relationship with literal style and experimentation with material. Scores for all categories were compared and significant positive relations were found in regards to emotion, identity and storytelling. There was also a significant positive realtionship between interest and imagination as well as interest and emotion. These findings add to research about motivation in adolescent art through the means of expression of the self.

Heliobacterium modesticaldum (H. modesticaldum) is an anaerobic photoheterotroph that can produce molecular hydrogen (H2) when it is fixing dinitrogen (N2). In addition, electrons can be injected into this organism via an electrode and redox mediator in a light-dependent fashion, as shown recently by the Redding and Jones research groups. These factors make H. modesticaldum an ideal organism for use in a microbial photoelectrosynthesis cell, in which electricity can be used to power specific metabolic processes that produce a desired compound (e.g. H2). However, the injection of electrons into this organism is not optimal, which may limit the H2 production rate. There is a gene (HM1_0653) in the genome encoding a multi-heme cytochrome c that is similar to the proteins known to be used for exit of electrons in the well- known electrode-respiring bacteria (e.g. Geobacteria). RNA-sequencing in the Redding lab has shown that the HM1_0653 gene is very poorly expressed in H. modesticaldum. Boosting expression of this cytochrome could lead to faster electron transfer into the cells and thereby more H2 production via photoelectrosynthesis. In order to gain a deeper understanding of this protein, it was expressed in E.coli by two different versions: (1) the entire gene and (2) a truncated gene with an additional hexahistidine tag (truncHM1_0653). Both cultures had a pink color, indicating the biosynthesis of cytochrome. It was discovered that the HM1_0653 protein was likely released into the medium and shows the most promise for ease of purification of HM1_0653. Furthermore, we explored protein expression in H. modesticaldum using the current transformation system in the Redding Lab, but the combination of gene toxicity and copy number of the vector resulted in cloning difficulties in E.coli. An alternative vector may prove more successful.

Yet much remains unknown regarding

MicroRNAs (miRNAs) are 17-22 nucleotide non-coding RNAs that regulate gene expression by targeting non-complementary elements in the 3’ untranslated regions (3’UTRs) of mRNAs. miRNAs, which form complex networks of interaction that differ by tissue and developmental stage, display conservation in their function across metazoan species. Yet much remains unknown regarding their biogenesis, localization, strand selection, and their absolute abundance due to the difficulty of detecting and amplifying such small molecules. Here, I used an updated HT qPCR-based methodology to follow miRNA expression of 5p and 3p strands for all 190 C. elegans miRNAs described in miRBase throughout all six developmental stages in triplicates (total of 9,708 experiments), and studied their expression levels, tissue localization, and the rules underlying miRNA strand selection. My study validated previous findings and identified novel, conserved patterns of miRNA strand expression throughout C. elegans development, which at times correlate with previously observed developmental phenotypes. Additionally, my results highlighted novel structural principles underlying strand selection, which can be applied to higher metazoans. Though optimized for use in C. elegans, this method can be easily adapted to other eukaryotic systems, allowing for more scalable quantitative investigation of miRNA biology and/or miRNA diagnostics.

In this study, the role of attention in facial expression processing is investigated, especially as it relates to fearful facial expressions compared to happy facial expressions. Facial fear processing plays a critical role in human social interactions and survival, and this has previously been studied mainly in animal models. This study, however, was accomplished with the presentation of images of actors with happy and fearful facial expressions in three spatial frequency formats, as it is hypothesized that images at different spatial frequencies may be processed via different pathways. These images were presented to human participants in two experiments. In Experiment I, facial expression was task-relevant as participants were asked to discriminate between “happy” and “fear” expressions with reaction time (measured in seconds) and accuracy recorded. In Experiment II, facial expression was task-irrelevant, as participants were asked simply to discriminate between photographs of males and females, again with reaction time and accuracy recorded. Overall, the results comparing happy and fearful facial expressions in Experiment I were not significant. The results comparing happy and fearful facial expressions in Experiment II exhibited similar insignificant results except for accuracy in certain spatial frequencies, which were found to be significant. These results suggest that fearful facial expressions are processed more accurately than happy facial expressions when attention is focused on other variables in the image rather than when attention is focused on the facial expressions themselves.

The purpose of this project was to create a workshop for adults without dance experience to immerse them in dance as a form of therapy. The goals of this workshop are to foster physical awareness and physical creativity. There are four main areas the participants would get experience in from this workshop including self-care/awareness, expression, gesture and choreography. These four areas are the ways that the two main goals manifest themselves. For this project I reviewed research by many different professionals from the therapy, psychology, self-care, dance, and dance therapy fields. Their studies as well as my own experience aided in putting together my themes for the workshop and class activities. I include my methodology, lesson plans, a workbook, and my reflection on the process.

Artistic expression can be made more accessible through the use of technological interfaces such as auditory analysis, generative artificial intelligence models, and simplification of complicated systems, providing a way for human driven creativity to serve as an input that allow users to creatively express themselves. Studies and testing were done with industry standard performance technology and protocols to create an accessible interface for creative expression. Artificial intelligence models were created to generate art based on simple text inputs. Users were then invited to display their creativity using the software, and a comprehensive performance showcased the potential of the system for artistic expression.
