Chronic restraint stress impairs hippocampal-mediated spatial learning and memory, which improves following a post-stress recovery period. Here, we investigated whether brain derived neurotrophic factor (BDNF), a protein important for hippocampal function, would alter the recovery from chronic stress-induced spatial memory deficits. Adult male Sprague-Dawley rats were infused into the hippocampus with adeno- associated viral vectors containing the coding sequence for short interfering (si)RNA directed against BDNF or a scrambled sequence (Scr), with both containing the coding information for green fluorescent protein to aid in anatomical localization. Rats were then chronically restrained (wire mesh, 6h/d/21d) and assessed for spatial learning and memory using a radial arm water maze (RAWM) either immediately after stressor cessation (Str-Imm) or following a 21-day post-stress recovery period (Str-Rec). All groups learned the RAWM task similarly, but differed on the memory retention trial. Rats in the Str-Imm group, regardless of viral vector contents, committed more errors in the spatial reference memory domain than did non-stressed controls. Importantly, the typical improvement in spatial memory following recovery from chronic stress was blocked with the siRNA against BDNF, as Str-Rec-siRNA performed worse on the RAWM compared to the non-stressed controls or Str-Rec-Scr. These effects were specific for the reference memory domain as repeated entry errors that reflect spatial working memory were unaffected by stress condition or viral vector contents. These results demonstrate that hippocampal BDNF is necessary for the recovery from stress-induced hippocampal dependent spatial memory deficits in the reference memory domain.

In developed countries, municipalities deliver drinking water to constituents through water distribution systems. These transport water from a treatment plant to homes, restaurants, and any other site of end use. Proper water distribution system infrastructure functionality is a critical concern to city planners and managers because component failures within these systems restrict or prevent the ability to deliver water. The reduced capacity to deliver water forces the health and well being of all citizens into jeopardy. The breakdown of a component can even spark the failure of several more components, causing a sequence of cascading failures with catastrophic consequences. To make matters worse, some forms of component failures are unpredictable and it is impossible to foresee every possible failure that could occur. In order to prevent cataclysmic losses that are experienced during system failures, the development of resilient water distribution infrastructure is vital. A resilient water distribution system possesses an adaptive capacity to mitigate the loss of service resulting from component failures. Traditionally, infrastructure resilience research has been retrospective in nature, analyzing the infrastructure system after it suffered a failure event. However, this research project takes water distribution resilience research in a new direction. The research identifies the Sensing Anticipating, Adaptation, and Learning processes that are inherent in the current operations of each component in the water distribution system (pumps, pipes, valves, tanks, nodes). Additional SAAL processes have been recommended for the components that lack adaptive management in current practice. This workis unique in that it applies resilience theory to water distribution systems in an anticipatory manner. This anticipatory application of resilience will provide operators with actionable process for them to implement during failure situations. In this setting, resilience is applied to existing systems for noticeable improvements in operation during failure situations.

Abstract Communication scholars have begun to contribute to the theoretical understanding of resilience as a dynamic and collaborative process, which can be talked into being (Buzzanell, 2010). Previous research has examined the role of resilience in romantic couples, however, has focused disproportionately on heterosexual couples. This offers a limited, and singular understanding of how resilience is developed and sustained in romantic relationships. To better understand the scope and breadth of resilience, this study examined five same-sex couples through an in-depth qualitative case study analysis. The purpose of this study was to contribute to the small body of existing data, as well as, enhance our understanding of how resilience works in other contexts. Data was analyzed for thematic patterns, and compared to existing data on same-sex relationships. The findings supported that resilience is a collaborative process that is facilitated by communication. There were some discrepancies from the data collected in this study compared to previous findings; however, due to the small sample size, findings from this study cannot be generalized to the larger population.

This paper will use a national cross-sectional survey approach to look at the association between trainable mind-body qualities (mindfulness and self-compassion) with well-being and resilience in 111 college students across the U.S.. Specifically, it will investigate (1) the relationship between trainable qualities (mindfulness, self-compassion) and the resilience, and subjective well-being in students, and (2) compare how these variables were distributed based on enrollment in a college course on compassion. After examination of descriptive statistics and Pearson correlations, comparative analyses were also employed to determine whether enrollment in compassion college courses had any relationship to one’s scores. Results: Respondents included 12 students enrolled in Compassion college course, and 99 students who were not. Both mindfulness and self-compassion showed significant positive correlations with well-being and resilience in all students, and in subgroups based on enrollment at p < .01. Additionally, students enrolled in the course averaged 3 points higher scores across all measures except resilience, where scores were about the same. Conclusions: In all college students, regardless of their enrollment in Compassion, well-being and resilience are positively correlated with both mindfulness and self-compassion. Furthermore, scores based on enrollment in “Compassion” yielded higher levels of mindfulness, self-compassion, resilience, and well-being.

Using the case of a children's home in Nepal, Ama Ghar, this study utilizes concepts of resiliency and community service to assess children's ability to react and recover from disaster. As earthquakes continues to strike rural and urban populations \u2014 from Mexico to Italy \u2014 learning the recovery stories of the 2015 Nepal Earthquake from the Ama Ghar family may further understanding on how to promote resiliency for youth in a post-disaster stage. Although community service in many respects has been supported as a successful youth development tool in Western contexts, researchers call for a more extensive look that compares variables of community service through a global lens. Because of the research backing the benefits that community involvement has on promoting civic responsibility, citizenship, and human and societal well-being, this study proposes that facilitating an active role through community service is a positive way to generate resilience among a child. After conducting in-depth interviews on Ama Ghar caretakers and alumni, it was ultimately concluded that there is a positive relationship between community service and overall resilience of a child. It was found that different forms of discussions of disaster, community service, and resilience shows the complex interconnectedness among these attributes and how this relationship accounts for the building up of resiliency among children who have faced disaster such as the earthquake in Nepal. Learning about the nurturing of children within the blended Ama Ghar family and their vivid first-hand experiences may be scalable however more in-depth research should be conducted to fully understand the complex factors that contribute to the rebuilding of well-being for disaster-affected youth.

Rasopathies are a family of developmental syndromes that exhibit craniofacial abnormalities, cognitive disabilities, developmental delay and increased risk of cancer. However, little is known about the pathogenesis of developmental defects in the nervous system. Frequently, gain-of-function mutations in the Ras/Raf/MEK/ERK cascade (aka ERK/MAPK) are associated with the observed pathogenesis. My research focuses on defining the relationship between increased ERK/MAPK signaling and its effects on the nervous system, specifically in the context of motor learning. Motor function depends on several neuroanatomically distinct regions, especially the spinal cord, cerebellum, striatum, and cerebral cortex. We tested whether hyperactivation of ERK/MAPK specifically in the cortex was sufficient to drive changes in motor function. We used a series of genetically modified mouse models and cre-lox technology to hyperactivate ERK/MAPK in the cerebral cortex. Nex:Cre/NeuroD6:Cre was employed to express a constitutively active MEK mutation throughout all layers of the cerebral cortex from an early stage of development. RBP4:Cre, caMEK only exhibited hyper activation in cortical glutamatergic neurons responsible for cortical output (neurons in layer V of the cerebral cortex). First, the two mouse strains were tested in an open field paradigm to assess global locomotor abilities and overall fitness for fine motor tasks. Next, a skilled motor reaching task was used to evaluate motor learning capabilities. The results show that Nex:Cre/NeuroD6:Cre, caMEK mutants do not learn the motor reaching task, although they performed normally on the open field task. Preliminary results suggest RBP4:Cre, caMEK mutants exhibit normal locomotor capabilities and a partial lack of learning. The difference in motor learning capabilities might be explained by the extent of altered connectivity in different regions of the corticospinal tract. Once we have identified the neuropathological effects of various layers in the cortex we will be able to determine whether therapeutic interventions are sufficient to reverse these learning defects.

The interplay between Post Traumatic Stress Disorder (PTSD) and self-efficacy lies in the efficacy-activated processes that comprise an individual’s cognitive and belief systems. Previous research shows that low self-efficacy contributes to development and maintenance of mental disorders like PTSD, while high self-efficacy influences ability to visualize, implement, and maintain success scenarios (resilience) related to effective mental coping. Negative cognition makes it difficult to pursue a coping success scenario in the presence of overriding self-doubt and often arises because a traumatic event has made it difficult to retrieve positive self-identities or has reactivated negative self-identities. Consistent with this model, we predict that a negative association exists between self-efficacy and PTSD onset susceptibility. We employed a pre-test/post-test design using a susceptibility/resilience questionnaire to assess predisposition to PTSD. Vignettes, designed to either raise or lower self-efficacy, were used to separate participants into groups and revealed a significant interaction between low and high self-efficacy across the pre- and post-tests, supporting the assertion that high self-efficacy guards against PTSD onset susceptibility while low self-efficacy may make someone more susceptible to developing PTSD-related symptoms.

The RAS/MAPK (RAS/Mitogen Activated Protein Kinase) pathway is a highly conserved, canonical signaling cascade that is highly involved in cellular growth and proliferation as well as cell migration. As such, it plays an important role in development, specifically in development of the nervous system. Activation of ERK is indispensable for the differentiation of Embryonic Stem Cells (ESC) into neuronal precursors (Li z et al, 2006). ERK signaling has also shown to mediate Schwann cell myelination of the peripheral nervous system (PNS) as well as oligodendrocyte proliferation (Newbern et al, 2011). The class of developmental disorders that result in the dysregulation of RAS signaling are known as RASopathies. The molecular and cell-specific consequences of these various pathway mutations remain to be elucidated. While there is evidence for altered DNA transcription in RASopathies, there is little work examining the effects of the RASopathy-linked mutations on protein translation and post-translational modifications in vivo. RASopathies have phenotypic and molecular similarities to other disorders such as Fragile X Syndrome (FXS) and Tuberous Sclerosis (TSC) that show evidence of aberrant protein synthesis and affect related pathways. There are also well-defined downstream RAS pathway elements involved in translation. Additionally, aberrant corticospinal axon outgrowth has been observed in disease models of RASopathies (Xing et al, 2016). For these reasons, this present study examines a subset of proteins involved in translation and translational regulation in the context of RASopathy disease states. Results indicate that in both of the tested RASopathy model systems, there is altered mTOR expression. Additionally the loss of function model showed a decrease in rps6 activation. This data supports a role for the selective dysregulation of translational control elements in RASopathy models. This data also indicates that the primary candidate mechanism for control of altered translation in these modes is through the altered expression of mTOR.

Depression, anxiety, and suicidal thoughts or actions are on the rise in adolescents (National Institute of Mental Health, 2015; Bridge, Asti, & Horowitz, 2015). Parents, school administrators, and therapists are searching for resiliency factors with in at-risk groups to aid students in need. In previous work, Luthar and Zigler (1992) reported that intelligent youth are more resilient than less intelligent youth under low stress conditions but they lose their advantage under high stress conditions. This study examined whether intelligence (reflected in grade point average; GPA) and maladaptive (internalizing and externalizing symptoms) behaviors are negatively related in adolescents, and tested whether level of stress, reflected in emotion regulation and friendship quality, moderated that association. It also probed whether the relationships differ by gender. Sixth-graders (N=506) were recruited with active parental consent from three middle schools. Adolescents completed self-report questionnaires Regarding demo graphics, maladaptive behaviors, emotion regulation, and friendship quality, and GPA data were collected from the school. Regression analyses found that GPA was negatively related to externalizing symptoms. Girls with poor friendship communication report significantly higher maladaptive behaviors. This relation was more pronounced for girls with high GPAs, as predicted. Results support the theory that intelligent female adolescents are more reactive under adverse circumstances. Future efforts should follow students through middle school into high school to evaluate whether friendships remain important to adjustment, hold for boys as well as girls, and have implications for relationship interventions.

The objective of this project was to develop a forgiveness training program to be used at a corporate level in addition to the current conflict management strategies. In addition to teaching the value of forgiveness, this search also touches on resilience and how forgiveness increases our personal resiliency. Forgiveness and resilience have been closely linked in previous forgiveness research as they relate to reconciliation. Based on the research, forgiveness has been widely talked about in relation to religious practices, however it is now being discussed in relation to communication. In order to understand forgiveness as a communication process, one has to understand where it began and how the definition of forgiveness has evolved overtime. This project looks at how forgiveness creates value for the individual in terms of the relationship and expresses why forgiveness and reconciliation are not mutually exclusive. Forgiving an individual does not always lead to reconciling the relationship; however, making it so the individuals can continue working together is the goal at work. A key part of understanding forgiveness is being able to identify what forgiveness is not, as much of what we have been taught from a young age is the exact opposite. Adult learners are much different from any other type of learners due to the level of life experience they have -- which can often make it more challenging to rewrite concepts, like forgiveness. This project identifies the best ways to teach adult learners through the use of interactive handouts and videos that demonstrate the power of forgiveness in our day-to-day lives.
